Abstract
소비자들의 건강에 대한 관심이 증가함에 따라, 최근 식품 산업에서는 조미료를 대체하는 천연 식품 소재의 이용이 각광 받고 있다. 효모는 GRAS로 분류되어 있으며 천연 풍미소재로 각광받고 있는 소재이다. 이러한 효모에서 얻을 수 있는 효모추출물을 천연 풍미 소재로 이용하기 위해서는 효모의 RNA 핵산 함량을 높이는 것이 중요하다. 그러나 효모가 다른 미생물에 비해 RNA 함량이 높음에도 불구하고 효모추출물 제조에 사용되는 대부분의 기존 효모는 특허화 되어있거나, 핵산 함량이 낮거나, 값이 비싸므로 산업적 이용을 위해 추가적인 연구가 필요한 상황이다. 본 연구에서는 효모의 RNA 생산량을 높이기 위하여 gTME(global transcription machinery engineering)를 이용하였다. gTME는 전반적인 전사체를 조절하는 주요 단백질을 변환하는 것을 통해 전사단계에서 새로운 형태의 다양한 phenotype를 생성하는 것을 목적으로 연구된 방법이다. 효모의 TATA binding protein인 SPT15의 무작위 돌연변이를 유발하여 RNA polymerase의 transcription factor의 활성에 변화를 주어 RNA 함량을 높일 수 있다고 기대해 볼 수 있다. gTME로 생성된 약 5만개의 Ura+ mutants를 일반배지와 KCl이 함유된 배지에 각각 tooth picking 하여 일반배지에서는 잘 자라고 KCl 배지에서는 잘 자라지 않는 1,114 균주를 선별하였다. 선별된 균주는 orcinol 방법으로 RNA 정량을 하였으며, 그 중 가장 높은 RNA 함량을 나타낸 M132-484 균주를 선별하였다. M132-484 균주와 모균주를 배양 36시간이 지난 후 RNA 함량을 비교해 본 결과, 탄소원으로 glucose, 질소원으로 peptone를 이용한 배양조건에서 모균주에 비해 RNA 함량이 약 27.6% 증가하였다. 또한 pH 4.5의 조건에서 RNA 함량이 16.8%가 추가적으로 증가되었음을 확인 하였다. 위와 같은 조건에서 1L 발효기를 이용하여 배양하였더니 RNA 함량이 약 23.6% 증가하는 것을 확인할 수 있었다. SPT15의 sequencing 각각 13, 16, 126 번째 위치한 codon이 alanine에서 glutamic acid로, isoleucine에서 lysine으로, alanine에서 valine으로 mutation이 발생하였음을 확인하였다. 나아가 천연 정미성 소재인 효모추출물을 보다 경제적으로 이용 할 수 있는 가능성을 보여주었다. ;Yeast extracts are natural flavor globally used for a variety of foods. Yeast is easy to culture and has relatively high RNA content, and yeast is used for flavorous nucleotide, the building block of RNA, such as 5'-IMP (5'-inosine monophosphate) or 5'-GMP (5'-guaonsine monophosphate). To produce yeast with high flavorous nucleotide content, selection or improvement of high RNA containing yeast is prerequisite. In this study, transcription factor SPT15 of S.cerevisiae was mutated using gTME method and SPT15 mutant library was established. After transformation into S.cerevisiae Y2805, mutants were screened for high RNA content yeast strains. Successful transformed mutant strains from gTME mutant library were able to survive on ura gene deficiency media, since ura3 gene was transformation selective maker. Secondly, acidic-RNase restricted strains were selected by using growth ability on environment containing K+ ion. Lastly, RNA contents of mutant strains were measured and selected strains were characterized to determine growth condition accumulating higher RNA content. Growth conditions of mutant with higher RNA content were determined using various carbon sources and nitrogen sources at different pHs. The mutant strain, M132-484, accumulated approximately 24.2% higher RNA content than wild type strain when glucose and peptone used at pH 4.5. In this mutant, it is found that there were three point mutations of SPT15. Interestingly, at 13th codon, alanine changed to glutamic acid in common with the other mutant strains with high RNA. The strategy may contribute to natural flavor industry by producing higher RNA content in yeast.
