Clonidine의 혈압강하작용에 미치는 Cocaine의 영향

Abstract

Clonidine은 중추성 혈압 강하제로 α_(2)-adrenoceptor에 대한 효능이 있다고 보고된 바 있으며, cocaine은 amines의 neuronal reuptak를 억제하여 norepinephrine의 작용을 증가시키므로 흰 쥐를 사용하여 cocaine으로 전 처리한 후 clonidine의 혈압 강화 작용에 미치는 영향을 관찰하였고, dopamine의 생합성 전구 물질인 L-dopa도 clonidine과 같이 중추성 교감 신경 흥분을 감소시킨다는 보고가 있어 L-dopa로 전 처리한 후 clonidine의 혈압 강하 작용에 미치는 영향을 관찰하였고, dopamine의 생합성 전구 물질인 L-dopa도 clonidine과 같이 중추성 교감 신경 흥분을 감소시킨다는 보고가 있어 L-dopa로 전 처리한 후 clonidine의 혈압 강하 작용에 미치는 영향을 관찰하였다. 이상을 a_(2)-adrenoceptor길항약인 yohimbine으로 전 처리한 후 clonidine을 투여하였을 때의 혈압 변화와 비교 검토하여 다음과 같은 결론을 얻었다. 1. 흰 쥐의 대뢰 정맥에 clonidine 120㎍/㎏ 단독 투여로 현저한 혈압 강하 작용이 나타났으며, cocaine 2.5㎍/㎏ 으로 전 처리하고 clonidine을 투여하였을 때 clonidine의 혈압 강하 작용이 clonidine단독 투여군에 비하여 억제되었다. 2. L-dopa 200㎍/㎏ 단독 투여로 혈압 강하 작용이 나타났으며, L-dopa로 전 처리하고 clonidine을 투여하였을 때 L-dopa의 혈압 강하 작용이 증가하였다. 3. yohimbine 800㎍/㎏으로 전 처리하고 clonidine을 투여하였을 때 clonidine의 혈압 강하 작용이 yohimbine에 의하여 길항되었다.;It has been reported that centrally acting antihypertensive agent, clonidine is α_(2)-adrenoceptor agonist. Cocaine prevents the reuptake of amines into the adrenergic neurones and potentiated the action of adrenaline. Thus, the effect of clonidine alone and in combination with cocaine on blood pressure has been investigated in chloral hydrate anesthetized rats. The intravenous administration of L-dopa, the metabolic precursor of dopamine as well as clonidine might be expected to diminish sympathetic nerve activity and reduce blood pressure in these rats. The effect of L-dopa alone and in combination with clonidine on blood pressure has been investigated in rats. Experimental comparisons with the influences of yohimbine, a typical α_(2)-adrenoceptor antagonist, on hypotensive effect are given. Administration of clonidine to rats pretreated with cocaine L-dopa and yohimbine respectively gives following results. 1. The intravenous administration to rats of clonidine (120ug/kg) predominently lowers blood pressure. Clonidine administration after cocaine(2.5mg/kg) pretreatment reduced the clonidine-induced decrease in blood pressure. 2. The intravenous administration of L-dopa (200ug/kg) showed the hypotensive effect. Clonidine (120㎍/kg) administration after L-dopa pretreatment increased the hypotensive effect of L-dopa. 3. Clonidine-induced decrease in blood pressure was antagonized by the α_(2)-adrenoceptor antagonist, yohimbine (800ug/kg).
