Abstract
Purpose: The purpose of this study was to evaluate whether Irradiated Allogenic Cancellous Bone combined with tetracycline could be effective on new bone formation in a rat calvarial defect model. Methods: Twenty-seven male Harland Sprague-Dawley rats were assigned to one of three treatment groups; Irradiated Allogenic Cancellous Bone (ICB^(??); Rocky Mountain Tissue Bank, Aurora, CO, USA)(0.1g/rat), Irradiated Allogenic Cancellous Bone /Tetra-cycline(Oxymycin cap 500mg, Chongkundang, Seoul, Korea) (100mg/rat) and non-graft control. This material was placed into 8mm calvarial critical sized defects (CSDs). They were measured histologically and radiographically following a 2, 4 and 8 weeks healing interval. One way ANOVA test was used to compare the difference between the control and graft group. Result: Radiographic density was statistically greater in a group treated with Irradiated Allogenic Cancellous Bone when compared with control group at 8 weeks (p<0.01). In the Irradiated Allogenic Cancellous Bone /Tetracycline group, defect size was also decreased but there was no significant difference between the group treated with Irradiated Allogenic Cancellous Bone/Tetracycline and control. In the histomorphometric analysis, new bone area of the Irradiated Allogenic Cancellous Bone group was greater than that in the Irradiated Allogenic Cancellous Bone/Tetracycline group at 4 and 8 weeks, but difference was not statistically significant (p>0.05). Conclusion: Irradiated Allogenic Cancellous Bone is effective graft material on bone formation in the rat calvarial defect model. Tetracycline does not enhance bone regeneration when it is combined with Irradiated Allogenic Cancellous Bone.;목적: 이 연구는 방사선조사 동종 망상골과 Tetrycycline의 병용이 쥐의 두개골 결손모델에서 골 형성에 효과적일 수 있는지를 알아보기 위해서 시행되었다. 실험 재료 및 방법: 27마리의 백서에 8mm의 두개골 결손 부위를 형성하여 방사선조사 동종 망상골, 방사선조사 동종 망상골과 Tetracycline을 병용하여 이식한 실험군과 이식재를 사용하지 않은 대조군으로 나누어 실험한 후 각각 2주, 4주, 8주 후에 방사선학적, 조직학적으로 관찰하였다. 대조군과 실험군사이의 차이를 비교하기 위하여 일원분산분석 방법으로 통계처리 하였다. 결과: 1. Radiographic density는 방사선조사 동종 망상골을 사용한 그룹이 8주에서 대조군과 비교하였을 때 통계학적으로 큰 차이를 보였다 (p<0.01). 2. 방사선조사 동종 망상골과 Tetracycline 을 병용한 그룹에서도 결손부 크기가 감소되었지만 대조군과의 두드러진 차이는 없었다(p>0.05). 3. 조직형태학적 분석에서 방사선조사 동종 망상골 그룹의 신생골 형성영역이 방사선조사 동종 망상골과 Tetracycline을 병용한 그룹보다 4주, 8주 모두 크게 나타났다 결론: 방사선조사 동종 망상골을 은 쥐의 두개골 결손 모델에서 골 형성에 있어 효과적인 이식재이나 Tetracycline은 방사선조사 동종 망상골과 병용되었을 때 골 재생을 강화하지 않는다.
