- Abstract
- 스테롤로부터 17-ketosteroid로의 미생물학적 전환은 스테로이드계 의약품 제조의 중요한 공급원이다.
따라서 본 연구에서는 스테롤로부터 17-ketosteroid를 생성시키는 Mycobacterium sp. NRRL B-3805의 세포추출액에서 스테롤 측쇄 분해 관련 효소의 활성과 성질을 조사하고 효소의 분리를 시도하였다.
1. Mycobacterium sp. NRRL B-3805 세포추출액에서 lithocholic acid 또는 5β-cholanic acid-3-one의 측쇄를 분해하여 5β-androstane-3, 17-dione으로 전환시키는 효소활성을 확인하였다.
2. Mycobacterium sp. NRRL B-3805의 세포추출액에 존재하는 스테롤 측쇄 분해 관련 효소는 Amberite XAD-2 또는 XAD-16에 흡착되는 성질을 보였다.
3. Mycobacterium sp. NRRL B-3805의 세포추출액을 ammonium sulfate fractionation과 DEAE-sepharose CL-6B column chromatography를 통하여 5β-cholanic acid-3-one의 측쇄를 분해하는 효소의 분리 정제를 시도하여 부분 정제하였다.;Sterol side chain cleavage which occurs in many bacteria, is key process for the conversion of sterol to 17-ketosteroid as a source for commercial preparation of steroidal drug.
We investigated the characteristics of sterol side chain cleavage activities in the cell-free extract of Mycobacterium sp. NRRL B-3805, and also tried purifications of the enzyme.
The result were as follows.
1. 20,000xg cell-free extracts of Mycobacterium sp. NRRL B-3805 showed the side chain cleavage activity to convert lithocholic acid or 5β-cholanic acid-3-one to 5β -androstane-3, 17-dione.
2. The side chain cleavage activity was bound to Amberite XAD-2 or XAD-16 resin, which might suggest a hydrophobic property of the activity protein(s).
3. A partial protein purification was achieved through ammonium sulfate fractionation and DEAE-sepharose column chromatography.
