家兎에서 實驗的으로 誘導된 면포에 Vitamin A acid 塗布가 미치는 影響에 관한 電子顯微鏡的 硏究

Abstract

좌창의 치료에는 여러가지 종류의 약제들이 이용되고 있으나 특히 13-cis-retinoic acid(RA)는 좌창 치료를 위한 국소도포제의 활성성분으로 널리 사용되며 치료효과가 우수한 것으로 알려져 있다. 저자는 RA의 어떠한 작용기전이 좌창 치료에 도움을 주는지를 알아보기 위하여 본 실험을 시행하였다. 실험방법으로는 파라핀유에 용해시킨 50% oleic acid를 가토의 양측 외이도에 1일 1회 3주 동안 도포하여 면포를 형성시킨 다음 좌측 귀를 대조군으로 하였으며 우측 귀에 0.1% RA를 1일 1회 3주간 도포하여 미세구조의 변화를 관찰하여 다음과 같은 결과를 얻었다. 1. RA도포 2주후 부터 각질세포층의 수가 현저히 감소하기 시작하여 도포 3주후에는 정상 모피지관의 각질세포층과 유사하였다. 2. 교소체와 변형 교소체 및 tight junction의 수는 RA 도포 2주후부터 감소하기 시작하여 도포 3주에는 현저히 감소되었다. 3. RA도포 기간이 길어짐에 따라 세포간 간격은 점차 넓어져 표피탈락의 정도가 심해졌다. 4. 대조군에서 거의 발견되지 않았던 피막과립은 RA도포 1주후부터 출현하기 시작하여 도포기간이 길어짐에 따라 현저히 증가되었다. 이상의 결과로 볼 때 RA는 세포간 결합력을 감소시켜 표피탈락을 초래하여 면포를 소실시키는 작용이 있다.;Comedones were induced at the external ears of rabbits by daily applications of 50% oleic acid in paraffin oil for 3 weeks. 13-cis-retinoic acid (RA) solution (0.1%) was applied to experimentally induced comedone observed. The morphologic changes around the epithelial lining of comedones were observed by electron microscopy to define the effect of RA on comedones. In follicles treated with RA, the thickness of the stratum corneum of the epithelium was markedly decreased with widening of intercellular spaces. The acid also produced decrement of the number of desmosomes and desmosomal bodies between horny cells. Abnormally appeared tight junctions in the stratum corneum and disappearance of membrane coating granules in the stratum granulosum were reversed to normal state, disappearance of tight junction and increment of membrane coating granules respectively by the application of RA. These observations suggest that RA has comedolytic effect by decreasing cohesiveness between horny cells.
