dc.description.abstractGelatin microcapsules containing Ftorafur, a hydrophillic anticancer agent, were prepared with congealable disperse-phase emulsification method. The preparation was based on dispersion of ftorafur-gelatin (Tween #40 or Span #20) solution in liquid paraffin. The F-G microcapsules were washed with cyclohexane and treated with benzethonium chloride, cationic surfactant, to prevent the microcapsules from aggregating. As the hardening time of F-G microcapsules, mean particle size was not changed in comparison with control. In the case of F-G microcapsules with Tween #40, Span #20, mean particle size decreased and narrow size distribution was observed. Ftorafur content of F-G microcapsules hardened with formaldehyde solution was not changed. In the case of F-G microcapsules with Span#20, ftorafur content of F-G microcapsules was much than that of F-G microcapsules without surfactant. T_(50%) of F-G microcapsules was less than ftorafur powder. The intrinsic dissolution rate of ftorafur in microcapsules with 1% Span#20 was 8.5 times smaller than that of intact ftorafur.;수용성 항암제인 Ftorafur의 Gelatin microcapsules (이하 F - G microcapsules 이라 한다.) 을 Congealable Disperse - Phase emulsification법에 의해 제조하였다. Liguid paraffin중에 Ftorafur와 Tween #40 혹은 Span #20을 함유하는 Gelatin 수용액을 분산시켜 F - G microcapsules을 제조하였다. F-G microcapsules의 세척용매로 Cyclohexane을 사용하였으며 세척액 중에 양이온성 계면활성제인 Benzethonium chloride을 첨가하여 응집을 방지하였다. F-G microcapsules 의 평균입자경은 Formaldehyde solution으로 경화시켰을 때 차이를 나타내지 않았으며 Tween #40, Span #20을 0.5 % 및 1 % 사용하였을 때 평균입자경의 감소와 비교적 균일한 입자도를 나타내었다. F-G microcapsules 중 Ftorafur의 함량은 경화시키지 않았을 때 10.17 %, Formaldehyde solution으로 30분, 1시간, 4시간, 24시간 경화시켰을 때 9.34 %, 9.32 %, 9.33 % , 9.33 % 이었다. Span #20을 첨가하였을 때 계면활성제를 사용하지 않았을 때보다 Ftorafur 함량이 증가하였다. T_(50%)는 원말에서 1.5분, 경화시키지 않았을 때 5분이며 30분, 1시간, 4시간, 24시간 경화시켰을 때 각각 6분, 7분, 7분, 8.5분이었다. Span #20 첨가로 다소 용출을 지연시켰다. Intrinsic dissolution rate는 Span #20을 1 % 첨가하였을 때 Ftorafur 원말에 비해 8.5배 감소하였다.-
