Abstract
Use of influenza vaccines that target highly variable surface glycoproteins hemagglutinin (HA) and neuraminidase (NA) causes inconvenience that the recipients must be vaccinated each flu season and that the vaccination cannot offer protection against diverse strains and subtypes of influenza virus beside the concurrently circulating strain. For this reason, development of influenza virus vaccines that offers broad heterosubtypic protection against multiple strains and subtype of influenza virus is needed. Due to the conservancy in protein sequences among different subtypes of influenza virus, vaccines that induce CD8 T cell response targeting the conserved influenza virus protein epitope may offer cross-protection against infections cause by different influenza virus subtypes. In this study, we generated recombinant replication-deficient adenovirus (rAd) vectored vaccine (rAd/NP) expressing the nucleoprotein (NP) of influenza A virus (A/PR/8/34). BALB/c mice were immunized intranasally with single-dose of rAd/NP or sublingually with rAd/NP using prime-and-boost regimen and subsequently challenge with lethal doses (10LD50) of homosubtypic or heterosubtypic influenza virus. Our results indicate that mice that were given i.n. immunization of rAd/NP were conferred with complete protecting following lethal homosubtypic challenge. Also, mice that were given i.n. immunization of recombinant vaccine at higher dose were conferred with protection following lethal heterosubtypic challenge. Additionally, our results demonstrate that delivery of rAd/NP via both immunization routes induced CD8 T-cell responses against immunodominant Kd-restricted NP147-155 epitope with intranasal immunization causing higher frequency of NP-specific CD8 T-cells recruitment to the lungs compared to that caused by the sublingual immunization. We believe that the observed homo- and hetero-subtypic protection may be conferred by mechanisms involving influenza NP-specific mucosal IgAs and NP-specific CD8 T cells. Together, these results suggest that a single intranasal administration of recombinant adenovirus-based vaccine encoding influenza NP protein is a potential vaccine regimen that could offer broad-ranged protection against various subtypes of influenza virus. ;현재 인플루엔자 백신은 인플루엔자의 표면 당단백질인 hemagglutinin (HA)와 neuraminidase (NA)를 타깃으로 만드는데 이 단백질들은 변이율이 높아 예방접종을 하더라도 다양한 백신주 및 아형에 대해 방어효능이 감소하고 매년마다 새로운 독감에 대비하여 예방접종을 해야 한다. 이러한 이유로 인플루엔자 바이러스의 각기 다른 아형에 대해서 넓은 방어능력을 제공해주는 백신개발이 시급하다. 다른 아형간 동일하게 보존되어있는 시퀀스가 있기 때문에 이 단백질의 에피토프를 타깃으로 CD8 T cell반응을 유도하는 백신은 다양한 종류의 인플루엔자의 감염에도 교차방어능력을 제공할 수 있을 것이다. 본 연구를 위해서 인플루엔자 A 바이러스 (A/PR/8/34)의 nucleoprotein (NP)를 발현하는 재조합 복제불능 아데노바이러스 백신 (rAd/NP)을 제조하였다. BALB/c 마우스에 rAd/NP백신을 단일용량으로 비강내로 면역화를 시키거나 프라임-부스팅 요법을 설하로 면역화를 시킨 뒤, 동형 혹은 다양한 아형의 인플루엔자 바이러스를 치사량 (10 LD50)으로 감염시켰다. 비강내로 rAd/NP 백신을 면역화 시킨 마우스 그룹에서 치사량의 동형 바이러스 감염에 대해 완벽한 방어능력을 보였으며 특히, 높은 용량을 면역화 시켰을 때는 동형뿐만 아니라 다른 아형의 인플루엔자 감염에도 방어능력을 보였다. 각 루트별로 rAd/NP를 면역화 시켰을 때 immunodominant Kd-restricted NP147-155 에피토프에 대한 CD8 T cell반응이 유도되었으며 설하 면역보다 비강내로 면역화된 마우스에서 NP-특이 CD8 T cell이 폐에 보다 많이 모여드는 것을 확인할 수 있었다. 또한 비강내 면역을 준 마우스 그룹에서만 virus-특이 점막 IgAs가 분비되었음을 미루어 볼 때, 인플루엔자 바이러스의 동형 및 각각의 아형의 감염에 대한 방어 능력은 NP-특이 점막 IgAs 와 NP-특이 CD8 T cell의 상호관계에 대한 메커니즘에 의해서 생성된 것이라 여겨진다. 이러한 연구 결과는 인플루엔자 NP 단백질을 발현하고 있는 아데노바이러스를 기반으로 제조한 백신을 비강내로 단일 투여했을 때 넓은 범위의 방어능력을 제공함으로써 인플루엔자 바이러스의 다양한 아형에 대한 교차방어능력을 유도할 수 있는 인플루엔자 백신 후보가 될 수 있음을 보여준다.
