dc.description.abstract1. Chitin synthesis is crucial for Candida morphogenesis. Therefor it is important to study the functional characterize chitin synthase inhibitors, which can be used as antifungal agents. Nikkomycin Z is a competitive inhibitor chitin synthases in fungi. It has been reported that it inhibits chitin synthases 1 and 3 but not 2 of Saccharomyces cerevisiae. In this study, nikkomycin Z inhibited all three chitin synthase isozymes of Candida albicans. IC_(50) values for CaChsl, CaChs2, CaChs3 were 15 μM, 0.8 μM and 13 μM, respectively. Nikkomycin Z also inhibited vegetative growth of C. albicans in a medium-dependent manner: growth inhibition was observed in Spider and corn meal agar plate, but not on Lee's plate. Growth inhibition by nikkomycin Z resulted from lack of septum and cell wall chitin, which in hun brought about cell lysis. Cell lysis was prevented to some extent by adding an osmostabilizer to the medium. Taken together, nikkomycin Z induced cell lysis by inhibition of the formation of septum and cell wall chitin in a growth medium-dependent manner. 2. Microtubule-dependent movement is fundamental to many biological processes including cell division and intracellulalar movement of membranous organelles in eukaryotic cells. The kinesin superfamily protein(KIF) is motor for the movement. In contrast to Saccaromyces cerevisiue, little is known about kinesin-like protein(KLP) in Candida albicans. Kinesin is composed of 4 domains with ATPase activity region: mortor, neck, stalk and tail. Similarity search with that region of murine ubiquitous kinesin heavy chain as a query revealed 6 contigs containing putative KLPs in the genome of C. albicans. The length of an open reading (ORF) of 375 amino acids, designated CaKar3, was short compared with S. cerevisiae KAR3 of 729 amino acids. CaKar3 was identified by using RT-PCR and lambda genomic library screening. CaKAR3 is a C-terminal motor protein of 687 amino acids encoded by non-disrupting gene. The amino terminal nonmotor region exhibited some divergence among fungal C-terminus kinesin including ScKAR3.;1. Chitin 합성은 병원성 곰팡이인 Candida albicans의 형태 전환에 중요한 요인이므로, chitin 합성을 저해하는 저해제의 작용 조건을 연구하는 것은 중요한 일이다. Nikkomycin Z는 chitin synthase에 대한 경쟁적 저해자(competitive inhibitor)로서, Saccaromyces cerevisiae의 세 chitin 합성효소 중 Chs1과 Chs3를 선택적으로 저해하는 것으로 알려졌다. 본 연구에서 nikkomycin Z는 Cancida albicans의 Chitin 합성효소 모두를 저해한다고 밝혔다. IC50 값은 CaChsl에 대해서는 15μM, CaChs2는 0.8μM, Cachs3는 13μM인 것으로 조사되었고, nikkomycin Z는 in vivo 상에서도 배지조건에 따라 C. albicans의 생장과 균사체 형태전환을 다르게 저해하는 양상을 보였다. 균사체 형성을 유도하는 Spider media, Cornmeal agar, Lee's media에서 각각 nikkomycin Z의 저해효과를 조사한 결과, Lee's media 에서는 C. albican의 생장을 거의 저해하지 못하였다. Spider 배지와 cornmeal agar 배지에서 nikkomycin Z에 의해 생장이 저해된 균주들은 septum과 cell wall chitin의 형성이 저해된 것을 확인할 수 있었다. 그러므로 세포내에서 nikkomycin Z에 의한 chitin 합성 효소의 저해와 그로 인한 세포생장 저해와 용혈현상은 배지조성에 의한 영향을 받는다고 볼 수 있다. 2. Saccaromyces cerevesiae와는 반대로 Candida albicans의 Kinesin-like protein (KLP)에 대해서는 잘 알려지지 않았다 yeast에서 human까지 KLP나 kinesin의 motor domain이 보존되는 subregion을 포함하고 있다. C. albicans의 genome안에 KLPs라고 추정되는 contigs 6개와 murine에 있는 kinesin heavy chain과 유사상 조사를 했다. C. albicans에서 KLP라고 추정되는 것들은 S. cerevisiae의 KAR3와 비교했을 때 ORF의 길이가 729 amino acids 인 Sckar3에 비해 375 amino acids로 짧았으나 밀접하게 관련되었다. CaKAR3의 sequence를 완성하기 위한 연구가 진행되었고 λ genomic library와 RT-PCR을 이용하여 CaKAR3을 분리하였다. CaKAR3은 674 amino acids의 C-terminal motor protein으로 밝혀졌다. CaKAR3의 N-terminal non-motor region에서 Sckar3를 포함한 곰팡이들의 C-Terminal kinesin과 약간의 차이를 볼 수 있었다.-
