dc.description.abstract쌍편모조류(dinoflagellates)는 진핵세포성 조류(algae)로서 진핵세포의 특징과 원핵세포의 특징을 모두 가지는 생물이다. 이러한 쌍편모조류의 한 종인 Gonyaulx polyedra는 항상 응축되어 있는 모양과 크기가 비슷해 보이는 많의 수의 염색체를 가지며, 전체 DNA량이 대단히 많은 것으로 알려져 있다. Ribosomal RNA 유전자는 모든 생물체에 존재하며, 공통된 기능을 가지며, 진화적으로 잘 보존된 염기서열로 되어 있으므로, 진화적 관계를 연구하는데 많이 이용된다. 이러한 ribosomal RNA 유전자의 염기서열을 결정하는 것은 ribosomal RNA 구조, 기능, 진화적 관계를 이해하는데 기초적 단계이다. Gonyaulx polyedra에서의 ribosomal RNA 유전자의 클로닝(cloning)을 통 해 본 생물체의 ribosomal RNA 유전자의 특성을 이해하고, 진화학적 위치를 알아보고자 이 연구를 행하였으며, 그 연구 결과는 다음과 같다. 1. Gonyaulx polyedra에서 ribosomal RNA 유전자를 단리하여 클로닝 (cloning)하였고, 각 절편의 염기서열이 분석되었다. 2. PCR을 행한 결과, Gonyaulx polyedra ribosomal RNA의 반복단위가 8000 nt 정도이며, 이들이 머리-꼬리 구조로 반복되어 있음을 예상하였다. 3. Gonyaulx polyedra ribosomal RNA의 염기서열을 분석한 결과, 그 크기와 염기서열의 상동성이 원핵세포보다는 진핵세포와 유사하다고 여겨지며, ribosomal RNA의 염기서열을 이용하여 작성한 계통분류도도 Gonyaulx polyedra가 진핵생물 집단에 속해 있음을 시사한다. ; The din flagellates have some primitive nucleate features and an intermediate evolutionary position between prokaryotes and eucalypts. Due to specific characteristics such as its ubiquity and low evolutionary rate, ribosomal RNA has been most widely used tool for studying molestation. The repeating units coding for 17-18S, 5.8S, 25-28S ribosomal RNAs are organized in tandem arrays. The determination of the primary structure of ribosomal RNA genes is prerequisite step in understanding the ribosomal RNA structure, function, biogenesis, and evolution. In this study, the small subunit ribosomal RNA gene, the large subunit ribosomal RNA gene, and the 5.8S ribosomal RNA gene and the internal transcribed spacer (ITS) of Gonyaulax polyhedral have been cloned and their sequences were analyzed. The small subunit of ribosomal RNA gene was 1775 nt, the D1-D 2 domain and K 6-D 8 domain of large subunit ribosomal RNA gene was 844 nt, 767 nt, respectively, ad the 5.8S ribosomal RNA gene was 159 nt. The first internal transcribed spacer (ITS1) was 191 nt, and the second internal transcribed spacer (ITS2) was 185 nt. And, the intergenic spacer of ribosomal RNA gene (IGS) was estimated 2200 nt. The 5800 nt transcribed sequences was separated by roughly 2200 nt intergenic spacer. The ribosomal RNA genes are repeated many times and arranged in a head-to-tail, tandem repeated manner. The repeating unit of ribosomal RNA gene of Gonyaulax polyedra was proposed 8000 nt iong. The lengths of ribosomal RNA, sequences alignments with typical orgamism, and phyloggeneetic analysis based on ribosomal RNA pointed that Gonyaulax polyedra was closely related to eukaryotes.-
