Abstract
Outbreaks associated with Vibrio spp. frequently occur in Asia because of the dietary habit of eating raw seafood. Up to now, 34 Vibrio spp. are discovered, 12 of which are pathogenic to human beings. Among them, Vibrio parahaemolyticus, V. vulnificus and V. cholerae are reported to be major human pathogens around the world including Korea; V. parahaemolyticus and V. cholerae cause gastroenteritis and V. vulnificus is associated with wound infection. In this study, a multiplex PCR was developed to detect the three major pathogenic Vibrio spp. using species-specific PCR primers. The multiplex PCR yielded three distinctive PCR products specific to the individual species. The multiplex PCR was successfully validated against Vibrio type strains including nine Vibrio spp. including five V. parahaemolyticus, two V. vulnificus and one V. cholerae and six other Vibrio spp. This multiplex PCR method was applied to Vibrio isolates collected from sushi restaurants in Busan, Korea, and most isolates were identified as V. parahaemolyticus In addition, whole cell protein pattern analysis using SDS-PAGE was performed to determine phylogenetic difference of 20 Vibrio type strains: five V. parahaemolyticus, two V. vulnificus, one V. cholera and twelve other Vibrio spp. including pathogenic V. alginolyticus, V. fluvialis, and V. mimicus. The Vibrio species showed similar protein band patterns and all the Vibrio spp. showed different whole cell protein profiles, compared one another. When the protein patterns of 36 Vibrio isolates were analyzed, all the isolates were indentified as V. parahaemolyticus. RAPD-PCR was used to determine the unique PCR product profiles of 20 Vibrio type strains and 36 isolates identified as the genus Vibrio. Twenty 10-mers of random primers were tested to obtain the specific patterns of each Vibrio strain. Two primer sets, OPA-10 and OPA-13, were shown to successfully differentiate 20 Vibrio type strains. The selected primers were tested for identification of 36 Vibrio isolates. Amplified PCR products of all the isolates were confirmed as V. parahaemolyticus. In this study, the multiplex PCR together with whole cell protein pattern analysis and RAPD-PCR suggest that this method may facilitate specific, rapid and simple identification of major Vibrio spp. involved in foodborne diseases.;아시아권에서는 해산물을 날 것으로 먹는 식습관이 있어 비브리오속에 의한 식중독 발생이 자주 일어난다. 지금까지 34가지의 비브리오 속이 발견되었으며 이 중 12가지 비브리오 속이 사람에게 병원성이 있는 것으로 알려져 있다. 병원성 비브리오균 중 Vibrio parahaemolyticus, V. vulnificus와 V. cholerae가 한국 및 전 세계적으로 주요 식중독 균으로 알려져 있다. V. parahaemolyticus와 V. cholerae는 위장염을 일으키며 V. vulnificus는 장염 뿐만 아니라 패혈증과도 관련이 있다. 이번 연구에서 첫 번째로, multiplex PCR로 이들 세 가지 주요 식중독균을 검출하기 위해 종 특이적 프라이머를 개발하였으며, 세 가지 비브리오 속에 특이적인 각기 다른 세가지 PCR 산물을 나타내었다. Multiplex PCR은 다섯 가지의 V. parahaemolyticus, 두 가지의 V. vulnificus, 그리고 한 가지 V. cholerae를 포함한 총 20가지 비브리오 속에 대해 성공적으로 수행되었다. 개발한 프라이머의 확인을 위해 부산지역 횟집에서 채취한 비브리오 분리균주에 적용한 결과, 모든 균은 V. parahaemolyticus로 동정되었다. 다음으로 계통적 차이를 확인하기 위한 다섯 가지 V. parahaemolyticus, 두 가지 V. vulnificus, 그리고 한 가지 V. cholerae와 병원성의 V. alginolyticus, V. fuluvialis, V. mimicus를 포함한 총 20가지 비브리오 속에 대한 SDS-PAGE를 이용한 세포 단백질 패턴 분석이 수행되었다. 같은 비브리오속은 같은 단백질 밴드 패턴을 보였으며 서로 다른 종 간에는 각기 다른 밴드 패턴을 나타내었다. 36가지 분리균주의 단백질을 추출하여 밴드 패턴을 분석한 결과, 모든 균주는 V. parahaemolyticus인 것으로 나타났다. 마지막으로 RAPD-PCR 방법을 이용하여 20가지의 비브리오 표준균주와 36가지 분리균주에 대해 DNA 프로파일 분석을 실시하였다. 20가지의 10-mer oligonucleotide primer를 시험한 결과 OPA-10과 OPA-13 두 가지 프라이머를 이용하였을 때 각 비브리오 균주에 대해 각기 다른 특이적 밴드를 확인할 수 있었다. 선택된 두 프라이머를 36가지 비브리오 분리균주에 적용한 결과, 증폭된 밴드간에 약간의 차이를 보이나 주 밴드로 보았을 때 모두 V. parahaemolyticus로 나타났다. 비브리오 속에 대한 세 가지 실험의 결과로, multiplex PCR과 SDS-PAGE를 이용한 단백질 패턴 분석, 그리고 OPA-10, OPA-13 프라이머를 이용한 RAPD-PCR 방법을 함께 이용한다면, 비브리오균에 의한 식중독 사고의 발생 시 종간의 구분이 가능한 특이적이고 신속한 비브리오균의 검출이 가능할 것으로 생각된다.
