- 청정 에너지원인 수소의 미생물에 의한 생산성 증대를 위하여 Clostridium butyricum 균체의 최적 고정화 조건을 결정하고, 고정화 균체의 수소 생성 및 glucose를 이용한 발효 양상을 관찰한 후에 C. butyricum의 발효 대사 산물을 소비하여 효율적으로 수소를 생성할 수 있는 Rhodopseudomonas sphaeroides 고정화 균체와의 혼합 수소 생성계 가능성을 검토하였다.
절대 혐기성 수소 생성 세균인 C. butyricum을 kappa-carrageenan, agar, alginate를 담체로 이용하여 entrapment법으로 고정한 결과 2% kappa-carrageenan gel에, gel내 균체의 농도가 10% 되게 하였을때 가장 수소 생성량이 많았다. 고정한 C. butyricum 균체는 생균체와 비교하여 30%에 달하는 총 수소 생성량 증가를 보였으며, 공기에 노출되어도 수소 생성 저해 효과를 받지 않았고, 장기간 저장 및 재사용이 가능하였다.
Clostridium butyricum 고정화 균체에 의한 glucose로부터의 회분 발효시 대사 산물 생성 양상은 생균체의 대사 산물 생성 양상과 유사하였으며, 단독 연속 수소 생성계에서 반응시킬 경우 dilution rate = 0.0736 h^(-1)일 때 최고 수소 생성 속도(56.6 ㎕·wet cell ㎎^(-1)·h^(-1))를 나타내었다.
광조사하에서 유기산을 이용한 수소 생성능을 가진 광합성 세균 Rhodopseudomonas sphaeroides를 고정화 하여 clostridial metabolites인 유기산(acetate, butyrate)을 기질로 하였을때 수소 생성을 하는지 확인하였다.
두 균주의 고정화 세포 혼합 반응계에 의한 수소 생성량은 한 gel안에 두 균체를 혼합시켜 고정하여 이용하는 것보다, 두 균체를 각각 2% carrgeenan gel에 고정하여 각 습균체량 비율이 C. butyricum : R. sphaeroides = 1 : 9 되게하여 이용하는 것이 총수소 생성량이 많았으며 dilution rate = 0.073 h^(-1) 일 때 57.5 ㎕·wet cell ㎎^(1-)·hr^(1-)의 속도로 수소를 생성하였다.;Hydrogen production by immobilized cells of Clostridium butyricum NCIB 9576 and Rhodopseudomonas sphaeroides K-7 were investigated.
Whole cells of Clostridium butyricum NCIB 9576, an hydrogen-producing strict anaerobic bacteria, were entrapped in 2% kappa-carrageenan gel. Total amounts of hydrogen produced by immobilized cells of C.butyricum was 30% increased than that of intact cells.
Immobilize cells had an resistance to inhibition effect on hydrogen production by air-exposure and showed good storage stability within 3 months. In batch culture, the pattern of clostridial metabolites production was similar to that of intact cells. In continuous culture, the optimum dilution rate was 0.0736 h^(-l) and the hydrogen production rate was 56.6μ1ㆍmg^(-1) wet cellㆍh^(-l).
Immobilized cells of Rhodopseudomonas sphaeroides K-7 were capable of producing molecular hydrogen by consuming clostridial metabolites (acetate, butyrate) under light irradiation (12,000 Lux). For a efficient bioconversion of glucose into hydrogen, mixed culture of immobilized cells of both anaerobic bacteria was performed. 15-20% of total amounts of hydrogen production was increased than that of monoculture of Clostridium butyricum immobilized cells and the optimum mixing ratio of two strains for mixed culture was C.butvricum : R.sphaeroides = 1 : 9.
