dc.description.abstractNerve growth factor(NGF)는 일부 말초 및 중추 신경세포의 분화와 생존을 증진시킨다고 알려져 있으므로, 신경세포의 사멸로 인한 질병의 치료제로 사용될 수 있다고 기대되고 있다. 그러나, 이 실험은 NGF가 포도당 결핍 상황에 있는 쥐의 부신 신경 암세포(rat adrenal pheochromocytoma; PC12) 의 사멸을 촉진시킨다는 상반되는 결과를 보여 주었다. NGF에 의해 촉진된 세포 사멸은 외부 calcium농도의 영향을 받고 있고, L-type calcium channel 억제제인 nifedine에 의해 감소되었고 또한, 세포 내 저장소로부터 방출되는 calcium을 조절하는 ryanodine에 의해서도 억제되었다. 이 결과들은 세포 내 calcium농도의 변화가 NGF에 의한 세포 사멸 촉진의 중요한 요인이라는 것을 간접적으로 보여주고 있다. 다음 단계로 NGF에 의해 촉진되는 세포 사멸이 대표적인 사멸 형태로 알려진 necrosis와 apoptosis 중 어느 과정을 통하여 일어나는지 알아보았다. PC12 세포가 포도당이 결핍된 상황에서 NGF에 의해 형태적으로는 수축하면서 죽기 때문에 apoptosis라고 생각되었지만, DNA의 붕괴가 서서히 일어나고, comatin condensation이 일어나지 않는 점들로 보아 apoptosis 보다는 necrosis과정을 거치는 것으로 보인다. 따라서, 포도당 결핍 시 NGF에 의해 촉진된 PC12세포의 사멸은 내부 calcium수준의 증가를 매개로 하는 apoptotic-like necrosis 과정을 통하여 일어 난다고 보여진다.;Nerve growth factor (NGF) is known to promote differentiation and survival in several peripheral and central neurons. Thus, it is widely believed that NGF may serve as a therapeutic agent for many types of neuronal diseases. However, NGF promoted cell death in PC12 cultures that was induced by glucose withdrawal. Such an effect of NGF was dependent on extracellular calcium concentration and was blocked by nifedipine, an L-type calcium channel blocker. Also, ryanodine, a modulator for releasing calcium from intracellular stores, could block the effect of NGF on cell death. All these results suggest that intracellular calcium may be a critical factor in causing the NGF-promoted cell death. To determine which type of dying processes, necrosis of apoptosis, was responsible for the NGF-promoted cell death, several parameters related to cell death were examined. Morphologically, PC12 cells were shrunken when exposed to NGF in the absence of glucose, indicating that apoptosis may occur in the NGF-promoted cell death. However, delayed DNA degradation and the lack of chromatin condensation in PC12 cells subject to NGF-promoted cell death argued against the idea that the NGF-promoted cell death was apoptotic. Therefore, I conclude here that the NGF-promoted cell death in PC12 cell cultures deprived of glucose in an apoptotic-like necrosis and mediated by the increase of intracellular calcium.-
