- 여러가지 종류의 small GTP-binding 단백질이 동물 조직에서 발견 되고 있는데, 이러한 단백질이 식물 세포에서도 존재 하는가를 조사 하였다.
[α-(32)^P]GTP-binding 실험을 통하여 조사한 결과 21, 23, 및 27kDa의 단백질이 단자엽 식물인 옥수수와 쌍자엽 식물인 콩의 막과 세포질에 모두 존재하는 것으로 나타났다. 이 세가지 단백질은 GTP 유사체인 Gpp(NH)p와 GDP를 함께 처리한 binding 실험에서 GTP와 특이하게 결합하는 것으로 나타났으며, GTP이외에 다른 누클레오티드와는 결합하지 않았다. 또한, 막에 hydroxylamine을 처리했을 때 막에서 떨어져 나오는 것으로 보아 막에 fatty acylation 상태로 붙어 있다고 여겨진다.
이상의 단백질들에 GTPase 작용이 있는가를 확인하기 위하여, 27kDa 단백질을 콩 hypocotyl의 crude한 막을 이용하여, Sephacryl S-300 gel filtration, Phenyl-sepharose CL-4B column, Hydroxyapatite column, Q-sepharose column chromatography의 과정을 통하여 부분적으로 분리, 정제 하였고, 이 27kDa 단백질에 GTPase activity가 있음을 확인 하였다.
이상의 결과들로 미루어 식물에도 동물의 small GTP-binding protein과 유사하게, GTP에 대한 specific binding 성질과 GTP를 가수분해하는 GTPase activity를 가진 단백질들이 존재하고 있음을 확인 하였다.;There is a family of homologous proteins known to be small GTP-binding proteins which have a GTP-binding domain and GTPase activity in various animal tissues. To identify this family of proteins in plant cells, GTP-binding study was performed with electroblotted plant proteins following SDS-PAGE using [α^(-32)P]GTP. Three small GTP-binding proteins (21, 23, and 27kDa) which have a specific GTP binding properties were identified in the membrane and cytosolic fractions of both monocotyledon (Zea mays L. ) and dicotyledon (Glycine max). Moreover, these three small GTP-binding proteins were gradually decreased when membranes were treated with hydroxylamine. This results indicate that three small GTP-binding proteins in plant cells are fatty acylated to membrane lipids.
The 27kDa protein was partially purified from hypocotyl membranes of Glycine max, following Sephacryl S-300 gel filtration, Phenyl -sepharose CL-4B column chromatography, Hydoxyapatite column chromatography, and Q-sepharose column chromatography. This 27kDa protein was found to have both GTP-binding and GTPase activities.
