Abstract
Lad는 Lck SH2 domain과 결합하며, TCR signaling에 중요한 adaptor 단백질로 최근에 알려졌다. 이 논문에서는 Lad의 새로운 기능을 규명하기 위해서 Lad의 세포내 위치와 Lad가 결합하는 단백질을 연구하였다. Jurkat T cell에서 Lad는 TCR stimulation에 의존하여 많은 세포신호전달 단백질들이 모이는 GEM fraction으로 모이는 것을 확인하였다. 더욱이 T cell과 APC가 만나서 형성하는 immunological synapse에서 Lad 는 c-SMAC에 위치한다. 또한 Lad는 TCR activation시 Lad가 빠른 시간안에 (5 분) synapse 부위로 옮겨 가는 것을 볼 수 있었고, 조금 지체된 시간에는 (15, 30분) synapse에서 벗어나는 현상을 볼 수 있었다. 늦은 시간에서는 (3 시간 이후) Lad가 synapse의 반대 방향에 존재한다. 아울러6시간동안 TCR stimulation을 주었을 때는 Lad가 phosphotyrosine과 같이 위치함을 확인하였다. 이는 Lad가 T cell activation시 early와 late 단계에서 모두 중요한 기능을 한다고 생각이 된다. 또한 Lad가 adaptor단백질임을 감안하여 다른 단백질들과 결합하는지를 알아보았다. 첫번째 Lad의 어느 부위가 p56Lck와 결합하여 tyrosine-phosphorylation되는지 확인하였고, 이 부분을 통해서 TCR-stimulated IL-2 promoter 활성을 중가 시키는 것을 알았다. 두번째로 yeast two hybrid screening을 통해서 여려가지 Lad 와 결합하는 단백질들을 ( Vav, Nck, Grb2 and PLC-γ) 찾았으며, 실제로 이들이 단백질-단백질 결합하여 어떻게 T 세포 항원 수용체의 신호전달에 기능을 하는지 밝히고자 하였다. Nck, Grb2 and PLC-γ와 Lad는 Lck kinase 의 작용이 있을때에만 결합하는 것을 293T 세포에서 관찰 할 수 있었다. 또한 Vav, Grb2 and PLC-γ 는 Lad 와 in vivo 결합을 하는 것을 확인하였다. 이로써 Lad 는 많은 다른 신호전달 단백질들과의 결합을 유도하는adaptor 단백질로써 APC에 의해 T 세포가 활성화되는 immunological synapse에서 활성화의 초기에 작용을 하지만, T 세포의 활성화가 계속 유지되는 동안에 Lad가 계속 관여할 것으로 생각된다. ; An adaptor protein, Lad, was previously cloned as a binding partner of Lck SH2 domain and shown to be important in TCR-mediated T cell activation(1,39,40). In this report, to investigate further the function of Lad, I studied the subcellular localization and binding partners of Lad. Upon TCR stimulation of Jurkat T cells, Lad was recruited to GEM fraction, where central signaling molecules were concentrated. Furthermore, upon conjugation of T cells with APC, Lad was colocalized in the central-SMAC at an immediated-early stage of T cell activation (5 min). Subsequently, Lad translocated to a distal site within immunological synapse (15min ? 1hr). And finally, at the later stage (after 3hrs), Lad was redistributed to the entire plasma membrane as punctated structures, and in approximately 30% of conjugated samples, Lad was localized at the opposite site of the contact. Strikingly, the distribution of Lad exactly overlapped with that of tyrosine phosphorylation throughout the stage of T cell activation up to 6 hrs. These data show that the localization of Lad is dynamically regulated and Lad functions at both early and later stage of T cell activation. Additional alternative approaches to understand functions of Lad, I also studied the binding partners of Lad. First, employing the various point mutants of Lad, tyrosine residue in the Lad responsible for binding was mapped, and it was found that the binding of Lad to Lck is required for function of Lad in T cell activation leading to IL-2 gene expression. Since the overexpression of mutants incapable of binding to Lck abolished the TCR-stimulated IL-2 promoter activation. Secondly, based on the yeast two hybrid screening results with Lad as a bait, Lad was found to interact with PLC-γ, Grb2 and Nck in an Lck-dependent manner upon coexpression in 293T cells(1,39,40). Additionally, the in vivo binding of Lad to PLC-γ, Grb2 and Vav was confirmed using immunoprecipitation. and western analysis. Taken together, these data show that during T cell activation by APC, Lad acts in the immunological synapse as an adaptor protein recruiting various signaling molecules and the function of Lad is not only confined in the early stage but also sustained throughout the later stage of T cell activation.
