In Pakinsonian brains. the level of zinc in substantia nigra (SN) is known to be higher than in normal brains. However, the direct effect of Zn^(2+) on SN neurons has not yet been elucidated. Therefore, to explain the role of Zn^(2+), I examined the effect of Zn^(2+) on the electrical properties of neurons that were acutely isolated from the SN area of young rat brains, using current- and voltage-clamp methods of whole-cell recording mode. Current-clamp studies showed that Zn^(2+) induced dramatic hyper-polarizations from SN neurones at resting state. Also Zn^(2+) shortened the durations of spikes elicited by current injections (0.05 nA; 200 ms) and developed fast afterhyperpolarizations. As a result, Zn^(2+) appeared to increase firing frequencies of evoked spikes. All these events occurred dependently on the concentrations of Zn^(2+) applied. Voltage-clamp studies revealed that Zn^(2+) decreased a transient potassium current (A-type) and increased a slowly inactivating potassium current. These results strongly suggest the modulation of voltage-gated potassium channels as a candidate responsible for the changes in excitability of SN neurons by Zn^(2+). | - |
