Abstract
During early development of Xenopus laevis, embryonic cells can change their fate to mesoderm in response to mesoderm-inducing signals such as Activin and Nodal. However, their responding ability is lost for a certain period of time shortly after gastrulation. This process is termed the loss of mesodermal competence (LMC). To understand its mechanisms, I analyzed the transduction of Nodal signaling at LMC. The onset of LMC correlates with the inability of Smad2, the principal intracellular Nodal signal transducer, to accumulate in the nucleus. However Smad2 can be phosphorylated at its C-terminal serines by Nodal stimulation even during the period of LMC. This temporal inability of Smad2 nuclear translocation is correlated with phosphorylation in the linker region of Smad2. Previously Ca2+-calmodulin-dependent protein kinase II (CamkII) has been shown to inhibit Smad2 signaling. To understand the correlation between the Smad2 linker phosphorylation and the CamKII activation during LMC, dissociated animal cap cells were treated with Activin and CamKII inhibitors. However, these artificial processes result variable loss of cells and inconsistent results. Therefore, I devised a simpler and more reliable method for investigating Smad pathway and characterizing the mechanism of LMC.;Xenopus laevis의 초기 발생 과정 동안, 배아 세포는 낭배기가 끝난 후 어던 특정 시기에 Activin과 Nodal 과 같은 중배엽 유도 신호에 반응하는 능력을 잃게 되는데 이 과정을 loss of mesodermal competence (LMC) 라고 한다. LMC 작용을 이해하기 위해 LMC에서 Nodal의 신호 전달에 대해 연구하였다. LMC의 시작은 Nodal 신호의 전달물질인 Smad2가 핵 안에 축적되는 능력을 상실하는 것과 관련되어 있다. 그러나 Smad2는 LMC과정 중에도 Nodal 신호에 의해 C-terminal이 인산화 될 수 있다. 이러한 Smad2의 핵 안에 축적되는 능력에 대한 일시적인 제한은 Smad2 linker region의 인산화와 관련되어 있다고 알려져 있다. 이러한 신호전달과정에 대해 연구하기 위해서는 세포를 흩었다가 관찰하는 방법이 사용되어 왔는데 이 방법은 세포에 스트레스를 주고 불안정하게 만든다. 이 논문에서는 세포가 안정화된 상태로 좀 더 쉽게 연구하기 위한 실험 기법과 이 실험 기법을 통해 Smad2의 인산화에 Camkinase가 미치는 영향에 대해 연구하였다.
