dc.description.abstractFor the first time, an ammonolysis?based technique utilizing ethyl fluoroacetate as a dispersed solvent was used to encapsulate a model drug, progesterone, into poly-(d,l-lactide-co-glycolide) microspheres. The objective of this study was to evaluate the new microencapsulation technique in terms of microsphere morphology, encapsulation efficiency, thermal behavior, the residual ethyl fluoroacetate and the physical status of progesterone. First of all, an HPLC analytical methodology was developed, validated and applied for the determination of the amount of progesterone encapsulated in the microspheres. It was found that sample preparation, HPLC chromatographic conditions and peak analysis methods affected the assay accuracy of progesterone contents in the microspheres. As far as process parameters were concerned, the ammonia to ethyl fluoroacetate molar ratio and progesterone payload decisively influenced the characteristics of the microspheres. The median diameter yielded from our microencapsulation process was 209 ㎛. The encapsulation efficiencies of microsphere prepared using 3 mL of ammonia solution were between 57.49 and 93.72 %. On the contrary, encapsulation efficiencies of microspheres, when 9 mL of ammonia solution was used, ranged from 93.63 to 100.07 %. With increasing progesterone payloads, more crystal progesterone appeared inside the microspheres. Also, the residual ethyl fluoroacetate in the microsphere was decreased as a function of progesterone payload. All our results demonstrated that the rapid completion of ammonolysis led to the conversion of the water-immiscible ethyl fluoroacetate into water-soluble solvents, thereby providing an efficient tool for microsphere solidification. In conclusion, an ammonolysis-based microencapsulation process would contribute to encapsulation of various bioactive materials into PLGA microspheres.;Poly-(d,l-lactide-co-glycolide) (PLGA)를 이용하여 제조한 microsphere는 생분해성 및 생체적합성을 모두 갖추고 있어 널리 이용되고 있다. 이 실험에는 신속한 microsphere solidification를 위해 암모니아 분해 반응을 이용하여 유기 용매를 물에 섞이는 산물로 분해하는 새로운 microencapsulation 방법을 사용하였다. 이 실험에서는 암모니아 분해 반응이 용이한 ethyl fluoroacetate를 분산 용매로 사용하였으며, microsphere에 봉입할 시험 약물로는 프로게스테론을 사용하였다. 프로게스테론 봉입률을 분석하기 위하여 메탄올과 물을 혼합한 이동상을 이용하는 HPLC로 분석하였다. 이 과정에서 microsphere 시료를 녹이는데 사용한 tetrahydrofuran이 이동상과의 solvent strength차이로 피크 형태 변형을 일으키는 것을 확인하였다. 이 때문에 피크 면적과 피크 높이를 사용하였을 때 각각 다른 분석 수치가 나오는 결과를 얻었다. 본 실험에서는 이 현상을 규명하고 progesterone 봉입률 분석법을 확립하였다. Microsphere 제조 시에 사용한 프로게스테론의 양, 유기 용매에 대한 암모니아 용액의 몰 비가 프로게스테론 봉입률, 형태, 입자 크기, 프로게스테론의 물리적 상태, thermal behavior, 잔류 용매 함량과 같은 microsphere의 특성에 영향을 미쳤다. 입자 크기는 평균 209 nm였으며, 봉입률은 암모니아 용액 3 mL을 사용하여 제조한 경우 57.49 ~ 93.72 %였고 암모니아 용액 9 mL을 사용하여 제조한 microsphere를 분석한 경우에는 93.63 ~ 100.07 %로 나타났다. 프로게스테론 사용량이 늘어날수록 결정형으로 존재하는 프로게스테론이 많아지는 것을 확인하였다. 약물이 양이 많아질수록 잔류하고 있는 유기용매의 함량은 줄어드는 것을 확인하였다. 본 실험의 암모니아 분해를 이용한 microsphere 제조는 신속하고 효율적인 microencapsulation 방법이 될 것으로 기대된다.-
