Abstract
A. A Kinetic Study on Aminolysis of 2-Pyridyl X-substituted-benzoates: Effect of Changing Leaving Group from 4-Nitrophenolate to 2-Pyridinolate on Reactivity and Mechanism Second-order rate constants (kN) have been measured spectrophotometrically for nucleophilic substitution reactions of 2-pyridyl X-substituted-benzoates 2d-h with a series of alicyclic secondary amines in H2O at 25.0 ± 0.1 oC. The kN values for the reactions of 2a-e are slightly smaller than the corresponding reactions of 4-nitrophenyl X-substituted-benzoates 2d-h (e.g., kN1a-e/kN8d-h = 1.1 ~ 3.1), although 2-pyridinolate in 2d-h is ca. 4.5 pKa units more basic than 4-nitrophenolate in 1a-e. The Brønsted-type plot for the aminolysis of 8f (X = H) is linear with βnuc = 0.77, which is typical for reactions reported previously to proceed through a stepwise mechanism with breakdown of a zwitterionic tetrahedral intermediate T± being the rate-determining step (RDS), e.g., aminolysis of 4-nitrophenyl benzoate 1c. The Hammett plot for the reactions of 8d-h with piperidine consists of two intersecting straight lines, i.e., ρ = 1.71 for substrates possessing an electron-donating group (EDG) while ρ = 0.86 for those bearing an electron-withdrawing group (EWG). Traditionally, such a nonlinear Hammett plot has been interpreted as a change in RDS upon changing substituent X in the benzoyl moiety. However, it has been proposed that the nonlinear Hammett is not due to a change in RDS since the corresponding Yukawa-Tsuno plot exhibits excellent linear correlation with ρ = 0.85 and r = 0.62. Stabilization of substrates 8d-h in the ground state has been concluded to be responsible for the nonlinear Hammett plot. B. A Kinetic Study on Aminolysis of 2-Pyridyl X-substituted-benzoates in Acetonitrile A kinetic study is reported for aminolysis of 2-pyridyl X-substituted-benzoates 2a-i with a series of alicyclic secondary amines in MeCN at 25.0 ± 0.1 oC. The Brønsted-type plot for the reactions of 2-pyridyl 3,5-dinitrobenzoate 2a is linear with βnuc = 0.58. It is proposed that the reactions of 2a-i in MeCN proceed through a forced concerted mechanism through an intramolecular H-bonding interaction in the aprotic solvent. The Hammett plot consists of two intersecting straight lines, while the Yukawa-Tsuno plot exhibits an excellent linear correlation with ρ = 1.28 and r = 0.63, implying that the nonlinear Hammett plot is not due to a change in rate-determining step but is caused by stabilization of substrates possessing an electron-donating group through resonance interactions. The reactions of 2a-i with piperidine proceed through a forced concerted mechanism in which nucleophilic attack and leaving group departure occur concertedly through a 6-membered cyclic transition state. This idea is supported by a normal deuterium kinetic isotope effect. Analysis of activation parameters also supports a concerted mechanism. C. Kinetic Study on Aminolysis of 4-Pyridyl X-substituted-benzoates: Effect of Substituent X on Reactivity and Reaction Mechanism A kinetic study is reported for nucleophilic substitution reactions of 4-pyridyl X-substituted-benzoates 3c-h with a series of alicyclic secondary amines in H2O. The Brønsted-type plot for the reactions of 4-pyridyl benzoate 3f is linear with βnuc = 0.71. The corresponding reactions of 2-pyridyl benzoate 2f, which is less reactive than 3f, resulted in also a linear Brønsted-type plot with βnuc = 0.77. The fact that the more reactive 3f results in a smaller βnuc value appears to be in accord with the reactivity-selectivity principle. The aminolysis of 3f has been suggested to proceed through a stepwise mechanism in which breakdown of the intermediate is the rate-determining step (RDS). The Hammett plot for the reactions of 3d-h with piperidine consists of two intersecting straight lines, i.e., ρ = 1.47 for substrates possessing an electron-donating group (EDG) and ρ = 0.91 for those possessing an electron-withdrawing group (EWG). In contrast, the corresponding Yukawa-Tsuno plot exhibits excellent linear correlation with ρ = 0.79 and r = 0.56. Thus, it has been concluded that the nonlinear Hammett plot is not due to a change in the RDS but is caused by stabilization of the ground state of the substrates possessing an EDG through resonance interaction between the EDG and the C=O bond of the substrates. D. Electronic Nature of Substituent X Governs Reaction Mechanism in Aminolysis of 4-Pyridyl X-Substituted-Benzoates in Acetonitrile A kinetic study is reported for aminolysis of 4-pyridyl X-substituted-benzoates 3a-i. Plots of pseudo-first-order rate constants (kobsd) vs. [amine] curve upward for the reactions of substrates possessing a strong electron withdrawing group in the benzoyl moiety(3a-d) but are linear for the reactions of those bearing an electron-donating group (3e-i), indicating that the electronic nature of substituent X governs the reaction mechanism. The k1k2/k–1 and k1k3/k–1 values were calculated from the intercept and slope of the linear plots of kobsd/[amine] vs. [amine], respectively. The Hammett plot for k1k2/k–1 consists of two intersecting straightlines, while the Yukawa-Tsuno plot exhibits an excellent linear correlation with ρ = 0.41 and r = 1.58, implying that the nonlinear Hammett plot is not due to a change in rate-determining step but is caused by stabilization of substrates possessing an electron-donating group through resonance interactions. The small ρ suggests that the k2/k–1 ratio is little influenced by the nature of substituent X. The Brønsted-type plots for aminolysis of 4-pyridyl3,5-dinitrobenzoate 3a are linear with βnuc = 0.98 and 0.79 for k1k2/k–1 and k1k3/k–1, respectively. The effect of amine basicity on the microscopic rate constants is also discussed. E. Kinetics and Reaction Mechanism of Aminolyses of Benzyl 2-Pyridyl Carbonate and t-Butyl 2-Pyridyl Carbonate in Acetonitrile Second-order rate constants (kN) have been measured spectrophotometrically for the reactions of benzyl 2-pyridyl carbonate 4 and t-butyl 2-pyridyl carbonate 5 with a series of alicyclic secondary amines in MeCN at 25.0 ± 0.1 oC. Substrate 5 is much less reactive than 4 and the steric hindrance exerted by the bulky t-Bu group in 5 has been attributed to its decreased reactivity. The Brønsted-type plots for the reactions of 4 and 5 are linear with βnuc = 0.57 and 0.45, respectively. Thus, the reactions have been concluded to proceed through a concerted mechanism, although the current reactions were expected to proceed through a stepwise mechanism with a zwitterionic tetrahedral intermediate T±. It has been proposed that the rate of leaving-group expulsion is accelerated by the intramolecular H-bonding interaction in T± and the “push” provided by the RO group through the resonance interaction. Thus, the enhanced nucleofugality forces the reactions to proceed through a concerted mechanism. The reactivity-selectivity principle (RSP) is not applicable to the current reaction systems, since the reaction of the less reactive 5 results in a smaller βnuc than that of the more reactive 4. Steric hindrance exerted by the bulky t-Bu group in 5 has been suggested to be responsible for the failure of the RSP. F. A Kinetic Study on Aminolysis of Benzyl 4-Pyridyl Carbonate in Acetonitrile: Effect of Changing Leaving Group from 2-Pyridyloxide to 4-Pyridyloxide on Reactivity and Reaction Mechanism A kinetic study is reported for nucleophilic substitution reactions of benzyl 4-pyridyl carbonate 6 with a series of alicyclic secondary amines in MeCN. The plot of pseudo-first-order rate constant (kobsd) vs. [amine] curves upward, which is typical for the reaction proceeding through a stepwise mechanism with two intermediates, a zwitterionic tetrahedral intermediate T± and its deprotonated form T–. Dissection of kobsd into the second- and third-order rate constants (i.e., Kk2 and Kk3, respectively) reveals that Kk3 is significantly larger than Kk2, indicating that the reactions proceed mainly through the deprotonation pathway (i.e., the k3 process). It is found that that the reactions reported previously to proceed through T± and T– show a small k2/k–1 ratio by decreasing k2 and/or by increasing k–1. An intramolecular H-bonding interaction, which was proposed to accelerate the rate of leaving-group expulsion (i.e., an increase in k2) for the aminolysis of benzyl 2-pyridyl carbonate 4, is absent for the reactions of 6. Thus, it is concluded that the reactions of 6 result in a small k2 (and/or a small k2/k–1 ratio), which causes the reactions to proceed through a stepwise mechanism with T± and T– as intermediates. G. Aminolysis of Y-Substituted-Phenyl 2-Methoxybenzoates in Acetonitrile: Effect of ortho-Methoxy Group on Reactivity and Reaction Mechanism Second-order rate constants (kN) were measured for aminolyses of Y-substituted-phenyl 2-methoxybenzoates 7a-i and 4-nitrophenyl X-substituted-benzoates 8a-k in MeCN at 25.0 oC. The Brønsted-type plot for the reactions of 7a-i with piperidine curves downward, indicating that a change in rate-determining step (RDS) occurs. The Hammett plot for the reactions of 8a-k with piperidine consists of two intersecting straight lines, which might be taken as evidence for a change in RDS. However, the nonlinear Hammett plot has been suggested not to be due to a change in RDS, but due to stabilization of the ground state of substrates possessing an electron-donating group (EDG) (e.g., 8i-k) through a resonance interaction, since the corresponding Yukawa-Tsuno plot exhibits an excellent linear correlation with ρ = 0.53 and r = 1.52. The ρ value found for the reactions of 8a-k in MeCN is much smaller than that reported previously for the corresponding reactions in H2O (i.e., ρ = 0.75). It is proposed that the reactions of 8a-k in MeCN proceed through a forced concerted mechanism due to instability of T± in the aprotic solvent, while the reactions of 7a-i proceed through a stepwise pathway with a stabilized T± through an intramolecular H-bonding interaction. H. Decomposition of Paraoxon and Parathion by Amines, HOO– and OH– Ions: Reaction Mechanism and Origin of the α-Effect The second-order rate constants have been measured spectrophotometrically for the reactions of paraoxon 9 and parathion 10 with a series of alicyclic secondary amines, OH− and HOO− ions in H2O at 25.0 ± 0.1 oC. A linear Brønsted-type plot with βnuc = 0.40 was obtained for the reactions of 9 with amines and OH−. The reaction has been concluded to proceed through a concerted mechanism. HOO− deviates positively from the linear Brønsted-type plot, implying that the α-effect is operative. The magnitude of the α-effect (kHOO−/kOH−) was found to be ca. 55 for the reaction of 9 and 290 for that of parathion 10, indicating that HOO− is highly effective in decomposition of the toxic phosphorus compounds although it is over 4 pKa units less basic than OH−. Among the theories suggested as origins of the α-effect (e.g., TS stabilization through an intramolecular H-bonding interaction, solvent effect, and polarizability effect), polarizability effect appears to be the most important factor for the α-effect in this study, since the polarizable HOO− exhibits a larger α-effect for the reaction of the more polarizable substrate 10.;본 논문은 아래와 같이 8가지의 친핵성 치환반응에 관한 것으로 각각의 논문 개요는 다음과 같다. (A) 물에서 2-pyridyl X-substituted-benzoates 2d-h와 고리형 이차 아민의 친핵성 치환 반응에서 이차 속도상수(kN)를 분광학적인 방법으로 측정하였다. 2d-h의 이탈기 2-pyridinolate는 4-nitrophenyl X-substituted-benzoates 1a-e의 이탈기 4-nitrophenolate 보다 약 4.5pKa 단위 더 강한 염기이지만, 2d-h반응의 kN값은 1a-e보다 약간 작다(kN1a-e/ kN2d-h = 1.1~3.1). 2-pyridyl benzoate 2f의 가 아민 분해반응에 대한 Brønsted-type plot은 ßnuc값이 0.77이고 R2 = 0.991인 직선이다. 2-pyridyl benzoate 2f는 4-nitrophenyl benzoate 1c 처럼 zwitterionic tetrahedral intermediate T±가 분해되는 단계가 속도결정단계(Rate-Determining Step, RDS)인 stepwise 메커니즘으로 진행되는 전형적 반응이다. 2d-h와 piperidine반응의 Hammett plot은 2개의 교차하는 직선으로 구성된다. Electron-donating group (EDG)을 가지는 기질은 ρ = 1.71이고, electron-withdrawing group (EWG)은 ρ = 0.86이다. 전통적으로 이런 비선형 Hammett plot은 benzoyl 부분에서 X 치환기의 변화에 따라 속도결정단계가 변하는 것으로 설명되어왔다. 그러나 본 연구에서 관찰된 비선형 Hammett plot은 Yukawa-Tsuno plot이 ρ = 0.86, r = 0.62 인 좋은 직선을 가지기 때문에 속도결정단계의 변화 때문이 아니라고 제안할 수 있고 비선형 Hammett plot의 원인은 공명구조에 의한 기질의 바닥상태가 안정화되기 때문이라고 결론지을 수 있다. (B) MeCN에서 2-pyridyl X-substituted-benzoates 2a-i와 고리형 이차 아민의 가 아민 분해반응을 수행하였다. Brønsted-type plot은 ßnuc값이 0.58이고 R2 = 0.998인 직선이었다. 2-pyridyl X-substituted-benzoates 2a-i 는 이탈기인 2-pyridyl 부분과 아민의 분자내 수소결합 상호작용에 의해서 2-pyridyloxide의 nucleofugality를 증가시켜 2a-i의 반응을 concerted 메커니즘으로 진행하게 한다. Hammett plot은 2개의 직선으로 구성되는데, X치환기가 EWG에서 EDG으로 변할 때 기울기 ρ는 1.33에서 1.94로 변하였다. 이러한 비선형 Hammett plot은 전통적으로 RDS의 변화로 설명했다. 그러나 본 연구에서 관찰된 비선형 Hammett plot은 Yukawa-Tsuno plot이 ρ = 1.28, r = 0.63 인 좋은 직선을 가지기 때문에 속도결정단계의 변화 때문이 아니라 바닥상태에서 기질 2a-e의 안정화가 비선형 Hammett plot의 원인이라고 결론지을 수 있다. 또한 2f와 중수소로 치환된 piperidine과의 반응에서 정상적인 동위 원소 효과가 나타나는 사실로부터 6각형 고리 구조를 통하여 친핵체의 공격과 동시에 이탈기의 이탈이 일어나는 concerted 메커니즘이라고 결론지을 수 있었다. Activation parameter를 구해 분석해 봄으로써 위에서 제안한 반응 메커니즘을 보다 명확하게 증명할 수 있었다. (C) 물에서 4-pyridyl X-substituted-benzoates 3a-e와 고리형 이차 아민의 친핵성 치환 반응을 수행하였다. 4-pyridyl benzoate 3c 반응의 Brønsted-type plot은 ßnuc값이 0.71인 직선이었다. 2-pyridyl benzoate 2c는 3c보다 반응성이 작지만 Brønsted-type plot의 ßnuc값이 0.77인 직선이었다. 반응성이 더 좋은 3c가 더 작은 ßnuc 값을 갖는 사실은 반응성-선택성 원리(Reactivity Selectivity Principle, RSP)에 일치하여 나타난다. 3c의 가 아민분해 반응은 중간체의 분해가 속도결정단계(RDS)인 stepwise 메커니즘으로 진행된다. 3a-e와 piperidine의 반응에서 Hammett plot은 2개의 교차되는 직선으로 구성되는데, 기질이 EDG을 가질 때 기울기 ρ는1.47이고 EWG을 가질 때 ρ는 0.91이다. 반면에, Yukawa-Tsuno plot은 ρ = 0.79, r = 0.56인 좋은 직선 관계를 나타냈다. 그러므로 본 연구에서 관찰된 비선형 Hammett plot은 속도결정단계의 변화 때문이 아니라 기질의 C=O결합과 EDG사이의 공명 상호작용에 의한 바닥상태의 안정화 때문이라고 결론지었다. (D) MeCN에서 4-pyridyl X-substituted-benzoates 3a-i와 고리형 이차 아민의 가 아민 분해반응을 수행하였다. Benzoyl 부분에 강한 전자끌개 그룹(3a-d)을 가진 기질의 반응에서 유사일차반응 속도상수 (kobsd) vs. [amine] 의 그래프는 위로 휘어졌고, 반면에 전자 주개 그룹(3e-i)의 반응은 직선을 나타냈다. 이것은 치환기 X 의 성질이 반응메커니즘에 영향을 미친다는 것을 의미한다. k1k2/k–1와 k1k3/k–1값은 각각 kobsd/[amine] vs. [amine]의 직선 그래프에서 기울기와 절편으로부터 계산되었다. k1k2/k–1의 Hammett plot은 2개의 교차하는 직선으로 구성되는데, Yukawa-Tsuno plot은 ρ = 0.41, r = 1.58인 좋은 직선 관계를 나타냈다. 본 연구에서 관찰된 비선형 Hammett plot은 속도결정단계의 변화 때문이 아니라 EDG을 가진 기질의 공명 상호작용에 의한 바닥상태의 안정화가 원인이 된다. 작은 ρ값은 k2/k–1 비가 치환기 X의 성질에 의해서 적게 영향을 받는다는 것을 나타낸다. 4-pyridyl 3,5-dinitrobenzoate 3a의 가 아민 분해반응에서 Brønsted-type plot은 직선이고 k1k2/k–1와 k1k3/k–1의 βnuc 값은 각각 0.98와 0.79이다. 그리고 미시적 속도 상수에서 아민의 염기도 효과를 논의 하였다. (E) MeCN에서 benzyl 2-pyridyl carbonate 4 그리고 t-butyl 2-pyridyl carbonate 5 와 고리형 이차 아민의 반응에 대하여 이차 속도상수(kN)를 분광학적으로 측정하였다. 기질 5는 4보다 반응성이 작은데 기질 5의 커다란 t-Bu group에 의한 입체장애 때문에 반응성이 감소되었다. 4과 5의 반응에 대한 Brønsted-type plot의 βnuc값은 각각 0.57과 0.45인 직선이다. 그러므로, 현재 반응이 zwitterionic tetrahedral intermediate T±를 갖는 stepwise 메커니즘으로 진행된다고 예상되었음에도 불구하고, 반응은 concerted 메커니즘으로 진행된다고 결론지었다. 이탈기가 떨어지는 속도는 T±의 분자내 수소결합 상호작용과 RO group의 “push”에 의한 공명 상호작용에 의해서 가속화되었다. 그러므로, 강화된 nucleofugality에 의해서 반응이 concerted 메커니즘으로 진행되었다. 반응성-선택성 원리(RSP)는 현재 반응 체계에서 해당되지 않는데, 반응성이 작은 5의 반응이 반응성이 더 큰 4보다 작은 βnuc값을 가지기 때문이다. 5의 커다란 t-Bu group에 의한 입체장애는 RSP가 적용되지 않는 원인이 된다. (F) MeCN에서 benzyl 4-pyridyl carbonate 6와 고리형 이차 아민의 친핵성 치환반응을 수행하였다. 유사일차반응 속도상수 (kobsd) vs. [amine]의 그래프는 위로 휘어지는 곡선을 나타냈고, zwitterionic tetrahedral intermediate T±와 그것의 deprotonated 형태인 T−, 즉, 두 개의 중간체를 가지고 stepwise 메커니즘으로 진행된다. kobsd를 이차속도상수(Kk2), 삼차속도상수(Kk3)로 나누었다. Kk3는 Kk2보다 상당히 큰 값을 가지는데, 이것은 아민의 농도가 진한 부분에서는 반응이 주로 deprotonation 과정(k3 과정)을 거쳐 진행된다는 것을 가리킨다. 이전에 보고되었던 benzyl 2-pyridyl carbonate 4의 반응은 forced concerted 메커니즘으로 진행되었다. 분자내 수소결합은 4의 반응에서 이탈기의 이탈속도를 가속화 시키므로(k2의 증가), concerted 메커니즘으로 진행된다. 그러나, 6의 반응에서는 이런 수소결합에 의한 k2의 증가는 구조적으로 불가능하다. 그러므로 수소결합의 유무가 상반되는 반응메커니즘의 원인이 된다고 주장하였다. (G) MeCN에서 Y-substituted phenyl 2-methoxybenzoates 7a-i와 4-nitrophenyl X-substituted-benzoates 8a-j의 가 아민 분해에 대하여 이차 속도상수(kN)를 측정하였다. 7a-i와 piperidine의 반응에 대한 Brønsted-type plot에서 아래로 휘어지는 곡선은, RDS의 변화가 일어나는 것을 의미한다. 8a-j와 piperidine의 반응에서 Hammett plot은 2개의 직선으로 구성되어 있는데, 이것은 보통 RDS 변화의 증거로 제시되었다. 그러나, 본 연구에서 관찰된 비선형 Hammett plot은 RDS의 변화 때문이 아니라 EDG을 가진 기질에서 공명상호작용을 통한 바닥상태의 안정화 때문이라고 할 수 있다. 왜냐하면 Yukawa-Tsuno plot에서 ρ = 0.53, r = 1.52인 좋은 직선 관계를 나타내기 때문이다. MeCN에서 8a-j 반응은 이전에 보고된 H2O에서의 반응(ρ = 0.75) 보다 훨씬 작은 ρ값을 갖는다. MeCN에서 8a-j의 반응은 비양성자성 용매에서 중간체 T±의 불안정성 때문에 forced concerted 메커니즘으로 진행하고, 7a-i의 반응은 분자내 수소결합 상호작용을 통하여 중간체 T±가 안정화되어 stepwise 메커니즘으로 진행한다고 제안하였다. (H) paraoxon 9, parathion 10 과 고리형 이차아민, OH− 그리고 HOO−이온의 반응에서 이차 속도상수(kN)를 분광학적으로 측정하였다. 9와 고리형 이차아민과 OH−의 반응에서 Brønsted-type plot은 βnuc값이 0.40인 직선이므로 반응은 concerted 메커니즘으로 진행된다고 결론지었다. HOO–는 직선 Brønsted-type plot에서 분명하게 벗어나는데, 이것은 알파효과를 나타낸다. 알파효과의 크기(kHOO−/kOH−)는 9의 반응에서 약 55배, 10은 290배로 관찰되었다. HOO−는 OH−보다 염기도가 4pKa 이상 작음에도 불구하고 9, 10과 같은 독성이 강한 인산 유도체를 더 효과적으로 분해한다.
