Abstract
This study was designed to further purify partially purified GI fraction which showed radiation-protective effects. To obtain GI fraction, Tris-HCI extraction, ammonium sulfate fractionation, heat inactivation, carboxymethyl-cellulose column chromatography, and sephadex G-75 column chromatography were conducted. Sephadex G-200 column chromatography was performed on GI fraction and three fractions, FⅠ, FⅡ and FⅢ were obtained. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis was carried out on these fractions, and as the results FⅠ fraction showed four bands, of which subunit molecular weights were 18,000, 38,000, 55,000 and 94,000 estimated from regression line for standard proteins and FⅡ fraction showed one band, of which subunit molecular weight was 22,000. FⅢ fraction showed one band of which subunit molecular weight was 18,500.;인삼에서 방사선 방어작용이 길다고 보고된 단백질 분획을 추출, 정제하여 활성성분을 밝혀내고 부작용이 없는 생약제제의 항암제를 개발하기 위하며 부분정제된 단백질에 대해 sephadex G-200 column chromatography를 시행하였다. 그 결과 세가지 분획, FⅠ, FⅡ 그리고 FⅢ를 얻었으며 각 분획에 대한 SDS-polyacrylamide gel electrophoresis를 시행하여 표준단백질의 이동도와 비교하여 각 분획이 함유하는 단백질의 subunit분자량을 추정하였다. 그 결과 FⅠ은 네개의 band를 나타내었고 그 분자량은 18,000, 38,000, 55,000 및 9 4,000이었고 FⅡ는 1개의 band를 나타내었고 그 분자량은 22,000이었으며 FⅢ 역시 1개의 band를 나타내었고 분자량은 18,500 이었다.
