Abstract
Aspergillus niger van Tieghem was cultured by the method of synchronous and submerged culture. Throughout the culture sporulation occurred and its life cycle and differentiation were completed in the experiments. Acid-phenol soluble proteins in this fungus were extracted at, each stage of life cycle. Those APS proteins were separated by polyacrylamide gel electrophoresis, then the profiles of proteins were observed and discussed. The results of studies were as follows: 1. APS protein bands on polyacrylamide gels were obtained. Those proteins were extracted out of growing hyphae after germination, conidiophore stage mycelia, presporulation stage mycelia and sporulation stage mycelia. 2. It was found that the fundamental and structural proteins exist in mycelia all over the differentiation as those are common and major in all stages. 3. Several APS proteins were found to be variable in their profiles on the gels, some of them appeared in the sporulating mycelia. 4. Molecular weight of APS proteins those appeared during differentiation particularly in sporulation was determined indirectly by the method of polyacrylamide gel electrophoresis. 5. Electrophoretic mobilities of some standard enzymic proteins those had been dissolved in acid-phenol solution were examined with polyacrylamide gel electrophoresis, then the mobilities of those proteins were compared against their molecular weights. ;검정곰팡이(Aspergillus niger)를 실험 재료로 하여 동조적 액침배양의 방법으로 생활사를 회전시키고 포자형성까지의 분화를 재현하였다. 또한 생활사의 각 단계에 있어서의 아세트산-페놀가용성 단백질을 추출하여 폴리아크릴아미드 겔 전기이동법으로 단백질의 양상(Profile)을 연구하였다. 연구결과는 다음과 같았다. 1. 포자의 발아로부터 균사생장, 생식기관인 포자형성까지의 각 생활단계의 아세트산-페놀 가용성 단백질의 전기이동상을 얻었다. 2. 이 전기이동상을 비교한 결과 기본적 구조단백질이 공통적으로 존재하는 것을 알았다. 3. 분화과정에 따라서 3~4개의 APS 단백질이 신생 또는 소실되는 것을 알았다. 4. 이들 단백질의 분화과정에 따라 신생 또는 소실되는 구조단백질의 분자량을 폴리아크릴아미드 겔 전기이동법에 의하여 간접적으로 측정함으로써 그 분자량을 결정하였다. 5. 표품효소 단백질을 아세트산-페놀에 용해시켜서 폴리아크릴아미드 겔 전기이동을 실시하여 단백질의 이동도에 대한 분자량의 상관곡선을 얻었다.
