Abstract
본 연구에서는 plantamajoside를 함유한 차전초 추출물의 항산화효과를 알아보기 위한 실험을 진행하였다. 본 연구는 이중맹검, 무작위 배정, 평행설계, 대조식품대조시험으로 진행되었으며, 모든 과정은 이화여자대학교 IRB에 따라 진행되었다. 피험자는 과체중이며 혈중 지질농도가 높은 30대 이상의 성인 남녀 40명으로, 신문광고, 지면광고와 인터넷 게시판을 통해 모집되었다. 연구 참여 기준에 의해 적합하다고 판정된 피험자는 2주 간의 phenolic compound를 함유하고 있는 식품에 대한 고갈식이 기간을 두었다. 2주 후 방문하여, 산화적 스트레스 유발을 위하여 고지방 식사 후 차전초 추출물의 섭취를 하게 하였으며 시험식품 섭취 후 6시간 동안 연구에 참여하였다. 40명의 대상자들을 무작위 배정을 통해 4군으로 나누었다. 군당 10명으로 대조식품군과 차전초 저용량 (5 g), 중용량 (10 g), 고용량 (20 g) 군으로 나누어졌다. 고지방식사로는 Egg Mc.Muffin, Sausage Mc.muffin 각각 하나씩과 hashbrown 2 조각을 섭취하도록 하였다. 그 후 대조군은 저과당, 말토덱스트린으로 만들어진 대조식품을 섭취하고, 시험식품군은 차전초 추출물을 용량별로 섭취하였다. 시험식품 섭취 전 공복상태와 시험식품 섭취 후 1, 2, 4 그리고 6시간 후에 정맥혈을 채취하여 지방산화와 관련하여 혈중 ox-LDL을 분석하였으며 지질대사와 관련하여 TG와 유리지방산 농도를 측정하였다. DNA 수준에서의 항산화 능력을 확인하기 위하여 고지방 식사로 인한 DNA 산화 정도를 얼마나 감소시켜 주는지 확인하기 위하여 comet assay를 수행하였다. 차전초 추출물 섭취군의 혈중 TG농도는 각각의 시험 물질 섭취 후 점차 증가하였고, 유리지방산 농도는 모든 시간대에서 차전초 추출물 고용량 군에서 낮은 경향을 보였지만 유의적이지는 않았다. 지질과산화 정도를 확인하기 위한 ox-LDL은 차전초 추출물 고용량 섭취군에서 다른 시험물질 섭취군에 비해 2시간과 4시간에 매우 낮아지는 경향을 보였으나 유의적이지는 않았다. 이것은 각 대상자 간에 차이가 커서 유의적인 결과를 얻지 못한 것으로 사료된다. comet assay를 통하여 DNA 손상 정도를 확인한 결과 시험물질 섭취 후 4시간까지는 유의적인 차이가 없다가 6시간 후에 3가지 용량의 차전초 추출물 섭취군이 대조군에 비하여 DNA 손상 정도가 유의적으로 낮아짐이 관찰되었다 (P = 0.0225). 결론적으로 차전초 추출물은 식 후 지방대사와 관련된 TG나 유리지방산 농도에는 변화를 주지는 않고 lipid 수준에서의 항산화 효과는 없는 것으로 관찰되었다. 하지만 산화적인 스트레스로 인한 DNA 손상 정도를 확인 한 결과 DNA 손상을 감소시켜줌을 확인할 수 있었다. 따라서 차전초 추출물이 지질수준에서 보다는 DNA수준에서 항산화 효과를 가지고 있다고 볼 수 있는 것으로 사료된다. 하지만 이번 연구는 단회 섭취를 통한 효과를 확인 한 것이므로 장기간의 차전초 추출물 섭취를 통한 차전초 추출물의 항산화능 평가 연구가 수행 되어야 할 것이다.;This study was performed to evaluate the antioxidant effects after acute consumption of Plantago asiatica L. using human subjects. This was a double-blind, randomized, placebo-controlled and parallel study. Fourty overweight hyperlipidemic subjects were recruited using newspaper and online advertisements on several websites in Seoul, Korea. Subjects were randomly divided into four groups. After 2-week run-in period, subjects visited the hospital and consumed either Plantago asiatica L. extract with low (5 g), intermediate (10 g), high (20 g) or placebo with a high-fat meal for loading oxidative stress in a fasting state. The high-fat meal have 976 kcal with consist of Egg Mc.Muffin, Sausage Mc.Muffin and two hashbrown petties. Blood samples were collected at fasting state and 1, 2, 4 and 6h after single-dose administration. The plasma oxidized-LDL (ox-LDL), triglycerides (TG) and free fatty acid (FFA) were measured for evaluate the antioxidant effects in lipid level. The ox-LDL concentrations were lowest at 120 min and 240 min in high dose of Plantago asiatica L. group. TG concentrations were not different among four groups at each time points. Postprandial FFA levels were lowest in high dose group at all time points. However, there was no significant effect of time and group interaction. For investigated in DNA level, we did comet assay. DNA-damage in lymphocytes was not significantly changes until 4 hours, but at 6 hours the percentage of DNA in the tail in Plantago asiatica L. groups were significantly decreased (P = 0.0225). According to this result, the single consumption of Plantago asiatica L. has no effect of modulation of lipid peroxidation in lipid level. However, Plantago asiatica L. extract have antioxidant effect in DNA level by protecting DNA from oxidative stress and reducing DNA-damage.
