Abstract
최근 전형적인 NK세포와는 달리 TCRαβ 를 발현하는 TNK세포가 발표된 이후 이들 세포에 대한 관심이 높아지고 있으며, 이를 계기로 T세포와 NK세포의 근원 및 TNK세포의 면역계에서의 역할에 대한 연구들이 진행되고 있다. MHC의제한을 받는세포독성T림프구(CTL)가CD3^+subset인반면자연살해 세포(Natural killer cell)는 CD3^­subset으로 알려져왔다. 그러나, 자연살해 세포가 T 세포계에 속한다는 유력한 증거가 있다. 성장한 쥐의 흉선에 존재하는 것으로 최근에 밝혀진 lymphocyte subset인 CD3^+NK1.1^+TCRαβ (TNK) 세포는 성장한 쥐의 CD4^­CD8^­ 흉선세포에서 발견할 수 있다. 본 연구는 표현형이 NK1.1^+, CD3^­ TCRαβ^+ CD4^­ CD8^­세포인 CTLL-2 세포주를 이용하여, IL-2 존재하에서 이 세포들이 사이토카인과 세포 활성화 인자에 의하여 CD8 양성 세포 또는 CD4 양성 세포와 같은 TNK 세포로의 전환이 가능한지를 관찰 하고자 하였다. 한편 Transforming growth factor -β는 매우 다양한 기능을 가지고 있는데, 넓게는 세포의 성장과 분화를 조절하고 면역체계내에서 다양한 조절기능을 수행한다. TGF-β가 림프구의 성장을 방해하는 요소로 기술된 기존의 여러 연구들과 달리, 부동화된 anti - CD3 항체에 의해 활성화된 쥐의 T 세포의 성장을 촉진시킬수 있다는 보고도 있다. 본 연구에서는 T 세포의 성장과 분화에 미치는 TGF-β의 역할을 조사하고, 이것의 효과를 T 세포의 근원(ontogeny)에 중요하다고 알려진 다른 신호들과 비교해 보았다. TGF-β로 처리한 결과 그 용량에 의존한 성장억제와 형태학적 변화를 보였다. 통상적인 실험경과중 5%미만의 세포에서 CD8α 양성을 보였으나, IL-2 + TGF-β로 처리했을 때는 38%의 세포에서 CD8α 가 양성으로 나타났다. 반면, 12-o-tetradecanoyl phorbol-2-acetate + Calcium ionophore, interferon -γ 그리고 tumor necrosis factor -α는 CD8 세포의 분포에 큰 차이를 보이지 않았다. IL-2로 처리했을 때 CTLL-2 세포의 20%에서 NK1.1가 표현되었고, 이는 IL-2 + TGF-β 처리시 이 역시 표현율이 약 75%까지 증가 되었다. 더 나아가서, CD8 양성을 보인 대부분의 세포는 세포질내 NK1.1이 관찰되었다. 이러한 결과들은 CD4­ CD8­(doble negative) 세포에서 CD8 전구체로써의 잠재력을 조사하고 NK1.1과의 연관관계를 규명하는데 유용한 본보기가 되었으며, 또한 T세포와 NK세포가 같은 근원을 가지고 있음을 증명하고 CD8 유전자 발현의 분자생물학적 조절에 관한 연구를 하는데 있어 또 하나의 길을 열어줄 것으로 생각된다. ; It has been proposed that the term natural killer (NK) cells be reserved to the CD3^- subset while the term MHC-restricted cytotoxic T lymphocyte (CTL) be used for CD3^+ subsets. However, there is a strong evidence that NK cell may belong to T cell lineage. Recently novel lymphocyte subsets, present in the adult murine thymus, CD3+NK1.1^+TCRαβ (TNK) cell is readily identifiable in freshly obtained murine adult CD4^­CD8^­thymocytes. We used CTLL-2 cell line, which is NK1.1^+, CD3^­ TCRαβ^+ CD4^­ CD8^­cells in the presence of IL-2, to elucidate these cells can be switched to CD8 positive or CD4 positive cells. The author investigated the role of TGF-β and compared this effect with other cell signals known to be important in T cell ontogeny. Treatment with TGF-β resulted in dose dependent growth inhibition and morphological changes. During routine passage, less than 5% of cells were CD8α positive, whereas 38% of cells expressed CD8α when treated with IL-2 plus TGF-β, whereas TPA plus Calcium ionophore, IFN-γ, or TNF-α caused no significant changes in the proportion of CD8 cells. Twenty percentages of CTLL-2 cells expressed NK1.1 with IL-2 treatment, and these expression was also increased up to about 75% with IL-2 plus TGF-β. Furthermore, the most of CD8 positive cells showed intracytoplasmic NK1.1. Our results indicated that these will be useful models to investigate CD8 precursor potentials in populations of CD4^-CD8^- (double negative) cells and relationship of NK1.1. These results also demonstrate that T cells and NK cells have same ontogeny and these model offer a way to study the molecular regulation of CD8 gene expression.
