Abstract
전자 주게로서 β-naphthol과 전자 받게로서 viologen이 유연성 polymethylene 사슬로 연결된 분자의 수용액 중에서의 평형과 이들 분자와 α-CD와의 내포 착화합물 형성에 관한 연구를 핵 자기 공명 분광학적 방법으로 수행하였다. Naphthol과 viologen이 알킬 사슬로 연결된 분자의 chemical shift는 naphthol과 viologen이 분리되어 있는 분자와 비교할 때 현저한 차이를 보였다. 이는 용액 중에서 고려하는 분자가 분자내 또는 분자간 착물을 형성하여 naphthol과 viologen 부분이 서로 평행하게 존재할 경우에 생기는 ring current 효과로 설명할 수 있다. β-Nap-O-C_(n)-MV^(2+)·2Cl^(-)분자의 chemical shift는 농도에 따라 크게 변화하며 이러한 농도 의존성으로부터 β-Nap-O-C_(3)-MV^(2+)·2Cl^(-)분자의 분자내, 분자간 전하 이동 착물 형성 상수를 구하고자 하였다. 그러나 실험 결과와 예상되는 결과와는 상이한 경향을 보였으며 이는 고려하는 분자가 이온의 성질을 띄기 때문으로 해석되었다. 시료의 농도에 따른 이온 세기의 변화를 줄이기 위하여 과량의 (0.3 M NaCl)의 염을 포함하는 용액에서 β-Nap-O-C_(3)-MV^(2+)·2Cl^(-) 분자의 Chemical Shift의 농도 의존성 실험을 반복함으로써 분자내, 분자간 전하 이동 착물 형성 상수 값을 구하였다. β-Nap-O-C_(9)-MV^(2+)·2Cl^(-), β-Nap-O-C_(12)-MV^(2+)·2Cl^(-)와 같이 알킬 사슬의 길이가 긴 경우에는 경우 농도에 따른 chemical shift의 기울기가 일정 농도에서 급격히 변하는 현상이 보여지는데 이러한 현상은 소수성 분자의 회합현상으로 설명될 수 있다. 관찰된 chemical shift를 분자의 총 농도의 역수에 대하여 도시함으로써 회합이 일어나기 시작하는 농도를 구하였으며 그 값은 두 경우에 각각 1.7mM, 10mM이었다. 분자내 전하 이동 착물의 구조에 대한 정보를 얻기 위하여 분자간의 상호작용이 없는 충분히 묽은 농도에서 chemical shift를 관찰하였으며 또한 컴퓨터를 이용한 분자 모델링을 병행하였다. 에너지 최소화 방법으로 최적화 시킨 분자내 전하 이동 착물의 구조는 naphthol과 viologen 부분이 서로 평행하게 위치하며, 이는 관찰되는 chemical shift로부터 유추되는 구조와 잘 부합된다. β-Nap-O-C_(n)-MV^(2+)·2Cl^(-)용액에 α-CD를 첨가하면 내포 착화합물이 형성되며, 이러한 착물 형성 반응의 속도에 따라 NMR 스펙트럼은 많은 영향을 받는다. 사슬이 짧을 경우(n=3, 5)에는 착물을 이룬 상태와 이루지 않은 상태와의 교환이 NMR 시간 척도에 비해 빠르므로 시간 평균값만을 관찰할 수 있으며, 이 경우 NMR 적정의 방법으로 α-CD와의 내포 착화합물 형성에 대한 평형 상수를 구하였다. 사슬이 긴 경우(n=9, 12)에는 교환 속도가 느려 두 상태가 따로 분리된 흡수선을 보이므로 흡수선 아래의 면적으로부터 평형 상수를 구하였다. 위에서 구한 내포 착화합물 형성의 평형 상수는 사슬이 길어짐에 따라 증가 하였다. 같은 실험을 여러 온도에서 반복하여 각 온도에서의 평형 상수를 구하였으며 구한 평형 상수의 온도 의존성으로부터 내포 착화합물의 형성에 따른 엔탈피와 엔트로피의 변화를 구하였다. 엔탈피의 변화는 사슬 길이에 거의 무관하게 음의 값을 갖으며, 이는 α-CD와 guest 분자 사이에 우호적인 반데르 발스 상호작용에 기인하는 것으로 여겨진다. 반면 엔트로피는 사슬이 길어짐에 따라 현저한 증가를 보였으며 이는 착물 형성됨에 따라 guest 분자 주위의 구조화된 물이 감소하는 효과로 설명될 수 있다. 따라서 사슬 길이가 증가하면서 평형 상수가 커지는 현상은 엔탈피의 감소가 아닌 엔트로피의 증가가 주 원인이 됨을 알 수 있다. 사슬의 길이가 n=3~7의 범위에서 길어 질수록 α-CD의 H_(3)의 CIS 값은 조금씩 downfield로 이동하였으며 이는 α-CD가 naphthol 부분에서 점차로 알킬 사슬 쪽으로 이동했음을 의미한다. 또한 n=9, 12의 경우 2D ROESY 스펙트럼에서 분자의 가운데 알킬 사슬과 α-CD사이의 교차 피크를 확인하였고 이로부터 CD가 guest 분자의 알킬 사슬에 위치함을 알 수 있었다.;The conformational equilnbria and complexation behavior with α-CD of a homologous series of β-Nap-O-(CH_(2))_(n-)MV^(2+) 2Cl- molecules are studied by proton NMR spectroscopy. These molecules are interesting because the electron donor (naphthol) and the acceptor (viologen) are linked via a flexible alkyl chain. Comparison of the proton chemical shifts of the linked compounds and those of disconnected constituents shows that the molecule exists in conformational equilibrium between the linear structure and the bent structure where the naphthol1 and the viologen groups have face-to-face configuration (intramolecular complexation). It is also found that the chemical shifts are strongly dependent on its concentration, which can be ascribed to formation of dimer molecules (intramolecular complexation). Further analysis, however, shows that the ionic strength of the solution plays an important role in concentration dependence of chemical shift for charged molecules. An excess NaCl solution was used to rule out the effect of ionic strength variation as the concentration of β-Nap-O-(CH_(2))_(n-)MV^(2+) 2C1- molecule being changed. The formation constants of inter- and intra- molecular complexation equilibria were obtained from nonlinear least squares analysis under this condition (K_(intra)=0.17, K_(inter)=14 M^(-1)). When the spacer alkyl chain becomes longer (n=9, 12), the molecule shows abrupt change of chemical shifts as the concentration increases, which is due to formation of aggregated species. The critical concentrations when the molecule starts to aggregate were evaluated to be approximately 1.7 mM for n=9 and 10 mM for n=12 molecules. Structures of intramolecular charge transfer complexes were also deduced from molecular modeling calculation. The energy-minimized structures agree well with the observed chemical shift changes as the chain length varies. Significant changes both in chemical shifts and in spectral shape are observed upon addition of α-CD due to complexation. The exchange rate is slow when the chain is long (n=9, 12), and separate peaks from the free and complexed species are observed. The complex formation constants can be obtained from the integrated signal intensities. The free and complexed molecules can not be distinguished by rapid equilibrium as the chain becomes shorter. Only the time-averaged signal can be observed in this case, and the complex formation constants are obtained by NMR titration method. The complex formation constant is found to become larger as the chain length increases. Enthalpy and entropy of complex formation were obtained from temperature dependence of the equilibrium constant. Negative enthalpy of complex formation indicates a favorable van der Waals interaction between the guest and CD molecules. Entropy of complex formation increases as the chain length increases, whereas enthalpy remains almost constant. This indicates that the increase in the formation constant as the chain length is dominated by the entropy factor, analogous to hydrophobic interaction. The chemical shift value of the complexed species gives insight on the primary inclusion site. Experimental data shows downfield shift as the chain length increases, which indicates that the primary inclusion site moves from the naphthol moiety to the alkyl chain. It was verified from the cross-peaks between alkyl chain and α-CD protons in ROESY experiment.
