- 종양괴사인자 (tumor necrosis factor-a, TNF-a)에 의해 유도되는 NF-kB의 활성화는 선천성 면역 반응 혹은 염증 반응에서 중요한 역할을 한다. NF-kB의 신호전달 과정에서 핵심이 되는 IKK complex는 700-900 kDa의 단백질 복합체로서 core protein은 IKK α, β, γ의 3 가지 subunit으로 구성되어 있다. 따라서 IKK complex의 구성 단백질을 찾는 것은 NF-kB 신호전달을 이해하는 데 있어서 중요하다. 앞선 논문에서 immune-affinity proteomic 방법으로 IKK complex의 또 다른 구성 단백질인 zinc finger protein 268 (이하 ZNF268)를 새로이 찾아 보고한 바 있다.
ZNF268은 alternative splicing에 의해 N-말단은 kru‥ppel-associated box (KRAB) domain이 없고, C-말단은 2개의 cysteine과 2개의 histidine이 24번 반복되는 구조를 갖는 단백질이다. 본 논문에서는 zS4 (spacer/first 4 C₂H₂ repeats)의 발현만으로도 NF-kB의 transcriptional activity가 증가함을 보였다. 흥미롭게도 TNF-a 자극을 주었을 때 NF-kB의 transcriptional activity의 증가가 더욱 증폭되었다. 이는 ZNF268이 NF-kB 신호전달 과정에 관여함을 시사한다. ZNF268의 24개의 zinc finger motifs 중 각각 NF-kB의 transcriptional activity와 IKK activity에 관련 있는 부분을 찾기 위해 ZNF268을 C₂H₂가 4번 반복하도록 8개의 fragment로 나눠서 cloning하였다. TNF-a 자극에 따라 8번 째 4-C₂H₂ fragment에 의해 NF-kB의 transcriptional activity가 현저하게 증가함을 확인하였다. 또한 8개의 4-C₂H₂ fragment 중 C₂H₂ #2, C₂H₂ #3이 IKK activity를 증가시킴을 보았다.
종합적으로 이러한 결과들은 IKK complex의 구성 단백질로 찾은 ZNF268이 TNF-α 자극에 의한 NF-kB의 신호전달 과정에서 중요한 역할을 담당하고 있음을 제시한다.;TNF-α-induced NF-kB activation plays a key role in innate immunity and inflammation. The IKK complex, consisting of two catalytic subunits, IKKα and IKKβ, and a regulatory subunit NEMO, plays major roles in signal-dependent activation of NF-kB. In an effort to identify additional components of IKK complex, we previously isolated the splice variant of ZNF268 as a novel IKK-interacting protein. ZNF268 is a protein of an exceptionally long repeated domain containing 24 C₂H₂ zinc finger motifs. Here, I show that only expression of zS4 (spacer/first 4 C₂H₂ repeats) is sufficient to increase the NF-kB transcriptional activity. A series of truncated mutants containing only 4 C₂H₂ repeats were generated. Upon stimulation by TNF-α, C₂H₂ #8 (amino acids 838 to 942) significantly enhances the transcriptional activity of NF-kB. Furthermore, the results of in vitro kinase assays revealed that the truncated mutants modestly increased the IKK activation. Particularly, the N-terminus of ZNF268s mainly increases IKK activity.
Collectively, these results suggest that the splice variant form of ZNF268 plays a positive role in TNF-α-induced NF-kB signaling pathway.
