Abstract
Histamine releasing factor(HRF), p23, 또는 p21이라고 알려져 있는 translationally controlled tumor-associated protein(TCTP)은 eukaryotes의 다양한cell type에서 풍부하게 발견되며 종 간의 sequence 보존 정도가 높고, 다른 단백질 family와는 homology를 가지지 않는 것으로 보고된 바 있다. TCTP는 cell growth와 human late allergy 반응에 관여하고 calcium binding 가능성 또한 확인되었으나 생물학적 기능이 명확히 밝혀지지는 않은 상태이다. TCTP의 또 다른 기능과 관련하여 Plasmodium falciparum TCTP는 heme 존재하에서 항말라리아 약인 artemisinin과 결합한다는 보고가 있다. Heme과의 반응으로 activatio된 artemisinin의 endoperoxide bridge가 열리면서 free radical을 생성하고 Plasmodium falciparum TCTP의 Cys14를 alkylation 시킬 것으로 추정되고 있다. 이 연구에서는 artemisinin의 항말라리아 작용기전을 규명하기 위해 artemisinin이 heme 또는 TCTP와 어떠한 결합 양상을 보이는지를 살펴 보았다. 이를 위해 homology modeling에 의한 artemisinin의 3차원 구조 구 성과 이 구조에 대한 heme과 artemisinin의 상호작용을 docking의 방법으로 연구하였다. Swiss-Model, Geno3D, COMPOSER, MODELLER과 같은 modeling program을 이용하여 Schizosaccharomyces pombe TCTP (PDB code 1H6Q)를 template로 P. falciparum TCTP의 3차 구조를 구성하였다. 각각의 program으로 생성된 4개의 model들을 2차 구조, 생물학적인 기능 그리고 PROSAII program으로 평가하여 MODELER model이 가장 합당한 구조로 판단되었고 이를 최종적인 P. falciparum TCTP의 구조로 사용하였다. 또 artemisinin과 heme 또는 P. falciparum TCTP 사이의 반응 양상은 Flexidock을 이용한 docking study로 살펴 보았다. Artemisinin과 heme의 docking 연구에서 heme의 Fe부분과 artemisinin의 peroxide ring의 중앙부분이 2.6A의 거리로 근접하게 docking되었다. 또. P falciparum TCTP의 MODELLER model과 artemisinin의 docking 연구에서는 C4-radical 부분이 Cys14의 2.48A 거리에서 sulfur를 향하여 docking됨으로써 지금까지 보고된 실험적인 결과와 일치하 였다. 본 연구는 artemisinin의 항말라리아 작용이 heme과의 결합과 TCTP의 alkylation에 기인할 가능성을 제시하였다. ;The translationally controlled tumor-associated proteins (TCTPs) are a highly conserved and abundantly expressed family of eukaryotic proteins that are implicated in both cell growth and human acute allergic response but whose intracellular biochemical function has remained elusive. In 1998, Meshnick reported that antimalarial drug, artemisinin, binds to Plasmodium falciparum TCTP in the presence of heme. That is, heme-catalyzed cleavage of the endoperoxide bridge in artemisinin forms a free radical, which is followed by a specific alkylation of P. falciparum TCTP. In our study to illustrate the mechanism of antimalarial activity of artemisinin, 3D structures of P. falciparum TCTP have been constructed by homology modeling method using NMR structure of Schizosaccharomyces pombe TCTP (PDB code 1H6Q) as a template. Homology modeling was performed by Swiss-Model, Geno3D, COMPOSER and MODELLER programs. The four models obtained by the four programs were evaluated by their secondary structures, biological functions and PROSAII program. As the result, the MODELLER model was chosen as the best one of them. To determine the binding modes between the artemisinin and P. falciparum TCTP, docking experiments were carried out using Flexidock program. Docking simulation of artemisinin with heme was first performed to show that the peroxide bridge of artemisinin was docked facing to Fe in heme in the distance of 2.6A. Then, the activated artemisinin was docked into the P. falciparum TCTP in 2.48A from the sulfur of Cys14. The interaction of the activated C4 radical of artemisinin with Cys14 of TCTP conformed the experimental data reported. Our studies provide evidence that the antimalarial effect of artemisinin may be due to the interaction with heme and alkylation of TCTP by the drug.
