Abstract
세포의matrix adhesion 동안에syndecan-4 (transmembrane heparan sulfate proteoglycan) 는 focal adhesions 과 stress fibers의 형성 과정에서 중요한 역할을 수행한다. 이전에 우리는 syndecan-4 의cytoplasmic domain은 Protein Kinase C-a (PKC-a) 와 직접적으로 결합하며, in vitro에서 PKC-a 의 기능을 활성화하는 것을 보여주었다. 그러나, syndecan-4의 조절 기전은 in vivo에서는 아직 밝혀지지 않았다. Mammalian two-hybrid assays 는 syndecan-4가 PKC-a 와 in vivo에서 결합하며, 이러한 결합은 syndecan-4의 cytoplasmic domain을 통해서 이루어 짐을 증명하였다. 또한, PKC의 활성화는 그들간의 결합을 증가시킨다. syndecan-4를 과 발현하게 되면, endogenous 한 PKC-a 의 경우에 한해서 다른 타입과는 다르게 증가하며, 섬유세포와 상피세포 모두에서 detergent-insoluble fractions내로의 이동의 증가를 보인다. 이는 정상적인 syndecan-4의 경우에 한해서 특이성을 지니며, cytoplasmic domain이 결여된 syndecan-4 mutant의 경우는 그렇지 않다. 더구나, syndecan-4를 과 발현 시킨 REF (rat embryo fibroblasts) 의 경우, phorbol 12-myristate 13-acetate (PMA) 를 장시간 처리하는 경우나 혹은 suspension 배양에 의한 PKC-a의 downregulation의 유도가 지연된다. 또한, PKC-a immune complex kinase assays는 syndecan-4의 과 발현이 세포막에서의 PKC-a 의 활성을 증가시킴을 보인다. 그러므로, syndecan-4 는PKC-a 와 결합 하며, 이를 통해 PKC-a 의 세포 골격 내로의 이동과 활성과 안정화를 조절한다. 또한, syndecan-4 는 phosphatidylinositol 4, 5-bisphosphate (PIP₂) 와의 결합에 의해 PKC-a 의 결합과 활성을 조절하는 것이 알려져 있으며, 이는 syndecan-4 가 PIP₂ 의 기능을 조절함을 내포한다. phosphoinositides 는 PLC-PH domain 과 결합하기 때문에, PLC-d 의 amino terminus 에서 분리된PH domain 을 green fluorescent fusion protein (GFP-PLC-d PH) 으로 만들어, GFP-PLC-d PH domain 의 세포막으로의 이동을 확인함으로써, PIP₂ 의 안정화 과정에서의 syndecan-4 의 효과를 볼 수 있다. COS7 세포에 Syndecan-4 가 과 발현할 경우, PLC-d PH domain은 약 1.5 배의 증가된 세포막으로의 이동을 보인다. 외부로부터 세포막내로 더해진 PIP₂ 의 degradation 은 syndecan-4 cytoplasmic domain 에 의해서 지연되며, 이는 syndecan-4 가 regulate the PIP₂ 안정화를 조절함을 나타내는 것이다. ; During cell-matrix adhesion, syndecan-4 transmembrane heparan sulfate proteoglycan plays a critical role in the formation of focal adhesions and stress fibers. Previously, we have shown that syndecan-4 cytoplasmic domain directly binds Protein Kinase C-a (PKC-a), and activate its activity in vitro. However, the regulating mechanism of syndecan-4 in vivo remains unclear. Mammalian two-hybrid assays showed that syndecan-4 interacted with PKC-a in vivo, and this interaction was mediated through syndecan-4 cytoplasmic domain. Activation of PKC increased their interaction. Overexpression of syndecan-4, but not mutant lacking its cytoplasmic domain, specifically elevated amount of endogenous PKC-a and enhanced the translocation of PKC-a into detergent-insoluble fractions in both fibroblasts and epithelial cells. In addition, rat embryo fibroblasts overexpressing syndecan-4 exhibited much slower downregulation of PKC-a by either prolonged treatment with phorbol 12-myristate 13-acetate or suspension culture. PKC-a immune complex kinase assays showed that syndecan-4 overexpression increased the activity of membrane PKC-a. Therefore, syndecan-4 seems to interact with PKC-a, and regulates cytoskeleton localization, activity and stability of PKC-a. Syndecan-4 is also know to interact with phosphatidylinositol 4, 5-bisphosphate (PIP₂) to regulate interaction with and activity of PKC-a, implying that syndecan-4 might also regulate the function of PIP₂. Since it is know that the PLC-PH domain interacts with phosphoinositides, the isolated PH domain from the amino terminus of PLC-d was made a green fluorescent fusion protein (GFP-PLC-d PH) and judged form membrane localization of GFP-PLC-d PH domain, the effect of syndecan-4 on PIP₂ stability was investigated. Syndecan-4 overexpression in COS7 cells induced increased membrane localization of PLC-d PH domain by ~1.5 folds. Degradation of PIP₂, which was exogenously added into the plasma membrane was also delayed by syndecan-4 cytoplasmic domain, therefore syndecan-4 also regulate the PIP₂ stability.
