Abstract
Voltage-gated calcium channels에 대한 아연 (Zinc)의 효과는 감소하는 현상만이 일부 보고되었을 뿐 그에 대한 생리적인 의미나 그것의 하위 mechanism에 대해서 구체화된 것은 없다. 특히 간질 (epilepsy)과 관련하여 low-threshold calcium channels이 거론되고, 다른 한편으로 간질에 대한 아연의 역할을 제안함을 볼 때 아연에 의한 low-threshold calcium channels의 작용 mechanism이 무관하지 않을 것으로 추론 된다. 이 논문에서는 whole-cell voltage-clamp technique을 사용하여 특히 thalamic relay neurons에서 firing patterns을 결정짓는 low-threshold calcium channels을 아연이 어떻게 감소시키는지에 대한 mechanism을 밝혀 보았다. 기존의 보고와 무관하지 않게, thalamic relay neurons에서도 아연은 low-threshold calcium currents를 농도 의존적으로 감소시켰으며 hill coefficient는 0.97을 나타내었다. 아연에 의한 low-threshold calcium currents의 감소를 설명할 수 있는 가능성은 다음과 같다. 우선, 아연에 의한 channel의 gating kinetics의 변화이다. 아연에 의한 currents의 감소가 gating kinetics의 변화에 의해 이루어진다면 activation kinetics는 느려질 것이고 inactivation kinetics는 빨라질 것으로 예측할 수 있다. 하지만 예상과는 달리, 아연은 activation과 inactivation의 gating kinetics에는 전혀 영향을 주지 않았다. 다음으로, 아연에 의한 activation과 inactivation에 대한 voltage-dependency의 변화를 살펴보았다. 아연에 의한 currents의 감소를 근거로 한다면 voltage-dependency는 아연에 의하여 inactivation은 hyperpolarized 방향으로, activation은 depolarized 방향으로 이동할 것으로 예상할 수 있다. 그러나 activation과 inactivation 모두 depolarized 방향으로 이동하였고, 이러한 변화가 아연에 의한 current의 현저한 감소를 설명하지는 못하였다. 마지막으로 fluctuation analysis를 통하여 아연에 의한 channels의 수와 single currents의 변화를 살펴보았다. Fluctuation analysis에 따르면 아연에 의한 감소패턴은 single currents의 감소보다는 channels 수의 감소에 더 잘 들어맞았다. 따라서, 아연에 의한 low-threshold calcium currents의 감소는 gating kinetics나 voltage-dependency의 변화와 무관하게 channels 수를 감소시킴으로써 이루어지는 것으로 추정된다. ; Although Zn^2+ is well known to reduce voltage-gated Ca^2+ currents in several preparations, no biophysical mechanisms underlying it have been yet completely elucidated. Here we thus examined, using whole-cell voltage-clamp technique, by which mechanism Zn^2+ reduced low-threshold Ca^2+ currents (LTCCs) that play an important role in determining firing patterns of thalamic relay neurons (TRNs). Zn^2+ dramatically reduced LTCCs in a concentration-dependent manner (IC50=55mM) with Hill coefficient of 0.97. Possible explanations for the reduction of LTCCs by Zn^2+ include changes in gating properties, the number of available channels, or single-channel conductance. Zn^2+ shifted both steady-state inactivation and activation of LTCCs to depolarizing directions so that voltage dependency of gating properties could not be the reason for the reduction of LTCCs by Zn^2+. Zn^2+ did not also change gating kinetics for either activation or inactivation. Accordingly, the reduction of LTCCs by Zn^2+ should not be due to changes in gating properties. Ensemble fluctuation analysis revealed that a model with a reduced number of channels fit the relations of fluctuation variance to current amplitude much better than that with a reduction in the single channel current. Therefore, we conclude here that the mechanism of block of LTCCs by Zn^2+ is by reducing the number of channels available to be activated.
