Abstract
During hepatitis B virus (HBV) infection, it has been generally thought that class I-restricted cytotoxic T lymphocytes (CTL) play a critical role in the viral clearance and immunopathology. In this study, the characteristics of class I MHC-restricted CTL responses to HBV antigens was investigated in C57BL/6 mice. Various synthetic peptides derived from HBV surface antigen (HBsAg) were screened for their binding affinity to H-2K^b molecules by MHC-stabilizing assay using RMA-S cell line. Among the 12 motif-fitting peptides, 5 peptides that have relatively high binding affinity were selected. To examine immunogenicity of the selected peptides, proliferation assays of lymphocytes from HBV antigen-primed C57BL/6 mice were performed with these peptides. Two peptides, designated C1 (FVQWFVGL) and D1 (ILSPFLPLL), showed stimulatory effects on proliferation of lymphocytes primed with HBV antigen. To define these peptides as MHC class I-restricted CTL epitopes, cytotoxic T cell responses were measured using lymphocytes primed with recombinant vaccinia virus expressing HBV envelope protein or recombinant HBsAg protein as an effectors and EL-4 pulsed with these peptides as a targets. As both of the peptides could sensitize EL-4 target cells for lysis by effector cells, it was possible to determine these two peptides, which are located near C-terminus of the HBV envelope small protein, as CTL epitopes in C57BL/6 mice. These results would be a basis of characterizing class I MHC-restricted CTL responses to HBsAg in C57BL/6 mice and contribute to the further study of immune responses to HBV infection.; Cytotoxic T lymphocyte (CTL)은 일반적으로 바이러스 감염에 대한 체내 면역 방어 작용에 중요한 역할을 담당하고 있으며, 이는 MHC class-I 분자에 제한되는 T 세포 수용체 (T cell receptor)가 항원으로부터 유래된 펩타이드를 인식하고, 그 항원을 제공하는 세포에 대한 독성을 나타냄으로써 이루어진다. 이러한 현상은 B 형 간염 바이러스 (Hepatitis B virus: HBV) 감염의 경우에 있어서도 마찬가지인데, 오히려 이 경우에는 HBV에 감염된 간 세포에 대한 독성이 지나쳐서 간염과 관련된 병리 현상을 나타나게 하는 주원인으로 알려져 있다. 따라서 CTL과 관련된 면역 반응을 연구하는 것은 간염 바이러스에 대한 전반적 면역 반응의 이해에 도움을 줄 것이며, 나아가 치료법의 개발에도 응용될 수 있을 것이다. 본 연구에서는, C57BL/6 마우스를 이용하여 HBV의 표면 단백질 (HBV envelope protein)중 CTL이 특이적으로 인식하는 펩타이드를 밝혀냄으로써 이 마우스를 이용한 다양한 면역반응 연구에 기초가 되고자 하였다. 먼저 특정 MHC 분자와의 결합에 필요하다고 알려진 anchor residue를 기초로 하여 HBV envelope protein의 전체 서열 중 8개에서 10개의 아미노산으로 구성된 다양한 펩타이드를 합성한 뒤, 이 펩타이드와 C57BL/6의 MHC class I 분자인 H-2Kb 사이의 결합력을 측정하였다. 이 결과로부터 H-2K^b에 결합할 수 있는 5가지의 펩타이드를 선택하였고, 이들이 실제로 CTL에 의해 인식되어질 수 있는 epitope 부위인지를 확인하기 위해 몇 가지 실험들을 수행하였는데, C1 과 D1 이라 표현된 두 종류의 펩타이드가 H-2K^b에 제한된 CTL에 의해 인식되는 것을 관찰할 수 있었다. 즉, HBV envelope protein부위를 발현하는 재조합 종두 바이러스 또는 재조합 HBV envelope protein에 의해 유도된 면역 세포가 이들 펩타이드의 존재하에 증식 반응을 일으키는 것이 관찰되었고, 유도된 면역 세포들 중에는 이들 펩타이드를 표면에 나타내는 세포에 대한 세포 독성을 갖는 CTL이 존재한다는 것을 확인할 수 있었다. 이 결과로부터 얻어진 두 가지의 CTL eptiope은 C57BL/6 마우스에서 HBV에 관련된 면역 반응을 연구하는데 크게 기여할 것이라 생각된다.
