Abstract
말기 암 치료에 있어서 화학요법은 중요한 역할을 한다. 그러나 화학 요법 시 발생하는 항암제 다 제 내 성은 치료에 큰 장애가 되고 있는데, 이러한 다 제 내 성을 유발하는 한 원인이 P-glycoprotein (P-gp) 의 과 발현이다. 현재까지 알려진 여러 기전 중 항암제 다 제 내 성을 극복하는 가장 확실한 방법은 p-gp의 과 발현을 억제하는 것이다. Antofine은 박 주 가리 과 (Asclepiadaceae) 식물인 산해 박 (Cynanchum paniculatum) 에서 유래한 phenanthroindolizidine alkaloid이다. 이것은 이미 항 염 증 및 강력한 암세포 증식 저해 작용이 있는 것으로 알려졌으나 mouse의 체내에 투여된 후, 빠르게 소실되어 생체 내에서 안정성 (stability) 이 현저히 낮다는 연구 결과가 있었기 때문에 실제로 약으로 개발되기는 어려운 상황이다. 따라서 antofine과 유사하거나 더 좋은 항암작용을 갖고, 내성도 극복할 수 있으면서 안정성은 높은 antofine의 유도체를 개발한다면 항암제의 좋은 후보물질이 될 수 있을 것으로 사료된다. 본 연구에서는 모 물질인 (-)-antofine을 positive control로 하여 (-)-antofine 유도체인 (-)-13-hydroxymethylantofine ((-)-HMA)의 paclitaxel 내성 사람 폐암 세포 주에 대하여 세포 성장 억제 활성과 내성 극복 기전에 대하여 알아 보았다. (-)-HMA는 (-)-antofine보다 활성이 10배 정도 낮긴 하지만, parent 세포인 A549와 비교해 볼 때 A549-Pa 세포에 대해서 나노 몰 농도 수준으로 충분히 좋은 활성을 보였으며 (IC50가 각각 SRB시 33.9 nM 및 45.7 nM, MTT시 28.8 nM과 41.2 nM), p-gp 발현을 (-)-antofine과 마찬가지로 24시간과 48시간에서 모두 저해하였다. 또한 p-gp의 mRNA인 MDR1의 발현 역시 24시간과 48시간에서 모두 저해하는 것을 RT-PCR (reverse transcription polymerase chain reaction) 과 real time RT-PCR을 통해 확인하였다. P-gp의 transporter 기능을 직접적으로 저해 하는지 확인 하기 위하여 rhodamin 123 assay을 수행한 결과 대조 군에 비해 (-)-HMA에 의해 세포 내에 축적된 rhodamin 123의 양이 증가하였다. (-)-HMA의 in vitro 실험에 따른 사람 폐암 세포 A549와 paclitaxel 내성 세포 A549-Pa의 성장 저해 활성을 바탕으로 사람 폐암 세포 A549와 paclitaxel 내성 세포 A549-Pa xenograft 누드마우스 실험 동물 모델에서 (-)-HMA의 종양 성장 억제 효능을 평가하였다. 그 결과 약물에 의한 몸무게 변화 및 기타 다른 독성은 확인되지 않았으며 A549 세포를 inoculation한 누드 마우스에 (-)-HMA를 투여하였을 때, A549 xenograft 종양의 성장을 5 mg/kg과 10 mg/kg에서 각각 54.93 % 및 60.66 %로 억제하였으며, A549-Pa 세포를 inoculation한 누드 마우스에서도 38.40 % 및 54.78 %로 억제하였다. 이러한 연구 결과를 바탕으로 (-)-HMA는 항암제 다 제 내 성을 극복할 수 있을 뿐만 아니라, 임상 적용도 기대해볼 수 있어, 항암제 개발 시 우수한 후보 물질이 될 수 있음을 확인하였다. Transforming growth factor-β (TGF-β) 는 세포의 기능을 조절하는 다양한 역할을 한다. 암의 초기 단계에서는 조직의 항상성을 유지하고 세포의 분화, 증식, 유착 등을 조절하여 암세포의 성장을 억제하지만, 암이 일정한 크기 이상으로 성장한 후기 단계에서는 상피세포의 중간 엽 세포로의 분화 (epithelial-mesenchymal transdifferentiation) 를 유도하고 암의 성장과 전이, 면역 기전의 회피 등에 기여함으로써 도리어 암으로의 진행을 촉진시킨다. 이러한 이유로 TGF-β 억제를 통한 암전이 억제제 개발은 큰 성과를 거두지 못하였다. 본 연구에서는 사람 전립선 암 세포주인 PC-3 세포에 대한 TGF-β type I receptor inhibitor (ALK-5 inhibitor) 인 IN-1130 (물질이름) 의 항전이 효능을 확인하고 그 작용기전을 연구하였다. 먼저 세포 성장 억제 효능 평가 시 PC-3에서 IN-1130의 세포 증식 억제 효과가 100 μM 이하에서는 미미한 것을 확인하였고, wound healing assay, colony dispersion assay, invasion assay 등을 수행한 결과 IN-1130이 PC-3 세포의 이동성과 침 윤성을 저해한다는 사실을 알 수 있었다. IN-1130의 PC-3 세포에 대한 항전이 기전을 알아보기 위해 matrix metalloproteinase-2 (MMP-2) 와 MMP-9의 발현 정도를 유전자 수준과 단백질 수준에서 살펴 본 결과 IN-1130이 TGF-β에 의해 증가된 MMP-2와 MMP-9의 발현을 유전자와 단백질 수준에서 감소시켰고, gelatin zymography를 통해 확인한 결과, TGF-β에 의해 증가된 MMP-9의 활성이 IN-1130에 의해 감소하는 것을 확인하였다. IN-1130이 EMT (epithelial mesenchymal transition) 인 E-cadherin의 발현에 유전자와 단백질 수준에서 영향을 미치는지에 대해 RT-PCR과 western blot analysis를 통해 관찰한 결과, TGF-β에 의해 E-cadherin의 mRNA와 단백질의 발현이 감소하는 것을 관찰하였고 IN-1130 처리에 의해 증가되는 것도 관찰하였다. 이러한 결과로서 ALK-5 억제제인 IN-1130은 항전이 효능이 있는 우수한 후보 물질이 될 수 있을 것으로 사료된다.;Chemotherapeutics are the most effective treatment for metastatic cancers. However, the ability of cancer cells to become simultaneously resistant to different drugs remains a significant impediment to successful chemotherapy. Multidrug resistance can also result from the over expression of p-glycoprotein (p-gp). One of the effective ways to overcome multidrug resistance is to inhibit the up-regulated p-gp. (-)-Antofine derived from Cynanchum paniculatum Kitagawa (Asclepiadaceae) exhibited a potential growth inhibition against various human cancer cell lines with the IC50 values of low nanomolar ranges. (-)-Antofine belongs to phenanthroindolizidine alkaloids. The mechanism of growh inhibition included the inhibition of DNA, RNA, protein synthesis, inhibition of dihydrofolate reductase activity, and induction of apoptosis. However, the comprehensive study for the investigation of mechanisms of phenanthroindolizidine alkaloids has not been fully elucidated yet. Although (-)-antofine exhibited a potential growth inhibition activity against various human cancer cell lines, there is a limit to apply (-)-antofine in clinical uses with structurally unstableity in vivo. On this line, novel (-)-HMAs which have an ability to overcome multidrug resistant as well as improved stability are still required. To determine the effect of (-)-HMA on the overcome the resistance and the cell viability, The paclitaxel-resistant human lung cancer cell A549-Pa was established by gradual exposure of paclitaxel with the IC50 value over 200 times compared to parent A549 cells. Although (-)-HMA showed 10 times lower inhibitory effect than (-)-antofine on the proliferation of A549-Pa cells and A549 parental cells. However, the IC50 values of (-)-HMA showed potectial activity with low nanomolar concentrations. The inhibitory effect against A549-Pa cells by (-)-HMA was also closely related to the down-regulation of p-glycoprotein expression on A549-Pa cells that overexpress P-glycoprotein. Further study exhibited that downregulation of P-gp by (-)-HMA was associated with the decreased expression of its mRNA MDR-1. The reversal effect of multidrug resistance of (-)-HMA was also evoked by the intracellular rhodamine-123 accumulation in A549-Pa cells. With the potential of (-)-HMA on the inhibition of human lung cancer cell and paclitaxel-resistant cell proliferation, The anti-tumor effect of (-)-antofine (5 and 10 mg/kg) using an in vivo tumor xenograft model was investigated. As a result, (-)-HMA significantly inhibited the tumor growth in both A549 and A549-Pa xenograft model without any overt side effect. These findings suggest that (-)-HMA might be a potential candidate as an antitumor agent along with the reversal effect of multidrug resistant cancers as well as the possibility of clinical application. Transforming growth factor-β (TGF-β) has a diverse effect in regulating cellular functions. In terms of tumorigenesis, TGF-β exhibits both tumor promoting and tumor suppressing activities. In early stages of tumor and normal epithelial cells, TGF-β functions as an antiproliferative factor which inhibits cell growth. However, most tumor cells may disturb the TGF-β pathway, turning the function from a tumor suppressor into a mediator of metastasis. On this line, TGF-β receptor antagonists have been reported to have an anti-cancer activity. In the present study, an anti-metastatic activity and the mechanisms of actions of ALK-5 inhibitor IN-1130 in cultured human prostate cancer PC-3 cells were investigated. First of all, SRB assay revealed that IN-1130 showed a very weak inhibitory effect on the proliferation of PC-3 cells with the IC50 values of over than 100 μM. However, IN-1130 inhibited TGF-β-mediated the cell migration in the wound healing and colony dispersion assay. IN-1130 also inhibited the enhanced invasion of PC-3 cells by TGF-β through Matrigel system. One plausible mechanism involved in the suppression of migration and invasion was associated with the down-regulation of MMP-9 and MMP-2 mRNA and protein expressions in PC-3 cells by IN-1130. In addition, gelatinolytic activity of MMP-9 in the zymography was also inhibited by IN-1130. In addition, IN-1130 up-regulated E-cadherin mRNA and protein expressions in PC-3 cells. These findings suggest that an ALK-5 inhibitor IN-1130 might have a potential of anti-metastatic activity, and thus will be a lead candidate for further development of anti-cancer agents.
