Abstract
Aminophosphonic acid 가 생물체에서 발견된 이래로 이에 대한 연구가 널리 행해지고 있다. 본 연구는 아미노산의 일종인 Aspartic acid 유사물질(analog) 2-Amino-3-phosp-honopropionic acid(PAL)가 흰쥐의 체내대상에 미치는 영향을 알아보고자 다음과 같은 과정에 의해서 합성하였다. 2-Amino-3-phosp-honopropionic acid의 합성 1. N-Acetyl-DL-alanine의 합성 2. Methyl-N-acetyl-DL-alaninate의 합성 3. Methyl-N-acetylchloro-DL-alaninate의 합성 4. Methyl-N-acetyldehydroalarinate의 합성 5. 2-Acetamidoacrylic acid의 합성 6. 2-Amino-3-phosphonopropionic acid의 합성 15% casein diet 에 2-Amino-3-phosphonopropionic acid를 0.1%, 0.2%, 0.3%, 수준으로 첨가하여 ?ㅅ Sprague-Dawley 종 28마리를 4군으로 나누어 4주간 사육하였다. 체중변화와 식이섭취량, 식이효율, 단백질 효율을 측정하였고, 혈청내의 inorganic phosphorus와 phospholipid, 간의 무게와 간내에 함유돼 있는 단백질과 지방함량, 지방내의 phospholipid량을 분석하였다. 그 결과를 요약하여 보면 다음과 같다. 1. 실험기간 동안 동물은 죽지않았다. 식이섭취량은 표준군에 비해 실험군이 낮았으며 PAL 첨가수준이 높을수록 식이 섭취량은 현저히 감소하였다. 2. 체중과 체중증가량은 표준군에 비해 실험군이 낮고 PAL 첨가수준이 높을수록 체중과 증가량은 감소하였다. 3. 사료의 효율과 단백질효율은 1, 2주에는 표준군에 비해 실험군이 낮았고 PAL 함량이 높을수록 효율은 낮아지는 경향을 보였으나 3, 4주에는 표준군과 실험군사이에 유의적인 차이가 없었다. 4. 혈청내의 inorganic phosphorus와 phospholipid는 실험군과 표준군 사이에 유의적인 차이가 없었다. 5. 간의 무게는 실험군이 표준군에 비해 상당히 적었으며, 실험군 사이에는 유의적인 차이가 없었다. 간내의 단백질과 지방함량은 실험군과 표준군사이에 유의적인 차이가 없었으나 지방내의 phospholipid는 0.3% PAL 첨가군이 가장 높았고 표준군, 0.2% PAL 첨가군 0.1% PAL 첨가군 순으로 군간에 유의적인 차이가 있었다. 이상에서 보여준 결과는 매우 흥미로우며, 2-Amino-3-phosphonopropionic acid (PAL)의 식이첨가가 맛에 영향을 주는 것이 아닌가 생각되며 간내의 phospholipid에 미치는 영향에 대해선 더 많은 연구가 있어야 할 것으로 생각되어 진다.;Since aminophosphonic acid was discovered in living organism, the biosynthesis and biological function of aminophosphonic acid have been extensively studied. The purpose of this project consists in the preparation of 2-amino-3-phosphonopropicnic acid and the study of biological function of 2-amino-3-phosphonopropionic acid by animal experiments. The following organic synthesis of 2-amino-3-phosphonopropionic acid was studied. 1. Synthesis of N-acetyl DL-alanine 2. Synthesis of Methyl N-acetyl DL-alaninate 3. Synthesis of N-acetylchloro DL-alaninate 4. Synthesis of Methyl N-acetyldehydroalarinate 5. Syntheais of 2-Acetamidoacrylic acid 6. Synthesis of 2-amino3-phosphonopropionic acid 2-amino-3-phosphonopropionic acid was supplemented to the 15% casein diet at the level of 0.1%, 0.2%, 0.3%, then the animals were divided into 4 groups, 7 rats each, and fed for 4 weeks. Body weight change, ad libitum fed food consumption were measured, and F.E.R., P.E.R. were calculated. Inorganic phosphorus and phospholipid in serum were measured, and weights of liver, content of totallipid, hospholipid, protein were also measured. The results were summarized as follows; 1. The experimental animal were not died during experimental period. The food intake of PAL-added group were less than the standard group and the food intake was decreased as the level of PAL was increased. 2. Body weight change of PAL-added group were less than the standard group and body weight change was decreased as the level of PAL was increased. 3. F.E.R. and P.E.R. of PAL-added group were less than the standard group, and P.E.R. and F.E.R. were decreased as the level of PAL was increased. during 1st end 2nd weeks. There was no difference in P.E.R. and F.E.R. between standard and PAL-added group during 3d, 4th weeks. 4. Inorganic phosphorus and phospholipid in serum were not significant in standard group and experimental group. 5. Weights of liver of PAL-added group was less than the standard group. There was not significantly different in protein and lipid content of the liver. However, phospholipid in total lipid content was significantly different. Phospholipid content was significantly higher in 0.3% PAL added group than standard group, phospholipid content in 0.1% PAL added group, 0.2% PAL added group was less than standard group.
