Abstract
Ahnak is a protein originally identified as a nuclear phosphoprotein in neuroblastoma and skin epithelium. A previous report indicated that central repeated units (CRUs) of Ahnak bind to Smad proteins and then regulate TGF-β signaling resulting in cytostatic effect. In this thesis, I provide detailed mechanisms of Ahnak-mediated potentiation of Transforming Growth Factor (TGF)-β signaling which leads to a negative regulation of cell growth serving as a novel tumor suppressor. To evaluate the function of Ahnak in tissues, we established Ahnak knockout (KO) mice. Ahnak KO mice showed stunted growth and reduced adipose tissue, suggesting a complex physiological effect of the Ahnak deficiency. Interestingly, the proliferation rate of Ahnak KO mouse embryonic fibroblasts (MEFs) was higher than that of WT. I introduced a construct expressing four CRUs (4CRUs, amino acid residues 3860-4412) into MEF cells isolated from Ahnak KO mice and analyzed cell proliferation rate. Add-back expression of Ahnak-4CRUs into Ahnak KO MEFs resulted in significant decrease to the level of WT MEFs. The results suggested that 4CRUs of Ahnak plays a critical role in cell growth. To verify the function of Ahnak in cell growth, we crossed mice carrying the mammary gland specifically expressed polyoma virus middle T antigen (MMTV-PyVT) with Ahnak KO mice, and the MMTV+/Tg Ahnak KO mice were compared to that in MMTV+/Tg WT mice at 6 weeks of age. The expression of proliferating cell nuclear antigen (PNCA), a cell proliferation marker, was significantly increased in MMTV+/Tg Ahnak KO compared to the levels in MMTV+/Tg WT. The result strongly indicated that Ahnak regulates c-Myc protein leading to controlling tumor growth. To analyze loss-of-function of Ahnak in tumor, we also analyzed Ahnak expression level in human breast cancer tissues. Expression of Ahnak was decreased in aggressive breast cancer tissues. These results suggest that Ahnak acts as a tumor suppressor and that its deficiency increases breast cell proliferation by potentiating TGF-β signaling. TGF-β plays an important role in cancer metastasis either. Therefore we investigated whether Ahnak protein regulates epithelial-to-mesenchymal transition (EMT), one of metastasis-promoting events. Several lines of evidence suggested that EMT is another biological process positively regulated by TGF-β/Smad3 signaling. EMT enables cancer metastasis by conferring motility and invasiveness to carcinoma. Briefly, EMT is characterized by morphological change, alteration of marker genes, disruption of cell-cell junction, cytoskeletal rearrangement, and acquisition of motility and invasiveness. In spontaneously transformed human keratinocyte cell line HaCaT, Ahnak was depleted using RNA interference (RNAi) for loss-of-function study. Morphological change upon TGF-β-induced EMT was not seen in the absence of Ahnak protein. Moreover alteration of marker proteins such as E- and N-cadherin was not observed in Ahnak depleted cells. Nuclear translocation of β-catenin, another character of EMT, was not seen in Ahnak-depleted cells. Orchestration of EMT is attributed by EMT-promoting transcription factors such as Snail (Snail1), Slug (Snail2), Twist1/2, and ZEB1/2. However Ahnak-depletion prevented induction of these EMT master transcription factors in response to TGF-β. Furthermore, cell migration was inhibited in the absence of Ahnak protein. It was due to irregular F-actin rearrangement which is necessary for stress fiber formation. Ahnak knockdown led to impaired cell invasion in Matrigel invasion assay. It was involved in aberrant expression of matrix metalloproteinases (MMPs) regulating extracellular matrix (ECM) remodeling. To address the function of Ahnak in detailed EMT signaling, how Ahnak protein exerts TGF-β/Smad3 signaling was investigated. Ahnak protein activated nuclear translocation of Smad3 in HaCaT cells. It promoted transcription of EMT-specific TGF-β/Smad3 target genes such as Connective tissue growth factor (CTGF), Collagen 1 A 2 (COL1A2), and Tissue inhibitor of metalloproteinase (TIMP) 1. Thus it was noticed that Ahnak protein positively regulates EMT. Lastly, it was examined whether Ahnak protein promotes cancer metastasis in vivo. To assess pulmonary metastasis, aggressively metastatic melanoma cell line was inoculated into WT and Ahnak KO mice intravenously. 21days later, Ahnak KO mice exhibited significantly decreased (less than half) lung metastatic colonies compared to WT. In WT, Endoglin (CD105), a pro-angiogenic Smad3 target gene, was highly expressed especially in peritumoral area consistently with other previous reports. However it was rarely expressed in Ahnak KO mice. Endothelial Endoglin expression was decreased in Ahnak KO mice compared to WT. These results indicated that tumor environmental Ahnak protein promotes cancer metastasis by activating tumor angiogenesis. Therefore we demonstrate that Ahnak protein suppresses tumor initiation while it promotes metastasis through TGF-β signaling.;Ahnak은 신경아종(neuroblastoma)과 피부 상피세포에서의 핵내 인산화 단백질(nuclear phosphoprotein)로서 처음 발견되었다. 이전 논문에서는 Ahnak의 중간 반복 단위 (central repeated units, CRUs) 부분이 Smad 단백질과 결합하고, TGF-β 신호 전달을 조절하여 세포의 성장을 억제한다는 것을 밝혀내었다. 이 학위 논문에서는 Ahnak 단백질이 새로운 항암인자 (tumor suppressor)로서 TGF-β 신호 전달을 강화시키고, 세포의 성장을 억제하는 자세한 기전을 연구하였다. Ahnak이 조직에서 하는 역할에 대하여 연구하기 위해 우리는 Ahnak 유전자 결핍 쥐 (Ahnak knockout mouse)를 제작하였다. Ahnak 유전자 결핍 쥐는 발육이 저해 되어 있었고, 지방 조직이 감소 되어 있었다. 이것은 Ahnak 발현이 없어지면 복잡한 생리적 효과가 나타난다는 것을 뜻한다. 흥미롭게도 Ahnak이 결핍된 쥐 배아 섬유아세포 (mouse embryonic fibroblasts, MEFs)는 야생형(wild type, WT)보다 세포 분열 속도가 더 높았다. Ahnak의 4CRUs를 Ahnak이 결핍된 MEFs에 발현시키면 분열 속도가 유의미하게 야생형만큼 감소하는 결과가 나왔다. 이 결과는 Ahnak의 CRUs가 세포 성장에 중요한 역할을 한다는 것을 뜻한다. 세포 성장에서 Ahnak의 역할을 연구하기 위하여 우리는 mammary gland specific polyoma virus middle T antigen (MMTV-PyVT) 발암 유전자를 발현하면서 Ahnak 유전자가 결핍된 쥐를 제작하여 6주령 시기에 야생형 쥐와 비교하였다. 세포 분열 표지 단백질인 proliferating cell nuclear antigen (PCNA)과 발암 유전자인 c-Myc이 야생형에 비하여 Ahnak 결핍 쥐에서 더 강하게 발현되어 있었다. 우리는 암에서 Ahnak의 기능 상실 (loss-of-function)을 분석하기 위하여 유방암 환자의 조직에서 Ahnak의 발현량을 분석하였고, 악성의 유방암에서 Ahnak의 발현이 감소되어 있는 것을 확인하였다. 이러한 결과는 Ahnak이 항암인자의 역할을 한다는 것과 Ahnak의 결핍이 TGF-β 신호 전달을 강화 시킴으로써 유방암 세포의 분열을 촉진한다는 것을 뜻한다. TGF-β는 암의 전이도 촉진을 한다. 우리는 전이 촉진 기능을 연구하기 위해 Ahnak 단백질이 epithelial-to-mesenchymal transition (EMT) 현상을 유도하는지에 대하여 알아보았다. EMT는 TGF-β/Smad3 신호 전달 체계에 의하여 강화 조절되는 또 다른 생물학적 현상이다. EMT는 운동성이 없는 상피세포에서 유래된 암세포가 운동성과 침투성을 갖게 함으로써 암의 전이를 유발한다. EMT의 특징으로는 세포 모양의 변화, 표지 단백질의 변화, 세포간 연결의 붕괴, 세포 골격의 재배열, 운동성과 침투성의 획득 등이 있다. 우리는 자연 암화된 인간 케라틴 생성 세포주 (keratinocyte cell line) 인 HaCaT 세포에서 RNA interference (RNAi) 를 이용하여 Ahnak 을 감소 시키고 기능 상실 연구 (loss-of-function study) 를 수행하였다. 그 결과 TGF-β에 의해 EMT가 유도되면 세포의 모양이 변하는데 RNAi에 의해 Ahnak 단백질이 감소 되면 그러한 현상이 나타나지 않았다. 그리고 Ahnak 단백질이 감소된 세포에서는 표지 단백질인 E-cadherin과 N-cadherin의 변화가 보이지 않았다. 또 하나의 EMT 현상인 β-catenin 단백질의 핵 내 이동 현상도 Ahnak이 감소된 세포에서는 보이지 않았다. EMT는 EMT 촉진 전사인자인 Snail (Snail1), Slug (Snail2), Twist1/2, ZEB1/2 등에 의하여 조절된다. 그러나 Ahnak 단백질이 감소 되면 TGF-β에 따른 Snail, Slug, Twist의 증가가 나타나지 않았다. 그리고 Ahnak 단백질의 결핍은 세포의 이동도 억제시켰다. 그것은 스트레스 섬유 (stress fiber) 가 형성 되는데 필요한 F-actin이 규칙적으로 재배열되지 못하였기 때문이었다. 더불어 Ahnak 단백질의 감소는 Matrigel 침투 실험에서 세포의 침투능력을 약화시켰다. 그것은 세포 외 기질 (extracellular matrix, ECM) 을 변형 시키는 matrix metalloproteinases (MMPs) 의 발현이 잘 이루어지지 않기 때문이었다. 세부적으로 EMT 신호 전달에서 Ahnak 단백질의 기능이 무엇인지 알아내기 위하여 Ahnak 단백질이 어떻게 TGF-β/Smad3 신호 전달에 참여 하는지 연구 하였다. Ahnak 단백질은 Smad3 단백질이 핵으로 더 잘 이동하도록 유도하였다. 그리고 그 결과 EMT 특이적으로 발현되는 TGF-β/Smad3 의 대상 유전자 (target gene) 인 Connective tissue growth factor (CTGF), Collagen 1 A 2 (COL1A2), Tissue inhibitor of metalloproteinase (TIMP) 1 등의 전사를 촉진시켰다. 그러므로 우리는 Ahnak 단백질이 EMT를 강화 시킨다는 것을 밝혔다. 마지막으로 Ahnak 단백질이 생체 내 암의 전이도 촉진 하는가에 대하여 연구하였다. 우리는 폐 전이를 연구하기 위해 야생형 쥐와 Ahnak 결핍 쥐에 전이성이 매우 큰 흑색종 (melanoma) 세포주를 정맥 주사 하였다. 21일 뒤에 Ahnak 결핍 쥐에서는 야생형에 비하여 폐 전이가 절반 이하로 감소 되어 있었다. 이 때 야생형에서는 Smad3의 대상 유전자이면서 신생 혈관을 유발하는 (pro-angiogenic) Endoglin (CD105) 단백질이 암 주위 조직에서 높게 발현되어 있는 것으로 나타났으며, 이것은 이전의 연구 결과에 부합하는 것이었다. 반면 Ahnak 결핍 쥐에서는 Endoglin이 거의 발현되지 않았다. 내피 세포가 발현하는 Endoglin 단백질의 양도 야생형에 비하여 Ahnak 결핍 쥐에서 감소되어 있었다. 이러한 연구 결과는 Ahnak 단백질이 암에 의해 혈관이 새로 만들어지는 것을 활성화 시킴으로써 암의 전이를 촉진한다는 것을 시사한다. 따라서 우리는 Ahnak 단백질이 TGF-β 신호 전달을 통하여 암이 시작 되는 것을 억제하는 반면에 암 전이는 촉진 한다는 것을 밝혀내었다.
