dc.description.abstractFor the improvement of continuous saccharification process, saccharification of starch by the immobilized amylase was investigated. Among the fungal amylases and bacterial amylases, a α-amylase from Bacillus licheniformis and a glucoamylase from Aspergillus oryzae were selected and were used throughout the experiments. From the point of Substrate affinity and dextranization condition of substrate, soluble starch pretreated at 70℃ to 80℃ for 20min to 30min and corn starch pretreated at 100℃ for 20min to 30min showed a higher amylase activity. Amylase immobilization to 10% polyacrylamide, 2% alginate, chitin, silica gel and oxirane acrylic bead was carried out by entrapment, adsorption and covalent binding method. From the results, immobilized amylase on chitin got the highest stability and efficiency in terms of the enzyme activity. The cross-linking agents of α-amylase for efficient immobilization were glutaraldehyde and acetaldehyde. The optimum ratio of α-amylase to chitin was 3 to 7(enzyme unit/μgchit The optimum condition for immobilized α-amylase on chitin was 80℃ to 90℃ and pH5.5 to 6.5. The enzyme was activated by the additions of Ca^(2+) Na^(+) and was stable at the wide range of temperature and pH ( 20℃-90℃ pH5.0-11.0). The cross-linking agent of glucoamylase for efficient immobilizatic was acetaldehyde. The optimum ratio of glucoamylase to chitin was 42 tc 7 (enzyme unit/mg chitin). The optimum condition for immobilized glucoamylase on chitin was 65℃ and pH5.5. The enzyme was activated by the additions of Ca^(2+),Co^(2+), Li^(+), Mg^(2+),and NH^(4+) to the reaction mixture and was stable at the range of 20℃-50℃ and pH5.0-7.0.;Amylase 고정화를 통한 연속 당화 공정을 개발하기 위해 선정된 액화, 당화 효소를 다양한 담체와 방법으로 고정화 시켜, 그중 가장 안정성과 효율이 높은 담체에 고정한 amylase의 반응 조건을 검토하여 고정화 α-amylase와 g1ucoamylase를 같은 반응기내에서 당화시키는 방법의 가능성을 검토하였다. Fungal amylase와 bacterial amylase 종에서 α-amylase는 Bacillus licheniformis에서 생산한 것을 glucoamylase는 Aspergillus oryzae로 부터 나온 효소를 선정하여 사용하였다. 기질은 친화성이 높은 가용성 전분과 옥수수 전분을 선택하여 호화 반응조건을 검토한 결과, 가용성 전분은 70-80℃에서 20-30min, 옥수수 전분은 100℃에서 20-30min 열처리 한것이 활성이 높았다. 담체로는 10% polyacrylamide 단독 혹은 5% ployvinylalcohol, 1% alginate, 1% polypyrrolidone과 혼합시켜 entrapment방법으로 고정했고 oxirane acrylic bead에 covalent binding, silica gel, chitin에 glutaraldehyde로 activation시켜 binding시켰고 2% alginate를 entrapment방법으로 고정시킨 결과, chitin이 α-amylase, glucoamylase 양쪽에서 효율과 안정성이 높아 고정화 담체로서 선정했다. 선정된 Chitin에 고정시킨 amylase의 고정화 조건과 최적 반응조건을 검토한 결과, α-amylase는 glutaraldehyde, acetaldehyde의 cross-linking agent를 사용하고 효소 3unit당 7㎍의 chitin으로 고정 했을때 고정화 효율이 증대 되었다. 고정화 후의 반응조건은 80-90℃, pH 5.5-6.5에서 최적을 보였고 Ca^(2+), Na^(+)에 의해 효소 활성이 촉진 되었으며 pH 5.0-11.0, 90℃까지의 온도에서 안정하였다. Glucoamylase는 acetaldehyde 사용시, 효소 42unit당 7㎎의 chitin을 사용 했을때 고정화 효과가 좋았다. 고정화 후의 반응조건은 65℃, pH 5.5에서 최적을 보였고 Ca^(2+),Co^(2+), Li^(+), Mg^(2+), NH^(+)_(4)에 의해 효소활성이 촉진 되었으며 pH5.0-7.0, 50℃까지의 온도에서 안정하였다.-
