Abstract
뇌의 한 부분인 basal ganglia는 개체의 움직임의 순서를 계획하고 그 시작을 일으키는 역할을 한다. 이러한 basal ganglia의 일부인 substantia nigra (SN)에는 dopamine을 합성하는 세포들이 존재하는데. 이러한 세포들이 어떤 요인에 의해 죽어 감으로써 파킨슨 씨 병이 생긴다. 이러한 병에 걸린 환자의 substantia nigra에는 특히 금속 이온들이 많이 모여 있는데, 그 하나가 바로 Zn^(2+)이다. 본 실험은 substantia nigra에 있는 신경세포의 Zn^(2+)에 의한 활성 변화를 조사한 것이다. Zn^(2+)는 SN neuron의 휴지기 전위 (resting membrane-potential)를 정상적인 상해보다 과분극 (hyperpolarization) 시켰다. 따라서 Zn^(2+)에 의해 세포의 활성이 일차적으로 저하된 듯이 보였다. 그리나 이후 제공되는 (+) 전류 자극에 의해 생기는 spike를 비교 해보면, 그 지속기간 (duration)이 감소하였고, afterhyperpolarization (AHP)이 증가하였다. 또한 spike 빈도 역시 늘어났는데, 이러한 것들로 보아 세포의 활성이 더욱 증가한 듯 보였다. 이러한 세포 활성의 변화는 특히 transient potassium current와 유사한 current의 감소와 slowly inactivating potassium current의 증가에 의하여 발생하는 듯 하였다. 이처럼 Zn^(2+)는 세포의 휴지기 전위를 과분극 시켰고, 동시에 다음 자극에 더욱 민감하도록 세포의 활성을 증가시켰다. 즉, 직접적으로 SN neuron의 행동에 영향을 주었으며, 이러한 세포들의 활성 변화는 한 개체의 행동까지도 변화시키리라 예상된다. 또한 이러한 금속 이온의 효과 및 작용기작을 분자 수준에서 밝힘으로 파킨슨 씨 병의 병리적 상황인 금속 이온의 축적이 만들어 내는 생리적 변화를 알아낼 수 있으리라 생각된다.;In Pakinsonian brains. the level of zinc in substantia nigra (SN) is known to be higher than in normal brains. However, the direct effect of Zn^(2+) on SN neurons has not yet been elucidated. Therefore, to explain the role of Zn^(2+), I examined the effect of Zn^(2+) on the electrical properties of neurons that were acutely isolated from the SN area of young rat brains, using current- and voltage-clamp methods of whole-cell recording mode. Current-clamp studies showed that Zn^(2+) induced dramatic hyper-polarizations from SN neurones at resting state. Also Zn^(2+) shortened the durations of spikes elicited by current injections (0.05 nA; 200 ms) and developed fast afterhyperpolarizations. As a result, Zn^(2+) appeared to increase firing frequencies of evoked spikes. All these events occurred dependently on the concentrations of Zn^(2+) applied. Voltage-clamp studies revealed that Zn^(2+) decreased a transient potassium current (A-type) and increased a slowly inactivating potassium current. These results strongly suggest the modulation of voltage-gated potassium channels as a candidate responsible for the changes in excitability of SN neurons by Zn^(2+).
