칠면조 적혈구 中 β-아드레날린 수용체에 대한(-)-[(3)^H] Dihydroalprenolol의 결합양상

Abstract

To characterize the β-adrenergic receptors in intact turkey erythrocytes, (±)-[^(3)H] Dihydroalprenolol radio-ligand binding was studied in the presence of excess (±)-Isoproterenol (agonist) or (±)-Propranolol (antagonist). Isoproterenol-and Propranolol-displaceable [^(3)H]-DHA binding showed linear Scatchard plot (Kd; 13.3 and 14.7 nM, Bmax; 1392 and 1679 sites/cell) and Hill coefficients were 0.9308 and 0.9619, respectively. These results indicate the presence of only one class of site without cooperativity.;칠면조 적혈구의 β-아드레날린 수용체의 특성을 살펴보고자 온전한(intact)칠면조 적혈구에 대하여 효능약(agonist) 및 길항제(antagonist)를 각각 displacer 로 사용하여 방사성표식 (radiolabeled) 길항제인 (-)-[^(3)H] Dihydroalprenolol의 결합양상을 비교하였다. Isoproterenol displaceable 및 Propranolol displaceable[^(3)H]-DHA결합은 전부 Scatchard plot에서 직선 ( KD;13.3,14.7nM,Bmax;1392,1679 sites/cell)을 나타내었으며, Hill coefficient는 0.9308, 0.9619로 1.0에 가까웠다. 이 결과로 미루어 효능약 및 길항제에 대하여 공히 [^(3)H]-DHA는 칠면조 적혈구 세포막 β-아드레날린 수용체에 있어 한 가지 결합특성을 가짐을 알 수 있었다.
