- Epithelial-mesenchymal transition (EMT)는 배아 형성과 종양 형성에 중요한 형태학적 변화 과정이다. EMT를 통해 상피 세포는 세포간 결합과 극성(polarity) 등, 그 특유의 성질을 잃고 간엽 세포화되어 이동성을 획득하게 된다. 이 과정에서 Transforming growth factor-β (TGF-β) signaling이 매우 중요한 역할을 한다. TGF-β는 Snail을 비롯한 EMT 조절 인자를 활성화하여 세포의 E-cadherin의 발현을 억제한다. 양성 종양 세포는 E-cadherin이 관여하는 세포간 결합을 잃고 악성 종양으로 변한다. 암세포는 EMT를 거쳐 이동성을 획득하고, 주변의 조직에 침투하여 혈관이나 림프관을 통해 이동한 뒤 다른 장기에 정착 후 증식하여 전이된다. 많은 실험과 임상 자료를 통해 TGF-β가 EMT에 미치는 영향 과, TGF-β signaling과 전이의 상관관계가 밝혀지면서 TGF-β signaling pathway를 표적으로 하는 항암제가 개발되고 있다.
본 논문에서는, TGF-β가 EMT와 유방암 전이에 미치는 영향과 TGF-β typeⅠreceptor kinase/Activin-like kinase 5 (TβRⅠ/ALK5) 저해제로 개발된 IN-1130의 항 전이 효과 및 기전을 알아보고자 유방암 세포주(MDA-MB-231, MCF-7)와 BALB/c xenograft mouse 동물 모델을 이용하여 실험하였다.
가장 먼저 TGF-β에 의한 EMT 관련 인자의 변화를 살펴 본 결과, in vitro에서 TGF-β는 EMT를 유도하는 전사 인자 Snail과 간엽 세포 지표인 N-cadherin의 발현을 증가시켰으며, 상피 세포 지표인 E-cadherin의 발현을 감소시켰다. 그리고 TGF-β에 의한 이러한 효과는 IN-1130 처치에 의해 억제되었다. IN-1130은 in vivo에서도 유방암 조직의 Snail 발현을 감소시키고 E-cadherin의 발현은 증가시켰다. 이와 함께 IN-1130 처치에 의해 동물 모델의 폐 조직에 나타난 전이성 병소(metastatic colony)의 수가 감소되는 것을 확인하였다. 또한 EMT에 의한 이동성 변화를 in vitro 실험으로 확인해본 결과, IN-1130은 TGF-β에 의해 증가된 세포의 이동성과 Matrigel을 통한 전이를 모두 억제하였다. IN-1130은 in vitro와 in vivo 모두에서 TGF-β에 의해 유도된 Smad2 단백질의 인산화를 억제하였다. 이를 통해 IN-1130이 TβRⅠ/ALK5의 작용을 저해하여 TGF-β signaling을 효과적으로 차단하는 것을 확인하였다. 유방암의 폐 전이 지표로 알려진 inhibitor of differentiation/DNA binding 1 (ID1)과 angiopoietin-like 4 (ANGPTL4)의 조절 기전을 살펴보기 위해 MDA-MB-231 세포로 실험한 결과, 두 지표 유전자의 발현이 Smad 의존적인 TGF-β signaling에 의해 증가되는 것으로 나타났으며, 이를 chromatin immunoprecipitation (ChIP) assay를 통해 각 유전자의 promoter에 Smad2/3가 결합하는 것을 보임으로써 증명하였다.
본 논문의 연구 결과는 IN-1130이 TβRⅠ/ALK5의 소분자 저해제로 유방암의 폐 전이를 효과적으로 억제할 수 있는 항암제로 개발될 수 있는 가능성을 제시하고, 나아가 전이 지표를 활용한 전이성 암의 진단과 치료에 기여할 수 있을 것으로 기대된다.;Epithelial-mesenchymal transition (EMT) is a crucial morphological event that occurs during embryogenesis and carcinogenesis. Through this process, epithelial cells lose their properties, including their polarity and cell-cell contacts, and turn into mesenchymal cells, and then acquire motility. Transforming growth factor-β (TGF-β) signaling has been shown to play an important role in EMT. In cancer cells, TGF-β activates specific transcription factors, especially Snail, a key regulator of EMT, which represses expression of E-cadherin. In many instances, epithelial tumors lose E-cadherin-mediated adhesions as they progress toward malignancy. Tumor cells undergoing Epithelial-mesenchymal transition (EMT) acquire motility, allowing them integrate into surrounding tissue, metastasize through intravasation, survival in circulation, extravasation and outgrowth at a distant organ. Besides the role of TGF-β on EMT, many clinical and experimental evidences revealed the correlation between TGF-β signaling and cancer metastasis. For this reason, investigators have developed many anti-metastatic drugs targeting TGF-β signaling pathway components.
In this study, we investigated effects of TGF-β on EMT and breast cancer metastasis. At the same time, we evaluated the anti-metastatic effects of IN-1130 which was developed as a TGF-β typeⅠreceptor kinase (TβRⅠ/ALK5) inhibitor on metastatic breast cancer cell lines (MDA-MB-231 and MCF-7) in vitro and BALB/c xenograft mouse model in vivo.
First of all, we examined the changes of cellular junction components such as E-cadherin and N-cadherin, and their transcriptional regulator Snail by TGF-β. As a result, TGF-β up-regulated mesenchymal marker N-cadherin and transcription regulator Snail which plays a crucial role as EMT inducer and these were inhibited by IN-1130 concomitant treatment. On the other hand, TGF-β down-regulated expression level of epithelial marker E-cadherin, which was recovered in human breast carcinoma cells by IN-1130 treatment. Therefore TGF-β promoted EMT and these effects were inhibited by IN-1130 in vitro. In addition, IN-1130 reduced the expression of Snail and elevated that of E-cadherin in vivo. Treatment with IN-1130 significantly decreased the number of lung metastatic colonies in vivo too. Also, IN-1130 attenuated TGF-β activated cell motility and invasiveness through the Matrigel in vitro. IN-1130 effectively blocked Smad2 phosphorylation induced by TGF-β in vitro and in vivo. Through these results, we could validate that IN-1130 effectively suppresses TGF-β signaling via inhibiting the action of TβRⅠ/ALK5. We confirmed that inhibitor of differentiation/DNA binding 1 (ID1) and angiopoietin-like 4 (ANGPTL4) known as lung metastasis signatures (LMS) were up-regulated by Smad-dependent TGF-β signaling. Evidence of binding of Smad2/3 to each promoter was obtained by chromatin immunoprecipitation (ChIP) experiments.
Together, our results suggest the possibility that IN-1130 will be a novel small molecular anti-cancer drug that potentially prevents breast cancer metastasis to lung. Furthermore, those also help improve the prognosis and treatment of various metastatic cancers including breast cancer.
