Abstract
미생물이 생산하는 고분자물질은 다양하고 독특한 성질을 지니고 있기 때문에 각종 산업에서 신소재로서 잠재력이 크며 미생물을 이용하여 발효에 의해 대량생산할 수 있으므로 유망한 산업화의 가능성을 가지고 있다. 즉 수용액의 성질을 변화시키거나 필름이나 막을 생성시키는 주요한 특징 외에도 생적합성, 생분해성, 친수성, 비독성 등의 특성으로 인하여 식품산업, 제약업, 의약분야에서 앞으로 널리 이용될 것으로 기대된다. 본 연구는 미생물이 생산하는 고분자물질의 하나인 Poly(r-glutamic aid)(PGA)를 생성하는 균을 분리하여 동정하고, 이 물질의 대량생산을 위한 최적배양조건과 그 물성에 대해 알아보고자 하였다. 청국장을 열처리한 후 분리된 균 중에서 PGA 생성이 가장 많은 균주를 선별하여 동정해 본 결과 Bacillus 속으로 보이고, 여러 면에서 Bacillus licheniformis와 유사하나 크기나 혐기적 조건에서 자라지 않는다는 점에서 차이를 보였다. PGA를 생성하기 위한 최적의 배양조건으로 초기 pH 6.5, 배양온토 37℃에서 초기접종량이 파장 660㎚에서 흡광도가 0.05되게 접종한 후 72시간 배양할 때 가장 많은 PGA를 생성하였고, 이때 배지성분으로 Glucose 3g, Ammonium sulfate 0.5g, L-glutamic acid 5g과 다른 여러 무기염과 함께 biotin 50μg을 100㎖에 첨가하였다. 배양액에서 균체를 제거한 후 Methanol에 침전시켜 Freeze dryer를 통해 powder상태의 PGA를 얻어 그 물성을 조사하였다. UV absorption spectrum 결과 230㎚이하에서 흡광도가 증가하는 전형적인 polypeptide양상을 보였고, PGA를 가수분해시켜 TLC로 확인해 봄으로써 PGA는 glutamic acid가 γlinked glutamic acid로만 이루어져 있음을 알 수 있었다. SEM, HPLC를 통해 PGA는 길이가 다른 peptide가 매우 broad하게 분포되어 있음을 알 수 있다. PGA의 용해도를 보면 H_(2)O, HCOOH, HCl, NaOH에는 잘 녹으나 각종 alcohol류에는 녹지 않는다. PGA는 수용액 상태에서 저절로 가수분해되고 extracellular degradation enzyme에 의해서도 분해되는데 enzyme에 의한 분해가 가수분해보다 더 빨리 진행된다. PGA의 relative viscosity(η_(rel))는 72시간 배양 후 분리된 PGA가 가장 높고, pH 6-8에서 안정하며 염기보다는 산성쪽에서 η_(rel)가 더 낮았다. salt의 첨가(0.1-1%)와 높은 온도(40-80℃)는 η_(rel)를 감소시키는 결과를 가져왔다.;The purpose of this work was to isolate biodegradable PGA producing bacterium, optimize culture condition for PGA production and characterize properties of PGA. A bacterial strain producing highly viscous PGA was isolated from Chung guk chang and supposed to be Bacillus sp. based on morphological, biochemical and physiological characteristics. The optimal culture conditions of Bacillus sp. for PGA production were initial pH 6.5, inoculum size 0.05 at O.D 660nm, and 72hr culture with the medium including glucose 3g, ammonium sulfate 0.5g, L-glutamic acid 5g and other minerals. Under this condition, the amount of PGA production was 1.04g/100㎖. PGA was easily degraded by hydrolysis and extracellular degradation enzyme and degradation by enzyme was faster than hydrolysis The results of UV absorption spectrum, TLC, HPLC and SEM indicated that PGA was composed of only glutamic acid through γ-linked polypeptide. The relative viscosity of PGA was very high when PGA was isolated from culture broth after 72hr cultivation, and was decreased under acid or alkali condition, high temperature and salt addition.
