- 간조직 마이크로좀내 약물대사 효소는 endoplasmic reticulum(ER)에 주로 존재하여 세포막과 결합되어 있고 여러 내인적 물질과 외인적 물질 및 많은 다른 이물질 대사에 관여하는 전자 전달계로서 작용한다. 또한 이 효소계는 mixed-function oxidase 또는 monooxygenase라 불리우는 중요한 효소군이 존재하며 이들 효소군에는 cytochrome P-450이라는 중요한 성분이 포함되어 있다. cytochrome P-450은 이 효소계에서 기질과 산소의 결합 장소이며 terminal oxidase로 작용한다.
본 연구에서는 진경제인 phenobarbital과 polycyclic hydrocarbon인 3-me-thylcholanthrene과 같은 화합물들의 투여가 미성숙과 성숙 흰쥐의 간조직 마이크로좀내에서 monooxygenase system에 관여하는 효소 활성과 aminopyrine-N-demethylase와 P-nitrophenol-O-demethylase를 측정하고 동시에 배설과 대사의 중요 경로인 UDP-glucuronosyltransferase 활성을 측정하여 다음과 같은 결론을 얻었다.
1. Phenobarbital 투여군은 cytochrome P-450과 NADPH-cytochrome C reductase의 활성이 미성숙과 성숙 흰쥐에서 모두 대조군에 비해 통계학적으로 의의있게 증가하였다.
2. Phenobarbital 투여군은 aminopyrine-N-demethylase와 P-nitroanisole-O-demethylase의 활성이 미성숙과 성숙 흰쥐에서 모두 대조군에 비해 통계학적으로 의의있게 증가하였다.
3. Phenobarbital 투여군은 기질이 P-nitrophenol과 bilirubin에서 UDP-glucuronosyltransferase 활성이 미성숙과 성숙 흰쥐에서 대조군에 비해 증가 하였으나 증가률은 bilirubin-UDP-glucuronosyltransferase의 활성이 더 높았다.
4. 3-Methylcholanthrene 투여군은 cytochrome P-450과 NADPH-cytochrome C reductase의 활성이 미성숙과 성숙 흰쥐에서 대조군에 비해 의의있게 증가하였다.
5. 3-Methylcholanthrene 투여군은 aminopyrine-N-demethylase 활성이 미성숙과 성숙 흰쥐에서 모두 감소하였으나 통계학적인 의의는 없었다. 그러나 P-nitroanisole-O-demethylase의 활성이 미성숙과 성숙 흰쥐에서 모두 증가하였다.
6. 3-Methylcholanthrene 투여군은 UDP-glucuronosyltransferase 활성이 기질이 P-nitrophenol일때 미성숙과 성숙 흰쥐에서 모두 의의있게 증가하였다. 기질이 bilirubin일때 미성숙과 성숙 흰쥐에서 모두 의의있게 감소하였다.;Effects of phenobarbital(PB) and 3-methylcholanthrene(3-MC) on the hepatic drug-metabolizing enzyme system and UDP-glucuronosyltransferase in immatures and adults liver of rats were investigated. Intraperitoneal administration of PB(75mg/kg, 72, 48 and 24hr before death) to immature and adult rats significantly increased cytochrome p-450 content 415.0% and 261.0% of control immatures and adults, respectively.
Also NADPH-cytochrome C reductase activity in PB-treated immatures and adults were significantly increased to 355.2% and 361.2 % of the control, respectively. Aminopyrine(AM) P-nitroanisole(NA)-metabolizing activity were increased by administration of PB to 1044.5% and 385.5% in immatures and, 179.8% and 244.2% in adults of the control, respectively. On the other hand, intraperitoneal administration of 3-MC (20mg,/kg, 72, 48 and 24 hr before death) to immature and adult rats significantly increased cytochrome p-450 content to 317.8% and 152.3 % of control immatures and adults, respectively. Also NADPH -cytochrome C reductase activity in 3-MC-treated immatures and adults were increased to 118.7% and 182.7% of the control, respectively. AM-metabolizing activity was decreased by administration of 3-MC to 96.0% in immatures and 93.9% in adults of the control, respectively. NA-metabolizing activity was increased by administration of 3-MC to 148.2% in immatures and 158.1% in adults of the control, respectively.
These results suggest that AM and NA-metabolizing enzymes in immature and adult livers treated with PB or 3-MC posses the capacity to be induced, and responsiveness of the drug-metabolizing enzyme system to 3-MC administration may differ from PB administration.
Substrate specificity of liver microsomal UDP-glucuronosyltransferase towards P-nitrophenol and bilirubin changes after treatment of rats with phenobarbital and 3-methylcholanthrene. Phenobarbital increases the glucuronidation of bilirubin, whereas 3-methylcholanthrene stimulates the glucuronidation of P-nitrophenol.
The results suggest that several forms of UDP-glucuronosyltransferase exit in rat liver microsome which are selectively induced either PB or 3-MC.
