dc.description.abstractRecently, protein and amino acid supplements including branched-chain amino acids (BCAAs) supplement are popularly consumed by people, who mainly believe that the supplement helps enhancement of physical performance. However, adverse effects such as neurotoxicity and altered neurobehavioral function upon BCAAs have been reported in brain. Therefore, it would be important to understand the effects of BCAAs on genome-wide gene regulation in brain, which may provide hints for what functional consequences BCAAs lead to. To characterize gene regulation upon BCAAs, genome-wide expression analysis was performed using microarray analysis. In the range of concentrations that did not directly affect cell viability, BCAA treatment influenced gene expression involving cell signaling pathways by double-stranded DNA binding and intra-cellular molecular functions such as chromatin organization. c-FOS and CBX1 were identified as candidate genes involved in changed functions of double-stranded DNA binding and chromatin organization, respectively. The mRNA expression change of c-FOS was not regulated by BCAA-included cyclic adenosine monophosphate (cAMP) response, since the activities of cAMP response elements incorporating the luciferase reporter were not altered upon BCAA treatment. Also, it was not changed by transient CBX1 overexpression in cells neither, possibly because CBX1-induced chromatin changes may require a longer-term of BCAA treatment than expected. Taken together, this study demonstrated the genome-wide gene expression changes in brain for the first time and the altered gene expressions upon BCAA treatment in brain cells indicated cell signaling response via c-FOS and chromatin changes by CBX1 may be major targets. However, further investigation by in vivo study is needed to confirm whether the changes in chromatin and signalings are connected each other and lead to altered brain functions.;본 연구에서는 뇌세포주에서 분지아미노산 처리에 의해 일어나는 유전체의 발현 변화를 전장수준에서 살펴보았다. 최근 분지아미노산 보충제는 운동선수, 군인, 체육관에서 정기적으로 운동하는 사람들이 근육에서 운동 능력 향상의 효능을 기대하여 많이 사용되고 있다. 그러나 최근 연구에서 고농도의 분지아미노산 처리가 세포에서 신경독성을 나타내거나 쥐에서 신경 행동 기능을 바꾼다는 이상 반응들이 보고되었다. 따라서, 본 연구에서는 분지아미노산 처리가 뇌세포에서 일으키는 전장 분석을 실시하여 주된 발현변화를 일으키는 유전자들을 분석함으로써 분지아미노산 보충제를 섭취에 대한 기초자료를 제공하고자 하였다. 본 연구에서는 뇌세포주에 세포 생존능에 영향을 미치지 않는 농도 내에서 분지아미노산 처리를 한 후, 마이크로 어레이를 이용하여 발현을 분석하였다. 그 결과, c-FOS와 같이 DNA 이중 나선에로의 결합에 의해 바뀌는 세포 신호 전달 경로와 CBX1와 같이 염색질 조직과 같은 세포 내 분자적 기능과 관련된 유전자 발현이 변화했다. 본 연구에서는 c-FOS의 발현 조절 기전을 밝히기 위해 두 가지 기능 연구를 수행하였다. 루시퍼레이즈 리포터 어세이를 사용하여 c-FOS의 발현이 프로모터 CRE에 의해 조절되지 않음을 확인하였다. 또한 일시적인 CBX1 과발현에 의해 c-FOS의 발현이 조절되지 않았는데, 이는 염색질 조직이 변화하기 위해서는 보다 긴 시간의 분지아미노산 처리가 필요하기 때문으로 보인다. 이와 같은 본 연구의 결과는 뇌세포주에서 분지아미노산 처리에 의해 일어나는 전장 유전체의 변화를 처음으로 분석하여 분지아미노산 처리가 뇌에 미치는 영향에 대한 기초 정보를 제공하는데 기여하였으나 그 정확한 기전을 보이는 데는 한계가 있었다. 앞으로 후보유전자에 대한 심도 깊은 연구 및 장기간 in vivo 모델을 통한 연구가 더 필요한 것으로 사료된다.-
