Sarcoma 180 암세포가 이식된 마이스에 있어서 NK 세포활성도 변화에 관한 연구

Abstract

NK cell은 종양세포와 virus 감염세포에 대항하여 생체방어의 중요한 역할을 하는 것으로 나타났다. NK cell의 작용기전에 대해서 완전히 알려지지는 않았지만 spontaneous lymphocyte mediated cytotoxicity (SLMC)를 나타낸다고 보고되었다. 자연적으로 발생된 암세포인 Sarcoma 180이 이식된 Swiss Webster mouse에서 암세포의 증식에 따른 NK세포활성도 변화를 추적하기 위하여 암세포의 이식전과 이식후 2일, 6일, 10일에 ^(51)Cr 방출법으로 각각 측정하였다. 암세포에 대한 splenocyte의 NK세포활성은 이식직후 높게 나타났는데 대조군에 비하여 100 : 1의 경우 18.6%, 50 : 1의 경우 11.7% 증가하였다가 그 후 감소하였다. 이식후 2일에 NK세포활성이 증가된 것은 소량의 암세포에 대항하여 즉각적인 생체방어의 역할을 수행한 것으로 생각되며 그후 증식에 의한 대량의 암세포에 의해 NK세포활성이 감소한 것으로 사료된다.;Natural killer cells appear to have important roles in host defense against nascent tumors and viral infection. The mechanism of natural killer activity has not been studied extensively, but it was reported that these cells had spontaneous Iymphocyte mediated cytotoxicity (SLMC). NK activity of Swiss Webster splenocyte was investigated before and at different time imtervals after transplantation of Sarcoma 180 tumor cells. The levels of NK activity against tumor cells in S.W. splenocyte was shortly augmented after transplantation. This augmented cytotoxicity reached a peak at 2 days after transplantation and then declined. The results indicate that this augmentation of cytotoxicity may be associated with immediate host surveillance against tumor cells and that decrease may be caused by rapid tumor growth.
