Poly-β-(1→4)-N-acetyl-D-glucosamine의 구조를 가지는 chitin은 자연계에서 cellulose 다음으로 풍부하게 존재하는 생고분자 물질이다. 1811년 Braconnot에 의해 버섯에서 chitin이 최초로 분리된 이후로 chitin에 대한 연구는 주로 생물체에서의 분포, 세균에 의한 분해 및 화학적 특성에 관해 이루어졌으나, 최근 들어 chitin 유도체 및 그 분해산물인 N-acetylchitooligo- saccharides의 의료, 농업, 식품, 폐수처리 및 화장품 분야에의 응용성에 관심이 집중되었다. 이에 N-acetylchitooligosaccharides의 생산에 chitinase의 이용 가능성을 타진해보기 위해 한국 토양으로부터 분리한 chitin 분해 균주인 Cytophaga sp. HJ로부터 chitinase를 순수분리하여 그 효소의 특성 및 분해산물을 확인하였고, 한편으로는 vector로 pUC18을 이용하여 동일한 균주의 chitin 분해 유전자를 Escherichia coli JM 109에 클로닝하였다. Cytophaga sp. HJ가 생성하는 chitinase 단백질을 30-60% ammonium sulfate 분별침전, 1차 DEAE-Bio gel A column, Octyl-Sepharose CL-4B column, 2차 DEAE-Bio gel A column을 통해 순수분리하였다. Chitinase의 분자량은 59.8KDa이었고, 비활성은 0.19U/mg이었다. Chitosan보다는 colloidal chitin이 chitinase에 대하여 우수한 기질로 작용하였으며, chitosan의 탈아세틸화도가 높을수록 분해가 되지 않았다. Colloidal chitin에 대한 chitinase의 Km은 3.57mg/mL, Vmax는 2.37U/mL, Kcat은 0.60U/mg이었고, 75-85% deacetylated chitosan에 대한 chitinase의 Km은 1.64mg/mL, Vmax는 0.13U/mL, Kcat은 0.08U/mg이었다. Chitinase의 최적 반응 온도는 50℃, 최적 반응 pH는 4.0이었으며, 효소 활성이 Hg^2+, N-bromosuccinimide에 의해서는 약 90% 정도 감소되고, Ag^+, DEP, Sb^3+에 의해서는 약 50% 정도 감소되었으므로 효소의 sulfhydryl기, tryptophan, histidine이 효소 활성의 발현에 중요한 역할을 할 것으로 추정되었다. 또한 chitinase에 의한 colloidal chitin의 분해산물은 N-acetyl-D-glucosamine과 di-N-acetylchitobiose가 대부분이었고, tri-N-acetylchitotriose는 미량으로 생성되었으며, 반응시간이 길어짐에 따라 N-acetyl-D-glucosamine이 더욱 더 많이 생성됨을 알 수 있었다. 다른 균주의 chitinase와 N-terminal amino acid sequence를 비교한 결과 유사성이 낮았으며, sequence 길이가 너무 짧아 확정하기는 곤란하지만 database 분석 결과 동일한 단백질은 발견되지 않았으므로 처음으로 분리된 단백질일 가능성이 있다고 생각된다. 한편 pUC18을 vector로 이용함으로써 Escherichia coli JM 109에 Cytophaga sp. HJ의 genomic DNA를 클로닝하여 최종적으로 3.3Kb의 chitinase 유전자를 얻었으며, 이 유전자를 함유하는 Escherichia coli JM 109은 chitin을 분해하는 성질을 나타내었다. 이러한 결과를 바탕으로 하여 최종 분리된 chitinase 유전자(chi 3.3)의 DNA sequencing 및 chitinase 단백질의 N-terminal amino acid sequence와의 비교분석에 관한 연구가 계속 진행되면 Cytophaga sp. HJ에 있어서의 chitinase 성질 규명에 큰 진전이 있을 것이라고 생각된다. ; Chitin, poly-β-(1→4)-N-acetyl-D-glucosamine, a cellulose-like biopolymer, is the second most abundant organic compound on earth. Since chitin was described for the first time by Braconnot (1811) as a component of mushrooms, researches on chitin were mostly directed toward the study of its occurrence in living organisms, its degradation by bacteria and its chemistry. But recently, research activities were concentrated on the applications of chitin derivatives and N-acetylchitooligosaccharides in the field of medicine, agriculture, food industry, water treatment and cosmetics. In order to find a possibility of applying a chitinase to production of N-acetylchitooligosaccharides, we purified a chitinase from Cytophaga sp. HJ isolated from sea sand in Korea, and studied on the characteristics of the chitinase. The chitinase gene from the same bacteria was cloned in Escherichia coli JM 109 by using pUC18 as a vector. The chitinase protein was purified by using 30-60% ammonium sulfate precipitation, the first DEAE-Bio gel A column, Octyl-Sepharose CL-4B column, and the second DEAE-Bio gel A column. The purified protein degraded chitin preferentially. The chitinase had a molecular weight of 59.8KDa and a specific activity of 0.19U/mg. The chitinase acted on colloidal chitin better than chitosan as a substrate, and didn t degrade chitosan with a high degree of deacetylation. For colloidal chitin and 75-85% deacetylated chitosan, Km values were 3.97mg/mL and 1.64mg/mL, Vmax values were 2.37U/mL and 0.13U/mL, and Kcat values were 0.60U/mg and 0.08U/mg, respectively. The optimum temperature and the optimum pH for the chitinase activity were 50℃ and 4.0 respectively. Hg^2+ and N-bromosuccinimide inhibited the activity by 90%. Ag^+, DEP and Sb^3+ also inhibited the activity by 50%. From this results, we inferred that sulfhydryl moiety, tryptophan and histidine may be involved in the expression of the enzyme activity. TLC analysis of the enzymatic reaction products showed that the chitinase mainly produced N-acetyl-D-glucosamine and di-N-acetylchitobiose, and tri-N-acetylchitotriose. And the more the reaction time was lengthened, the more N-acetyl-D-glucosamine was produced. In the amino terminal region, there was low sequence homology with other microbial chitinases. Although the sequences were too much short to get a completely definitive conclusion, the purified chitinase from Cytophaga sp. HJ could be a new protein because there was no protein with the same amino terminal amino acid sequence in database. Also, the chitinase gene was cloned in Escherichia coli JM 109 by using pUC18 as a vector. Finally 3.3Kb chi gene was obtained and a clone harboring the gene expressed chitinase activity by forming a clear halo in LB agar medium containing 0.25% colloidal chitin. Still, the nucleotide sequence determination of chi 3.3 gene is required to get the full amino acid sequence information of the enzyme and to compare the nucleotide sequence to N-terminal amino acid sequence data.
