dc.description.abstract | 본 연구는 온도가 증가함에 따라(가역적 열 젤화성질) 졸젤 전이를 보이는 수화젤 시스템은 그것의 간단한 가공과 외과적인 수술 절차 없이도 약제와 세포/줄기 세포에 심을 수 있는 전도 유망한 시스템 중 하나로써 제안되었다. 이 논문에서는, 폴리 펩타이드를 기반으로 한 하이드로젤을 연구하였다. 폴리 (에틸렌 글리콜) block 폴리 ( 알라닌 – co 페닐알라닌 ) (PEG-PAF) 수화젤은 온도가 증가함에 따라 졸젤 전이를 일으킨다. 3.0 ~ 7.0 wt. % 의 낮은 농도에서 졸-젤 전이를 보이며, FTIR, circular dichroism, 13CNMR,TEM사진, dynamic light scattering 연구를 통해서 졸-젤 메커니즘을 분석할 수 있었다. PEGPAF 수화젤은 졸-젤 전이 메커니즘에 따라 random coil에서 βsheets로 펩타이드의 작은 입체 구조 변화와 PEG의 탈수를 동반하는 마이셀 응집을 보인다. 또한 졸-젤 전이 온도와 젤 윈도우는 분자량, 구성 요소 또는 고분자의 입체화학을 다양하게 변화를 줌에 따라 조절할 수 있다. 폴리(에틸렌 글리콜)block폴리(알라닌) (PEGPA)은 나노 구조의 이차 구조 중요성과 폴리 펩타이드 블락 공중합체의 온도 민감성을 보기 위해 연구되었다. 생물학적 모티프로부터 잘 알려진 나노 구조 조절은 재료 과학자들에 의해 관심을 얻고 있다. L폴리알라닌의 βsheet구조는 양친성 공중합체인 폴리(에틸렌 글리콜)L/또는 DL폴리 알라닌 이중 공중합체의 졸-젤 전이처럼 섬유 나노구조를 발달시키는데 중요한 역할을 한다. L이성질체는 random coils에서 βsheets로 변화를 겪으며, 고분자 농도가 증가함에 따라 나노 섬유로 바뀐다. 반면에 DL이성질체는 어떠한 특정 나노 구조로의 변화 없이 random coil구조로 남아있는다. 미리 조립된 βsheet 이차 구조를 가진 L이성질체는 random coil구조인 DL이성질체보다 졸-젤 전이가 용이하며, 이는 오직 L이성질체만 생리학적으로 중요한 온도 범위인 20 ~ 40 oC에서 졸젤 전이를 보인다. PEGPAF의 주사 가능한 의약 전달 시스템으로써의 실현 가능성을 보기 위하여, 재료의 특징, 졸젤 전이 메커니즘, 그리고 생체 외/생체 내(쥐) 분화를 연구하였다. PEGPAF는 인산완충식염수 내에서는 안정하나, 쥐의 피하층 내에서는 분해되었다. 생체 외 실험에서는 포유류 조직의 피하층 내에서 보이는 카텝신 B, 카텝신 C, 그리고 엘라스타아제 같은 단백질 분해 효소들은 쥐 내에서 고분자 분해와 연관이 있다. 수용 인슐린 포뮬레이션의 싱글샷 (13.8 mg 인슐린/Kg)은 쥐 내에서 18일 넘게 혈당강하효과를 보인다. 또한 hGH 저장소가 만들어졌으며, 4일 동안 약의 방출 프로파일을 보였다. 이 논문은 폴리펩타이드의 계층 구조와 폴리펩타이드 수용액의 졸젤 온도민감성 전이 사이의 연관성, 조직공학의 insitu 젤 시스템, 세포/줄기 세포 치료, 그리고 의약 전달 시스템으로서의 가능성에 대한 기본적인 정보를 제공한다.;An aqueous system that undergoes a sol-to-gel transition as the temperature increases (reverse thermal gelation) has been suggested as one of the most promising implantable systems of pharmaceutical agents and cells/stem cells due to its simple fabrication and non-surgical implanting procedure. Here in this thesis, polypeptide based hydrogels are investigated. Poly (ethylene glycol)-block-poly(alanine-co-phenyl alanine) (PEG-PAF) aqueous solution undergoes sol-to-gel transition as the temperature increases. The sol-to-gel transition was observed at as low a concentration as 3.0 ~ 7.0 wt. %. Based on FTIR, circular dichroism, 13C-NMR, TEM images, dynamic light scattering studies, the sol-to-gel mechanism could be suggested. Micellar aggregation accompanying small conformational changes of the peptide from random coils to βsheets and dehydration of PEG is suggested as the sol-to-gel transition mechanism of the PEG-PAF aqueous solution. The sol-to-gel transition temperature and gel window could be tuned by varying the molecular weight, the composition, or the stereochemistry of the polymer. Poly (ethylene glycol)-block-poly(alanine) (PEG-PA) was investigated to see the significance of secondary structure in nanostructure formation and thermosensitivity of polypeptide block copolymer. Well-defined nanostructural control from biological motifs is gaining attention among the material scientists. The β-sheet structure of L-polyalanine plays a critical role in developing a fibrous nanostructure as well as sol-to-gel transition of amphiphilic poly(ethylene glycol)-L/or DL-polyalanine diblock copolymers. L-isomers underwent transitions from random coils to β-sheets, and to nanofibers as the polymer concentration increased, whereas DL-isomer remained as a random coil structure without developing any specific nanostructure. The L-isomer with a preassembled β-sheet secondary structure facilitates the sol-to-gel transition than the DL-isomer with a random coil structure. Thus, only L-isomer showed sol-to-gel transition in a physiologically important range of 20 ~ 40 oC. To see the feasibility of the PEG-PAF as an injectable drug delivery system, the material characteristics, the mechanism of the sol-to-gel transition, and in vitro/in vivo (rats) degradation were investigated. The PEG-PAF is stable in phosphate buffered saline, however, it degraded in the subcutaneous layer of rats. In vitro study showed that proteolytic enzymes such as cathepsin B, cathepsin C, and elastase that are present in the subcutaneous layer of the mammalian tissue might be responsible for the degradation of the polymer in rats. A single shot of an aqueous insulin formulation (13.8 mg insulin/Kg) showed a hypoglycemic effect over 18 days in rats. Also an hGH depot was made and showed the 4 day release profile. This thesis provides fundamental information on the relationship between hierarchical structures of polypeptides and thermosensitive sol-gel transition of the polypeptide aqueous solution and the potential as an in-situ gelling system for tissue engineering, cell/stem cell therapy, and drug delivery. | - |
