Chitosan을 이용한 조절 방출성 제제 개발을 위한 연구

Abstract

This study was conducted for the purpose of the controlled drug release by using biodegradable and biocompitible polysaccharide, chitosan. And also, colonic drug delivery system was expected since that chitosan ((1→4)-2-amino-2-deoxy-β-D-glucan) could be utilized by energy source of bacteria in colon. The result of the drug release from surface-crosslinked chitosan tablet using glutaraldehyde solution (cross-linker) and chondroitin encapsulated chitosan bead was obviously sustained comparing with chitosan tablet and gelled bead using TPP. Therefore, they may serve as a carrier for drugs targeted specifically to the colon.;생체 분해성과 생체 적합성이 우수한 polysaccharide인 chitosan을 이용한 조절 방출성 제제 개발을 목적으로 본 연구는 수행되었다. 또한, chitosan((1→4) -2-amino-2-deoxy-β-D-giucan )은 colon에 존재하는 bacteroids의 에너지 원천으로 사용되므로 colonic drug delivery system에의 적용을 목적으로 하였다. chitosan을 이용한 정제와 bead를 제조하여 약물 방출 실험을 행한 결과, glutaraldehyde 수용액에서 표면 가교시킨 chitosan 정제와 chondroitin encapsulated chitosan bead로부터의 약물 방출은 표면 가교시키지 않은 chitosan 정제와 tripolyphosphate로만 gelation시켜 만든 bead보다 현저히 지연되었고, colon bacteria에 의한 분해로 대부분의 약물 방출이 colon에서 일어날 수 있는 표적 지향(targeting)을 기대할 수 있었다.
