dc.description.abstractA series of 3,4,5-trimethoxycinnamic acid (TMCA) derivatives was prepared and evaluated for antinarcotic effects on morphine dependence in mice and binding affinities on dopaminergic and serotonergic receptors. Our research team found that the naloxone-induced morphine withdrawal syndrome was significantly suppressed by new synthetic TMCA derivatives (20 mg/kg/day). Most of TMCA derivatives were discovered to have high affinity to 5-HT1A receptor. The naloxone-induced morphine withdrawal syndrome was attenuated by 8-Hydroxy-2-(di-n-propylamino) tetralin ((+)8-OH-DPAT, 0.1 mg/kg/day, i.p.), a 5-HT1A receptor agonist. In cortical neuronal cells of mice, (+)8-OH-DPAT (1 μM) produced an elevation of the pERK 1/2 expression whose pERK levels were inhibited by WAY 100635, a 5-HT1A receptor-specific antagonist. In addition, the effect was increased by the TMCA derivatives and the derivatives-mediated changes in pERK levels were constrained by the WAY 100635. These results show that new synthetic TMCA derivatives have a potential anti-narcotic effect through acting as a 5-HT1A receptor agonist in mice and rats. Besides, TMCA and its derivatives, TMCA5, and TMCA9 - were prepared and evaluated for anti-anxiety and anti-depression effects on the condition of immobilization and electric shock stress in mice and rats, respectively. TMCA treated group exhibited significant suppressing anxiety and depressive behavior in EPM and FST. In addition, TMCA5 and TMCA9-treated group showed decreasing anxiety-like behaviors of antianxiety effects. These results suggest that TMCA and its derivatives could be regarded as perspective anxiolytic and antidepressive agents for the treatment of morphine dependence, anxiety, and depression.;본 연구에서는 3,4,5-trimethoxycinnamic acid (TMCA)와 TMCA 유도체들을 합성하여 설치류에서 향정신성 약물인 모르핀 해독효과와 항불안, 항우울 효능의 행동학적 규명 및 그 하위 메커니즘을 탐색하고자 하였다. 먼저, 설치류에서 향정신성 약물에 의존한 항마약 효과와 함께 도파민과 세로토닌 수용체에서의 결합친화력을 평가하기 위해 TMCA 유도체들을 사용하였다. 이 논문에서는 naloxone에 의해 유도된 모르핀 중독 신드롬이 새로이 합성된 TMCA유도체들 (20 mg/kg/day) 에 의해 유의하게 억제됨을 밝혀냈다. 또한 결합친화력 실험에서는 TMCA유도체 대부분이 5-HT1A 수용체와 매우 높은 친화력을 가진다는 결과를 도출해냈다. 이를 기반으로 5-HT1A 수용체의 agonist인 (+)8-OH-DPAT (0.1 mg/kg/day, i.p.) 를 설치류에 주입해 행동학 실험을 한 결과, naloxone에 의해 유도된 모르핀 중독 신드롬이 약화됨을 알 수 있었다. 뿐만 아니라, 마우스 일차 배양 신경세포에 (+)8-OH-DPAT 처리시 pERK1/2의 발현이 증가했으며, 증가된 pERK1/2 발현은 선택적 5-HT1A 수용체 antagonist인 WAY00635에 의해 약화되었다. 또 다른 실험법으로, 마우스와 랫드에서 불안 및 우울증상을 유도하기 위해 고정(immobilization) 및 전기 충격을 주었다. 마우스에서 불안 및 우울증상의 행동학적 측정법인 EPM과 FST를 통해 스트레스 군과 TMCA 주입 군을 비교한 결과, TMCA 주입 군에서 불안 및 우울증세가 완화되는 것을 규명하였다. 또한 랫드에서 (+)8-OH-DPAT을 대조군으로 사용하여 앞서 합성한 TMCA유도체와 유사한 TMCA5, TMCA9 유도체를 경구투여하여 항불안, 항우울 효과를 증명하였다. 이러한 결과들은 TMCA및 TMCA 유도체들이 설치류에서 5-HT1A 수용체 agonist 역할을 함으로써 잠재적인 항마약 효능이 있음과 더불어 불안 완화제 및 우울 관련 작용물질로도 매우 유용하게 사용될 수 있음을 시사한다.-
