Abstract
Human Fas-associated factor, hFAF1, was identified as Fas-associating molecule and a member of the apoptosis signaling complex. We identified that hFAF1 acts as a scaffolding protein by unveiling the interacting proteins and ubiquitin related domains. hFAF1 has multi-ubiquitin related domains including ubiquitin associated (UBA) domain, ubiquitin like 1, ubiquitin like 2 (UBL1, UBL2) and ubiquitin regulatory X (UBX) domain. Our laboratory found that UBX domain interacts with valosin containing protein (VCP), the polyubiquitinating targeting factor in the ubiquitin-proteasome pathway. N-terminal UBA domain was identified to recruit K48- and K63-linked poly-ubiquitinated proteins and plays a role to accumulate the ubiquitinating proteins. In our previous study, UBA domain is suggested a key modulator can regulate apoptosis. In this study, hFAF1 is investigated as an ubiquitin receptor focusing on the importance of UBA domain in ubiquitin-proteasome system. To understand the molecular regulation of UBA domain, at first I made efforts to identify polyubiquitinated UBA substrates to understand molecular functions of hFAF1. Secondly, in order to investigate the biological function of interaction of hFAF1 and VCP, I designed and produced VCP binding negative mutant of hFAF1 based on the information of post-translational modification following heat shock and NMR structure of UBX domain. Surprisingly, the interaction of VCP and UBX of hFAF1 affects the recruitment of polyubiquitinated proteins on UBA domain. This result suggests that VCP binding stabilizes interaction between UBA domain and its substrates. In the cases of other ubiquitin like domains, UBLs are known to interact with intra-and inter- UBA domains, we examined whether UBL domains interact with adjacent UBA domain. Overexpressed UBL1-UBL2 domains inhibit substrates binding to UBA domain but is not significant compare to impairment of VCP binding. We suggest that hFAF1 has fine regulation of UBA domain activity by using other ubiquitin related domain, UBLs and UBX domain via VCP binding. Recently, hFAF1 was also reported as a suppressor of NF-кB activity by cytoplasmic retention of NF-кB p65 via physical interaction and IKK complex inhibition. To investigate the role as a ubiquitin receptor in NF-кB activation pathway, I examined the UBA substrates recruitment with hFAF1 wild type and VCP binding negative mutant of hFAF1. The binding of substrates is increased in hFAF1 wild type by TNFΑ treatment and even in VCP binding negative mutant. This result suggests that hFAF1 acts as an ubiquitin receptor in TNFΑ signaling pathway and UBA domain has specific binding potential to recruit its substrates regardless the VCP binding. We examined whether hFAF1 plays important role in human cancer because hFAF1 accumulates polyubiquitinated proteins as proteasome inhibitors which are developed for anti-cancer drug. The effects of overexpression of various truncated hFAF1 and knock down in hFAF1 were assessed by colony formation and cell survival in HeLa cells. Using truncated hFAF1 study, we identified UBA domain playing major role in hFAF1 induced apoptosis in cancer cells. The overexpression hFAF1 and UBA domain increased apoptosis and inhibited colony formation and knock down vice versa. This was also confirmed in clinical samples of cervical cancer tissues. The expression levels of hFAF1 in cancer tissues are significantly decreased compared to normal tissues. These results suggest that hFAF1 is a novel candidate tumor suppressor by regulating ubiquitin proteasome pathway.;Human Fas-associated factor 1 (hFAF1) 은 Fas에 결합하는 단백질로 처음 알려졌으나 우리는 hFAF1의 결합 단백질을 찾음으로써 hFAF1을 ubiquitin proteasome system에 관여하는 scaffolding 단백질로 밝혔다. hFAF1은 ubiquitin associated (UBA) domain, ubiuquitin like 1, ubiquitin like 2 (UBL1, UBL2) domain, ubiquitin regulatory X (UBX) domain 등 ubiquitin과 구조가 유사하거나 ubiquitin과 결합하는 domain을 가지고 있다. 우리는 hFAF1이 UBX domain을 통해 valosin containing protein (VCP)와 결합하고 N-terminal의 UBA domain은 K48- K63- linkage의 polyubiquitination된 단백질과 결합한다. UBA domain은 세포내의 polyubiquitination된 단백질을 축적 하여 세포 사멸을 일으킨다. hFAF1의 UBA domain의 분자 수준의 조절을 이해하기 위해 우리는 UBA substrate을 찾는 시도를 하였다. 두 번째로 hFAF1과 VCP의 기능을 알아보기 위해 VCP와 결합하지 않는 hFAF1 mutant를 제작하였다. PTM과 UBX 의 구조를 기반으로 만들어진 이 mutant에서 놀랍게도 VCP결합하지 않는 hFAF1은 UBA domain의 subatrate와 결합하지 못하였다. 이 결과는 VCP의 binding이 UBA domain의 substrate binding을 안정화 시키고 있음을 의미하며 hFAF1의 ubiquitin관련 domain들 간의 조절이 있음을 의미한다. 또한 우리는 다른 ubiquitin 관련 domain인 UBL domain이 UBA domain에 어떤 역할을 하는 지 알아보는 실험을 하였다. 많은 UBL-UBA domain 단백질에서 UBA와 UBL domain의 상호작용이 알려져 있는데 hFAF1에서도 역시 UBL domain이 UBA domain과 작용 하여 UBA domain을 저해하는 것으로 나타났다. 최근 hFAF1은 NFκB, IKKβ와 결합하여 NFκB 활성을 저해한다고 보고되었다. NFκB의 활성화 과정에서 hFAF1의 ubiquitin receptor로서의 기능을 살펴보기 위해 TNF? 처리 후 wild type hFAF1과 VCP와 결합하지 못하는 hFAF1에서 UBA substrate결합을 실험하였다. TNF처리에 의해 UBA substrate 의 결합이 증가하였고 이는 hFAF1이 TNF신호전달 과정에서 UBA substrate 특이성을 가지고 ubiquitin receptor로서 작용하고 있음을 의미한다. 또한 VCP와 binding 하지 못하는 hFAF1에서 VCP 결합과 관련 없이 polyubiquitin 결합이 증가하는 것 또한 hFAF1이 UBA substrate 특이성이 있음을 의미한다. 우리는 hFAF1이 암세포에서 중요한 기능을 하는지 알아보기 위해 임상의 자궁 경부암 조직에서 hFAF1의 발현을 조사하였다. hFAF1은 정상세포에 비해 암세포에서 발현이 현저하게 낮았다. 우리는 hFAF1 knock down된 HeLa S3 세포에서 colony formation이 감소한 것을 보임으로서 in vitro에서 재현 하였고 이것이 UBA domain에 의해 세포사멸이 진행되는 것과 어떤 관계가 있는지 알아보기 위해 여러 형태의 hFAF1 발현시킨 HeLa 세포에서 colony formation을 관찰한 결과 암의 형성에 UBA domain이 중요하다는 것을 증명하였다. 이것으로 우리는 hFAF1을 새로운 tumor suppressor로서 제안하였으며 이 기능이 ubiquitin proteasome system을 조절 하여 일어난다는 것을 증명하였다.
