dc.description.abstractSeveral studies have shown that vascular smooth muscle cells (SMCs) express the cellular adhesion molecules in atherosclerosis and restonsis. Although the biological consequences of adhesion of monocytes and leukocytes to SMCs are not well characterized, the expression of adhesion molecules was most prominent in fibrous plaques and advanced atherosclerotic lesions. It has recently been shown that peroxiredoxinII (PrxII) suppresses the proliferation and migration of SMCs and, furthermore, is involved in the neointimal thickening of SMCs in balloon-injured carotid artery. But there has not been investigated for the effects of PrxII in monocyte adhesion, which is important process of atherosclerosis. Here, this study show that PrxII suppresses the tumor necrosis factor-α (TNF-α) induced monocyte adhesion to smooth muscle cells and expression of adhesion molecules, such as intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1). In order to expand our interest to look for additional factors that regulate monocyte adhesion to SMCs, I performed a large-scale adhesion assays using 43 protein candiates, which has previsouly been identified in balloon-injured animal model. Among the candidates, I found that intimal thickness-related receptor and PKA alpha catalytic subunit significantly influenced monocyte adhesion to SMCs when they were knock-downed in SMCs.;내피세포에서 발현되어 동맥경화와 혈관 재협착 및 혈관이식과 관련된 것으로 알려져 있는 adhesion molecule이 최근에 내피세포뿐 아니라 혈관의 평활근 세포에서도 발현된다는 것이 보고 되어지고 있다. 아직 평활근 세포와 단핵세포 및 백혈구의 adhesion에 관한 생물학적인 메커니즘은 밝혀지지 않았지만, 평활근 세포에서의 adhesion molecule이 plaque와 동맥경화 장애를 촉진시키는 역할을 한다는 것은 분명하다. 또한 최근 발표된 논문에 의하면 PeroxiredoxinII (PrxII) 라는 antioxidant protein은 platelet-derived growth factor (PDGF) 수용체의 위치 특이적인 인산화를 통해 평활근 세포의 증식과 이동을 억제하고 나아가 balloon-injury를 받은 경동맥의 neointima에 평활근 세포가 축적되는 것을 억제 시키는 것으로 밝혀져 있다. 그러나 아직 평활근 세포에 있어서 PrxII가 혈관장애에 있어서 중요한 adhesion에 끼치는 영향에 대한 연구는 없었다. 이에 이번 연구를 통해 PrxII가 혈관의 평활근 세포에서 intercellular adhesion molecule-1 (ICAM-1) 과 vascular cell adhesion molecule-1 (VCAM-1)의 발현을 억제함으로 tumor necrosis factor-α (TNF-α)에 의한 adhesion을 억제하고 있음을 in vitro 실험을 통해서 살펴보았다. 또한 혈관 재협착과 관련된 biomarker 및 target단백질을 찾는 실험을 위해 proteomics 분석법을 이용하여 balloon-injured 동물 모델에서 43개의 후보 단백질들을 찾았다. Intimal thickness-related receptor (ITR) 와 PKA alpha catalytic subunit (PRKACA) 는 후보 단백질에 포함되어 있는 단백질들로서, 이미 혈관장애와 관련이 있음이 보고된 바 있다. In vitro 에서 TNF-α에 의한 adhesion assay를 진행한 결과 ITR 은 감소하였고, PRKACA는 증가하는 양상을 보였다. 이 결과는 ITR이 biomarker로의 역할을, PRKACA이 target 단백질로서의 역할을 할 수 있음을 보여주는 의미있는 결과이다.-
