dc.description.abstractThe purpose of this study was to determine the effect of ginseng protein fraction on DNA repair capacity by measuring the amount of labelled thymidine incorporated into DNA in the PUVA treated cells. GI fraction which has been reported to have a radioprotective effect was obtained by isolating and partially purifying Korean white ginseng. Semiconservative DNA synthesis of CHO-Kl cells were inhibited by trimethylpsoralen + near ultraviolet light (300-400 nm) (PUVA treatment) and then UV light (254nm) was irradiated at 10-30J/㎡ to induce DNA damage. The DNA repair capacity was increased at first and then decreased as the UV dose increased. However, when cells were treated with ginseng protein fraction before or after UV irradiation, the DNA repair capacity was kept increasing as the UV dose increased. This result suggests that the ginseng protein fraction increases the DNA repair capacity of CHO-Kl cells damaged by UV irradiation.;방사선 방어작용이 있는 인삼단백분획이 손상된 DNA의 회복능력을 증가시키는지를 결정하기 위하여 DNA에 대한 ^(3)H-thymidine의 incorporation을 측정함으로써 자외선에 의한 DNA손상의 회복능력에 미치는 인삼단백분획의 영향을 관찰하였다. 인삼중의 단백질 분획을 추출 부분정제하여 방사선 방어작용이 있는 GI분획을 얻었다. CHO-Kl세포에 PUVA(trimethylpsoralen + near ultraviolet ligth)처리를 하여 DNA의 정상적인 반보존적 합성을 저해한 후, 10, 20, 30 J/㎡의 자외선을 조사하였을 때 세포의 DNA회복능력은 처음에는 증가하다가 곧 감소하였다. 그러나 인삼단백분획을 자외선 조사 전 또는 후에 처리하였을 때에는 DNA회복능력이 계속 증가함을 발견하였다. 이러한 결과는 방사선 방어작용이 있는 인삼단백분획이 자외선에 의해 손상된 세포의 DNA회복능력을 증가시킴을 나타낸다.-
