Clonidine의 혈압강하 작용에 대한 삼환계 항우울약의 영향

Abstract

중추성 혈압 강하제 Clonidine은 임상적으로 삼환계 항우울약과 병용시 혈압 강하작용이 감소된다고 보고가 있어 흰쥐를 사용하여 Amitryptylene, Imipramine으로 전 처리한 후 Clonidine을 투여하였을 때의 혈압의 변화를 관찰하였고 α_(2)-adrenergic 길항약 Yohimbine으로 전 처리한 후 Clonidine을 투여하였을 때의 혈압의 변화와 비교하여 삼환계 항우울약이 Clonidine의 중추성 혈압강하작용에 미치는 영향을 검토하여 다음과 같은 결론을 얻었다. 1. Clonidine 120 ㎍/kg 투여로 현저한 혈압 강하작용이 나타났으며 Amitryptylene 600 ㎍/kg으로 전 처리후 Clonidine을 투여한 군은 최대 혈압 강하작용이 Clonidine 단독 투여군에 비하여 23.9 % 감소하였으며 매우 (P<0.001) 유의적이었다. 2. Imipramine 600 ㎍/kg으로 전 처리후 Clonidine을 투여한 군은 최대 혈압 강하작용이 Clonidine 단독 투여군에 비하여 33.4 % 감소하였으며 매우 (P<0.001) 유의적이었다. 3. Yohimbine 800 ㎍/kg 전처리후 Clonidine을 투여한 군은 최대 혈압 강하작용이 Clonidine 단독 투여군에 비하여 16.4 % 감소하였으며 유의적 (P<0.05) 이었다.;From clinical experience it has been known that the hypotensive action of centrally acting Clonidine may be antagonized by simultaneously administered Tricyclic Antidepressants. Thus this experiment was set up to investigate the effect of Clonidine on bloodpressure and to compare the interactions separately pretreated Amitryptylene, Imipramine and classical α_(2)-adrenoreceptor antagonist Yohimbine with Clonidine in Chloral hydrate anaesthetized rats. The summarized results are as follows; 1. Administration of Clonidine 120 ug/Kg remarkably reduced the rat's blood pressure. And in the case of Clonidine administration after Amitryptylene 600 ug/Kg pretreatment, maximum hypotensive effect of Clonidine was significantly reduced (p < 0.001) by 23.9% compared with Clonidine alone. 2. In the case of Clonidine administration after Imipramine pretreatment, maximum hypotensive effect of Clonidine was significantly reduced (p < 0.001) by 16.4% compared with Clonidine alone. 3. Clonidine administration after Yohimbine 800ug/Kg pretreatment, maximum hypotensive effect was reduced (p < 0.05) by 16.4% compared with Clonidine alone. On according to the results Tricyclic Antidepressants antagonized the hypotensive action of Clonidine which is more potent than classical adrenergic antagonist Yohimbine. This is due to inhibitory action of Tricyclic Antidepressants decrease the Norepinephrine reuptake by α_(2)-adrenoreceptor stimulation of Clonidine.
