dc.description.abstractHalofuginone (HF), a derivative of plant-derived drug febrifugine, has been reported to have effects on a variety of diseases. HF has been shown to suppress autoiummune diseases by inhibition of Th17 cell differentiation throughthe activation of the amino acid starvation response (AAR). In addition, HF has been shown to inhibit progression of fibrosis by inhibiting TGFβ1-induced Smad2/3 phosphorylation. Recently, HF has been shown to activate AAR through the inhibition of proline charging of tRNAproby glutamyl-prolyl-tRNAsynthetase. In this study, I found thatHF inhibits TGFβ1-induced phosphorylation of Smad2/3 GCN2 dependently and eIF2α independently. In GCN2 -/- MEF cells, HF did not reduce phosphorylation of Smad2/3. However, in eIF2α (S51A) MEFs,in which wild type eIF2α is replaced by a non-phosphorylating mutant form,HF-reduced Smad2/3 phosphorylation was not changed. In addition, I found that HF inhibits BMP2-induced phosphorylation of Smad1.;식물로부터 추출한 약물 중 하나인 Halofuginone은 다양한 질병에 효과가 있다는 것이 보고되어 왔다. Halofuginone은 amino acid starvation response를 활성화시킴으로써 Th17 cell의 분화를 억제한다. 이것은 자가면역질환에 치료제로 사용될 수 있는 가능성을 제공한다. 또한 Halofuginone은 TGFβ1 signaling에서 활성화되는 Smad2/3 의 인산화를 억제하는데, 이것은 Smad2/3의 활성화로 인해 유발되는 fibrosis 질병을 또한 억제할 수 있다. 최근에 Halofuginone은 amino acid starvation response를 유도하는데 Halofuginone이 proline이 transfer RNApro에 charging되는 것을 억제함으로써 일어난다는 사실이 밝혀졌다. 이번 연구에서는Halofuginone이 TGFβ1 signaling에서 활성화되는 Smad2/3의 인산화를 억제하는데 GCN2에 의존적으로, eIF2α에 독립적으로 하는 것으로 나타났다. Halofuginone에 의해서Smad2/3 인산화가 감소하는데 GCN2-/- MEF 에서는 감소하지 않았고 eIF2α 기능을 하지 못하는 SV40-eIF2α(S41A) MEF에서는 감소하는 경향을 나타내었다. 또한 Halofuginone은 TGFβ1 superfamily인 BMP2가 유도하는Smad1의 인산화도 억제하는 것으로 나타났다.-
