Abstract
우리나라산 가물치의 내장중에 존재하는 Ornithine aminotransferase의 효소활성을 연구한 결과, 다음과 같았다. 각 내장을 cell fractionation하여 효소활성을 측정하였더니, Ornithine aminotransferase는 cytosol fraction에 70%∼84%, mitochondria fraction에 14∼22% 정도 들어있었다. 효소활성의 Optimum PH는 7.3이었으며, Michaelis 상수는 L-Ornithine에 대하여 0.432mM, α-Ketoglutaric acid에 대하여 0.238mM로 나타났다. Ornithine aminotransferase는 중금속 등에 의하여 효소활성이 방해되었다. 그 중, 특히 HgCl_(2)가 가장 큰 방해작용을 나타내었다. 내장 각 부위의 Ornithine aminotransferase의 활성크기를 비교해 보면, 간>위>신장>창자>심장>정소>비장>난소의 순서로 나타났다.;This report described the enzyme assay, partial purification, and general properties of ornithine aminotransferase in internal organs of Korean Ophicephalus argus. 1. The result which was accomplished cell fractionation for each internal organ, was that its' cytosol fraction had contained ornithine aminotransferase about 70%-84%, and its' mitochondria fraction had contained ornithine aminotransferase about 14%-22%. 2. The PH optimum of ornithine aminotransferase was found to be approx. 7.3. Its' Michaelis values (Km) found were: L-Ornithine, 0.435mM; -Ketoglutaric acid, 0.238mM. And the partially purified liver enzyme was inhibited by heavy metals; HgCl_(2), KCN, AgNO_(3), CdCl_(2), CuCl_(2), Specially, the greatest inhibition was observed with Mercury. 3. The relative activity of ornithine aminotransferase in internal organs was found to be ; Liver>Stomach>Kidney> Intestine>Heart>Testes>Spleen>Ovaries.
