Abstract
본 연구에서는 생리학적으로 중요한 활성산소종을 검출할 수 있는 형광 물질에 대하여 연구하였다. 목표에 따른 형광 검출 물질의 구조를 고안하고 화합물을 합성하여 그 특성을 연구하였으며 안정성 및 선택성을 가지고 있는지와 검출의 작용 원리, 생체 내 응용가능성 등을 연구하였다. 세부적인 연구 내용 및 결과는 다음과 같다. 1장에서는 과산화수소 (H2O2)를 효과적으로 검출하는 철(II) 금속 착화합물인 ZP1Fe2를 이용하여 turn-on 타입의 형광 변화를 보았다. 완충용액에서 과산화수소를 검출하여 22배의 형광 증가를 확인했다. 이러한 signal 변화는 과산화수소의 철 center의 활성화와 그에 따르는 리간드의 oxidative N-dealkylation 때문인 것을 모델 화합물 DPAPhOH를 이용한 메커니즘 연구를 통해 밝혀냈다. 또한 이 물질은 다른 활성산소종에 비해 과산화수소에 대해 선택성을 보였으며 생체 내 과산화수소에 대해 리소좀에서 기관 선택적인 검출을 하는 것이 확인되었다. 2장에서는 생체 내 신호전달과 세포소멸에서 중요한 역할을 하는 일중항산소 (1O¬2)를 검출하는 형광 센서에 대해 연구하였다. 일중항산소에 대한 형광 물질들은 일반적으로 anthracene base를 가지던 것에 비해 isobenzofuran base를 가지며 aryl 위치에 형광단을 치환하여 ratiometric한 형광 변화를 보일 수 있는 물질들을 고안하였다. 이 물질들은 [2+4] 페리 고리모양 반응을 통해 endoperoxide를 형성하며 이 때 가교산소가 생기면서 conjugation의 변화에 의해 ratiometric한 형광 변화를 보인다. 형광단의 크기가 커짐에 따라 반응 속도 상수가 커지는 것을 확인하였으며 다른 활성산소종에 비해 일중항산소에 대한 선택성과 물질의 안정성을 측정하였다. 나노 입자를 형성하여 수용액 상에서의 응용성과 세포 내의 일중항산소를 검출하여 ratiometric한 형광 변화를 보이는 것으로 생체로의 응용성도 확인하였다.;Herein, we report a novel sensing method involving the detection of fluorescence in response to the presence of intracellular H2O2 (Scheme. 1). The probe platform was readily constructed by complexation of iron ions and Zinpyr-1 (ZP1), a fluorescent zinc ion probe, however, the idea may be extended to any ensemble of iron and fluorescent metal ion sensors. We have proposed that the sensing mechanism involves activation of the Fe center by H2O2, followed by oxidative N-dealkylation of the ligand to liberate the fluorescence-quenching Fe ionophore. The H2O2 probe exhibited exceptional selectivity for H2O2 over other ROSs, such as •OH, t-BuOOH and O2•. Interestingly, the probe localized at the lysosomes of live HeLa cells, producing an organelle-specific response to intracellular H2O2. Biological singlet oxygen (1O2), a highly reactive oxygen species (hROS), is linked to various pathological and physiological processes in the human body, but molecular mechanisms of the 1O2-mediated processes remain unestablished. We created a series of fluorescence ratiometric 1O2 probes by attaching fluorogenic aryl groups to the 1 position of isobenzofuran, which undergoes the [2+4] pericyclic reaction with 1O2. The 1O2 probes displayed fluorescence ratiometric responses with excellent 1O2 selectivity over other ROSs, including hydrogen peroxide, hydroxyl radical, alkylhydroperoxide, and superoxide. The 1O2 response rate increased in proportional to the conjugation length of the aryl substituent of isobenzofuran. Fluorescence ratiometric 1O2 detection ability was conserved in the aqueous nanosuspension state of the probes. Finally, we successfully visualized intracellular 1O2 production by PMA (phorbol 12-myristate 13-acetate) stimulation of RAW 264.7 macrophage cells.
